Soil-Plant-Microbe Interactions in Stressed Agriculture Management: A Review

Overexpression of bacterial γ-glutamylcysteine synthetase in the cytosol of Populus tremula × P. alba produces higher glutathione (GSH) concentrations in leaves, thereby indicating the potential for cadmium (Cd) phytoremediation. However, the net Cd(2+) influx in association with H(+) /Ca(2+) , Cd tolerance, and the underlying molecular and physiological mechanisms are uncharacterized in these poplars. We assessed net Cd(2+) influx, Cd tolerance and the transcriptional regulation of several genes involved in Cd(2+) transport and detoxification in wild-type and transgenic poplars. Poplars exhibited highest net Cd(2+) influxes into roots at pH 5.5 and 0.1 mM Ca(2+) . Transgenics had higher Cd(2+) uptake rates and elevated transcript levels of several genes involved in Cd(2+) transport and detoxification compared with wild-type poplars. Transgenics exhibited greater Cd accumulation in the aerial parts than wild-type plants in response to Cd(2+) exposure. Moreover, transgenic poplars had lower concentrations of O2 ˙(-) and H2 O2 ; higher concentrations of total thiols, GSH and oxidized GSH in roots and/or leaves; and stimulated foliar GSH reductase activity compared with wild-type plants. These results indicate that transgenics are more tolerant of 100 μM Cd(2+) than wild-type plants, probably due to the GSH-mediated induction of the transcription of genes involved in Cd(2+) transport and detoxification.

Overexpression of bacterial γ-glutamylcysteine synthetase mediates changes in cadmium influx, allocation and detoxification in poplar.

An investigation was carried out to elucidate growth, anatomical, physiological, and major ROS detoxification pathways involved in the tolerance of A. tricolor under salinity stress. Both VA14 and VA3 varieties exhibited the reduction in relative water content (RWC), photosynthetic pigments, growth, increased electrolyte leakage (EL), and leaf anatomy adaptation under salinity stress, whereas VA14 was well adapted and performed better compared to VA3. Higher ROS accumulation was demonstrated in the sensitive variety (VA3) in comparison to the tolerant variety (VA14). Salinity stress changed the cellular antioxidant pool by increasing total carotenoids, ascorbate, proline, total polyphenol content (TPC), total flavonoid content (TFC), and total antioxidant capacity (TAC) in both varieties. Although a higher increment was demonstrated in the tolerant variety, the proline increment was much more pronounced in the sensitive variety. Non-enzymatic antioxidant, ascorbate, carotenoids, TPC, TFC, TAC, and antioxidant enzymes SOD and APX were noted to be a major H2O2 detoxifier in the tolerant A. tricolor variety, where there is a comparatively lower H2O2 load. It was complemented by GPOX and CAT activity at a comparatively higher H2O2 load (in the sensitive variety). SOD contributed to the dismutation of superoxide radical (SOR) both in the tolerant and sensitive varieties; however, it greatly contributed to the dismutation of SOR in the tolerant variety. The increase in SOD, ascorbate, and APX makes it predominantly evident that SOD and the AsA-GSH cycle had greatly contributed to quench reactive oxygen species (ROS) of the tolerant variety of A. tricolor.

The Response of Salinity Stress-Induced A. tricolor to Growth, Anatomy, Physiology, Non-Enzymatic and Enzymatic Antioxidants.

Moso bamboo (Phyllostachys edulis) represents one of the fastest-spreading plants in the world, due in part to its well-developed rhizome system. However, the post-transcriptional mechanism for the development of the rhizome system in bamboo has not been comprehensively studied. We therefore used a combination of single-molecule long-read sequencing technology and polyadenylation site sequencing (PAS-seq) to re-annotate the bamboo genome, and identify genome-wide alternative splicing (AS) and alternative polyadenylation (APA) in the rhizome system. In total, 145 522 mapped full-length non-chimeric (FLNC) reads were analyzed, resulting in the correction of 2241 mis-annotated genes and the identification of 8091 previously unannotated loci. Notably, more than 42 280 distinct splicing isoforms were derived from 128 667 intron-containing full-length FLNC reads, including a large number of AS events associated with rhizome systems. In addition, we characterized 25 069 polyadenylation sites from 11 450 genes, 6311 of which have APA sites. Further analysis of intronic polyadenylation revealed that LTR/Gypsy and LTR/Copia were two major transposable elements within the intronic polyadenylation region. Furthermore, this study provided a quantitative atlas of poly(A) usage. Several hundred differential poly(A) sites in the rhizome-root system were identified. Taken together, these results suggest that post-transcriptional regulation may potentially have a vital role in the underground rhizome-root system.

Moso bamboo can rapidly complete its growth in both height and diameter within only 35–40 days after shoot emergence. However, the underlying mechanism for this “explosive growth” remains poorly understood. We investigated the dynamics of non-structural carbohydrates (NSCs) in shoots and attached mature bamboos over a 20-month period. The results showed that Moso bamboos rapidly completed their height and diameter growth within 38 days. At the same time, attached mature bamboos transferred almost all the NSCs of their leaves, branches, and especially trunks and rhizomes to the “explosively growing” shoots via underground rhizomes for the structural growth and metabolism of shoots. Approximately 4 months after shoot emergence, this transfer stopped when the leaves of the young bamboos could independently provide enough photoassimilates to meet the carbon demands of the young bamboos. During this period, the NSC content of the leaves, branches, trunks and rhizomes of mature bamboos declined by 1.5, 23, 28 and 5 fold, respectively. The trunk contributed the most NSCs to the shoots. Our findings provide new insight and a possible rational mechanism explaining the “explosive growth” of Moso bamboo and shed new light on understanding the role of NSCs in the rapid growth of Moso bamboo.

Populus × euramericana (Pe) displays higher stable carbon isotope composition (δ(13)C) and intrinsic water use efficiency (WUEi) than Populus cathayana (Pc) under unlimited water conditions, rendering us to hypothesize that Pe is better acclimated to water deficiency than Pc. To examine this hypothesis, saplings of Pc and Pe were exposed to drought and subsequently re-watered. Pc and Pe exhibited distinct anatomical, physiological and transcriptional responses in acclimation to drought and re-watering, mainly due to stronger responsiveness of transcriptional regulation of genes encoding plasma membrane intrinsic proteins (PIPs), higher starch accumulation, δ(13)C, stable nitrogen isotope composition (δ(15)N) and WUEi , and lower reactive oxygen species (ROS) accumulation and scavenging in Pe. In acclimation to drought, both poplar genotypes demonstrated altered anatomical properties, declined height growth, differential expression of PIPs, activation of ABA signaling pathway, decreased total soluble sugars and starch, increased δ(13)C, δ(15)N and WUEi , and shifted homeostasis of ROS production and scavenging, and these changes can be recovered upon re-watering. These data indicate that Pe is more tolerant to drought than Pc, and that anatomical, physiological and transcriptional acclimation to drought and re-watering is essential for poplars to survive and grow under projected dry climate scenarios in the future.
