The invention discloses an rs12742393 polypeptide locus in a region-2 of an intron of a NOS1AP gene and a detection method thereof. Concretely, the polypeptide locus is a nucleotide at the position-121 in a sequence shown in SEQ ID No.1. The detection method comprises the following steps: extracting a genomic DNA from a sample by using the genomic DNA as a DNA template; designing a pair of nucleic acid primers that can be specifically amplified with an amplified product having the polymorphism of the nucleotide at the position-121 in the sequence shown in the SEQ ID No.1, and using the pair of primers to amplify target genes; determining the gene type of the rs12742393 polypeptide locus and detecting if the A to C single nucleotide polymorphism exists at the position-121 in the sequence shown in the SEQ ID NO.1. The invention establishes a method for quickly detecting the rs12742393 polypeptide locus in the NOS1AP gene in a batch sample.

OBJECTIVE Hepatocyte nuclear factor 1β (HNF1β) is a transcription factor that is critical for pancreatic cell formation and glucose homeostasis. Previous studies have reported that common variants of HNF1β were associated with type 2 diabetes in Caucasians and West Africans. However, analysis in the subjects from the Botnia study and Malmö Preventive Project produced conflicting results, and the role for HNF1β in type 2 diabetes susceptibility remains unclear. We therefore investigated common variants across the HNF1β gene in a Chinese population. RESEARCH DESIGN AND METHODS Fifteen tagging single nucleotide polymorphisms (SNPs) were analyzed for association with type 2 diabetes in subjects with type 2 diabetes (n = 1,859) and normal glucose regulation (n = 1,785). RESULTS Consistent with the initial study, we observed evidence that the risk G allele of rs4430796 in intron 2 was significantly associated with type 2 diabetes (odds ratio 1.16 [95% CI 1.05–1.29], P = 0.0035, empirical P = 0.0475). Furthermore, the at-risk G allele was associated with earlier age at diagnosis in the type 2 diabetic subjects (P = 0.0228). CONCLUSIONS The result of this study provides evidence that variants in the HNF1β region contribute to susceptibility to type 2 diabetes in the Chinese population.

Common Variants of Hepatocyte Nuclear Factor 1 Are Associated With Type 2 Diabetes in a Chinese

12(S)‐hydroxyeicosatetraenoic (12(S)‐HETE), an alternate arachidonic acid metabolite, has been recently examined in metabolic disease. However, the role of 12(S)‐HETE in diabetic kidney disease (DKD) remains unclear. We studied for the first time the relationship of serum 12(S)‐HETE and DKD and renal function parameters in a Chinese population.

The effects of tetrandrine (Tet) on seven isolated vascular strips of rabbit were compared. The influence of Tet, verapamil (Ver) and phentolamine (Phen) on noradrenaline (NA) dose-response curves were studied. Tet injibited the contraction evoked by high K+ (60 mM) in thoracic aortic strips and more prominently in mesenteric arterial strips. Phen competiticely antagonized NA. Tet and Ver also antagonized NA but in a noneompetitive manner. Calcium was antagonized by Tet and Ver noncompetitively.
The findings suggest that the Tet-induced telaxation on vascular smooth muscle results from its antagonism to calcium, similar to verapamil.

Effects of tetrandrine on seven vascular smooth muscles

The TM4 (UBAC2) protein, which contains 4 transmembrane domains and one ubiquitin binding domain, is mainly expressed in cell and nuclear membranes. The current research aimed to explore the role of TM4 in metabolic inflammation and to examine whether the ubiquitin–proteasome inhibitor PS-341 could regulate the function of TM4. The metabolic phenotypes of TM4 knockout (KO) mice were studied. We next explored the association between the polymorphisms of TM4 and obesity in a Chinese Han population. TM4 expression in the visceral fat of obese patients who underwent laparoscopic cholecystectomy was also analysed. Finally, the effect of PS-341 on the degradation and function of the TM4 protein was investigated in vivo and in vitro. TM4 KO mice developed obesity, hepatosteatosis, hypertension, and glucose intolerance under a high-fat diet. TM4 counterregulated Nur77, IKKβ, and NF-kB both in vivo and in vitro. The TM4 SNP rs147851454 is significantly associated with obesity after adjusting for age and sex (OR 1.606, 95% CI 1.065–2.422 P = 0.023) in 3394 non-diabetic and 1862 type 2 diabetic adults of Han Chinese. TM4 was significantly downregulated in the visceral fat of obese patients. PS-341 induced TM4 expression through inhibition of TM4 degradation in vitro. In db/db mice, PS-341 administration led to downregulation of Nur77/IKKβ/NF-κB expression in visceral fat and liver, and alleviation of hyperglycaemia, hypertension, and glucose intolerance. The hyperinsulinaemic-euglycaemic clamp demonstrated that PS-341 improved the glucose infusion rate and alleviated insulin resistance in db/db mice. PS-341 alleviates chronic low-grade inflammation and improves insulin sensitivity through inhibition of TM4 degradation.
