Journal ArticleDOI
Toward a clinical-grade expansion of mesenchymal stem cells from human sources: a microcarrier-based culture system under xeno-free conditions.
Francisco dos Santos,Pedro Z. Andrade,Manuel Abecasis,Jeffrey M. Gimble,Lucas G. Chase,Andrew M. Campbell,Shayne Boucher,Mohan C. Vemuri,Cláudia Lobato da Silva,Joaquim M. S. Cabral +9 more
TL;DR: The feasibility of expanding human MSC in a scalable microcarrier-based stirred culture system under xeno-free conditions is demonstrated and represents an important step forward for the implementation of a Good Manufacturing Practices-compliant large-scale production system of MSC for cellular therapy.
Journal ArticleDOI
Mesenchymal stem cells from umbilical cord matrix, adipose tissue and bone marrow exhibit different capability to suppress peripheral blood B, natural killer and T cells.
Andreia Ribeiro,Paula Laranjeira,Sandrine Mendes,Isabel Velada,Cristiana de Sousa Leite,Pedro Z. Andrade,Francisco dos Santos,Ana Henriques,Mário Grãos,Carla M.P. Cardoso,António Martinho,M. Luisa Pais,Cláudia Lobato da Silva,Joaquim M. S. Cabral,Hélder Trindade,Artur Paiva +15 more
TL;DR: Overall, UCM-, BM- and AT-derived MSCs hamper T cell, B cell and NK cell-mediated immune response by preventing their acquisition of lymphoblast characteristics, activation and changing the expression profile of proteins with an important role in immune function, except UCM-MSCs showed no inhibitory effect on B cells under these experimental conditions.
Journal ArticleDOI
Stem cell cultivation in bioreactors.
Carlos A V Rodrigues,Tiago G. Fernandes,Maria Margarida Diogo,Cláudia Lobato da Silva,Joaquim M. S. Cabral +4 more
TL;DR: This review describes recent developments in the cultivation of stem cells in bioreactors, particularly considerations regarding critical culture parameters, possible bioreactor configurations, and integration of novel technologies in the bioprocess development stage.
Journal ArticleDOI
Maximizing the ex vivo expansion of human mesenchymal stem cells using a microcarrier-based stirred culture system.
Gemma Eibes,Francisco dos Santos,Pedro Z. Andrade,Joana S. Boura,Manuel Abecasis,Cláudia Lobato da Silva,Joaquim M. S. Cabral +6 more
TL;DR: It is demonstrated that a microcarrier-based stirred culture system is adequate for human MSC expansion, using a low-serum containing medium, allowing the generation of significant cell numbers for potential applications in regenerative medicine.
Journal ArticleDOI
A xenogeneic-free bioreactor system for the clinical-scale expansion of human mesenchymal stem/stromal cells
Francisco dos Santos,Andrew Campbell,Ana Fernandes-Platzgummer,Pedro Z. Andrade,Jeffrey M. Gimble,Yuan Wen,Shayne Boucher,Mohan C. Vemuri,Cláudia Lobato da Silva,Joaquim M. S. Cabral +9 more
TL;DR: A xenogeneic (xeno)‐free microcarrier‐based culture system was successfully established for bone marrow (BM) MSC and adipose tissue‐derived stem/stromal cell (ASC) cultivation using a 1L‐scale controlled stirred‐tank bioreactor system under xeno‐free conditions.
