Abstract: SUBCUTANEOUS transplantation of coarse powders of acid-insoluble matrix of the shafts of long bones of rat to allogeneic recipients results consistently in the formation of new bone through endochondral ossification1. Haematopoietic bone marrow develops in the newly formed ossicle in adolescent rats2,3 and this experimental model is amenable to an investigation of hormonal control of initiation of haematopoiesis. Erythropoiesis is dependent on pituitary hormones4,5 since hypophysectomy resulted in anaemia in all animal species which have been studied. Hypophysectomy caused a prompt regression of many stem-cell erythroblastic leukaemias in rat6. We now report the profound influence of hypophysectomy on de novo erythropoiesis in matrix-induced transformation ossicles.

Abstract: The tails of newborn rats, consisting of slender continuous cones of avascular cartilage, were transplanted in subcutaneous spaces of allogeneic adolescents; colloidal carbon was injected in a vein before the transplants were harvested on days 7-28. Between 97 and 100% of the transplants were accepted, underwent differentiation into bone with bone marrow, and grew at a brisk rate. Acceptance was recognized by (i) a zebra-stripe effect, visible in the gross, resulting from accumulation of carbon in reticuloendothelial cells; (ii) increase of alkaline phosphatase [or-thophosphoric-monoester phosphohydrolase (alkaline optimum), EC 3.1.3.1] and of incorporated 45Ca in the transplant; and (iii) the presence of dense cortical bone with lacunae populated with osteocytes demonstrable by histology. Differentiation of the cartilaginous transplant into bone with subsequent growth of the transplant occurred in recipients fed sucrose as their sole ration.

Abstract: A set of feedings of a lipid solution of 7,12-dimethylbenz(a)anthracene given biweekly by gastric intubation to young adult female mice of the non-inbred CF-1 strain elicited lymphatic leukemia in 73% of the animals in 112 +/- 24 days. Brief ether anesthesia facilitated alimentary administration of hydrocarbons and steroids. Inoculation of whole blood of mice with lymphatic leukemia into the subcutaneous tissue of allogeneic newborn resulted in lymphosarcomas at the injection site sometimes associated with leukemia. The lymphosarcomas of infant mice regressed rapidly following administration of cortisone or dexamethasone. They frequently, but not invariably, recurred; 14% of the mice were free from lymphosarcoma 3 months after treatment with the glucocorticoids.
