Photoreduction of dioxygen in photosystem I (PSI) of chloroplasts generates superoxide radicals as the primary product. In intact chloroplasts, the superoxide and the hydrogen peroxide produced via the disproportionation of superoxide are so rapidly scavenged at the site of their generation that the active oxygens do not inactivate the PSI complex, the stromal enzymes, or the scavenging system itself. The overall reaction for scavenging of active oxygens is the photoreduction of dioxygen to water via superoxide and hydrogen peroxide in PSI by the electrons derived from water in PSII, and the water-water cycle is proposed for these sequences. An overview is given of the molecular mechanism of the water-water cycle and microcompartmentalization of the enzymes participating in it. Whenever the water-water cycle operates properly for scavenging of active oxygens in chloroplasts, it also effectively dissipates excess excitation energy under environmental stress. The dual functions of the water-water cycle for protection from photoinihibition are discussed.

Photorespiration and photoinhibition: Some implications for the energetics of photosynthesis

Current models of C3 photosynthesis incorporate a phosphate limitation to carboxylation which arises when the capacity for starch and sucrose synthesis fails to match the capacity for the production of triose phosphates in the Calvin cycle. As a result, the release of inorganic phosphate in the chloroplast stroma fails to keep pace with its rate of sequestration into triose phosphate, and phosphate becomes limiting to photosynthesis. Such a model predicts that when phosphate is limiting, assimilation becomes insensitive to both CO2 and O2, and is thus incapable of explaining the experimental observation that assimilation, under phosphate-limited conditions, frequently exhibits reversed sensitivity to both CO2 and O2, i.e., increasing O2 stimulates assimilation and increasing CO2 inhibits assimilation. We propose a model which explains reversed sensitivity to CO2 and O2 by invoking the net release of phosphate in the photorespiratory oxidation cycle. In order for this to occur, some fraction of the glycollate carbon which leaves the stroma and which is recycled to the chloroplast by the photorespiratory pathway as glycerate must remain in the cytosol, perhaps in the form of amino acids. In that case, phosphate normally used in the stromal glycerate kinase reaction to generate PGA from glycerate is made available for photophosphorylation, stimulating RuBP regeneration and assimilation. The model is parameterized for data obtained on soybean and cotton, and model behavior in response to CO2, O2, and light is demonstrated.

Univalent Reduction of Molecular Oxygen by Spinach Chloroplasts on Illumination

— In 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU) poisoned chloroplasts of algae and‘ higher plants the area over the fluorescence induction curve increases with biphasic first order kinetics (Melis and Homann, 1975). Two possibilities are considered to explain the biphasic nature of the area growth. The first is a sequential double reduction of the primary electron acceptor in system II while the second envisages a heterogeneity of its photochemical centers. The kinetic properties of the area growth after firing a single saturating flash proved to be incompatible with the predictions of the “sequential double reduction” model. This conclusion was corroborated by results obtained from a kinetic analysis of the area restoration process in the dark, and an analysis of the partially restored areas.



Assuming an existence of a heterogeneous pool of photochemical centers, the growth of the area over the fluorescence curve could be further analyzed to yield two components, a fast a-component, and a relatively slow β-component. The kinetic characteristics of these components, and the effect of a short saturating flash on their respective size, led to the conclusion that one type of photochemical center had a faster recombination rate of the photochemically separated charges and was less efficient in trapping excitation energy.

Heterogeneity of the photochemical centers in system ii of chloroplasts

Oxygen was taken up by both intact and broken chloroplasts when catalase was posioned. In confirmation of other work we found that oxygen enters the electron transport chain of isolated chloroplasts by oxidizing the primary photoreductant of system I. In isolated intact chloroplasts this reaction proceeds in addition to oxygen evolution by PGA reduction. The reductant produced by photosystem II does not react with oxygen at a significant rate. In normal leaves oxygen depresses chlorophyll fluorescence. However, this depression does not take place in DCMU poisoned leaves or in a mutant having a nonfunctional photosystem II; furthermore, another mutant with a weakly functioning photosystem I gave only a very small fluorescence depression with oxygen. This shows that the site of interaction of oxygen is at the reducing end of the electron transport chain. This view is supported by the extent of the fluorescence depression in leaves as a function of oxygen concentration which is very similar to the oxygen dependence of oxygen uptake by isolated chloroplasts. An oxygen requirement of isolated intact chloroplasts reducing PGA and nitrate was indicated by lower reaction rates and faster decay of activity under nitrogen than under air.

Effects of oxygen on the electron transport chain of photosynthesis.

Absorption and fluorescence spectra were measured for Chlamydobotrys stellata cultured either photo-heterotrophically on acetate or autotrophically on CO2 as well as during adaptation from hetero- to autotrophic conditions. Curve analyses of the absorption spectra at liquid nitrogen temperature suggest the presence of chlorophyll-a forms with their main absorption peaks at 663, 670, 678, 685, 693 and 707 nm. The proportion of the longer wavelength forms, 685, 693 and 707 nm, decreases during adaptation to autotrophic growth. The chlorophyll-b content of the photo-heterotrophic culture was very low.
