Journal ArticleDOI
Structure-function analysis of the human sialyltransferase ST3Gal I: role of n-glycosylation and a novel conserved sialylmotif.

TL;DR: Mutational analysis showed that residues His299 and Tyr300 of the new motif, and His316 of the VS motif, are essential for activity since their substitution by alanine yielded inactive enzymes, and the invariant Tyr residue plays an important conformational role mainly attributable to the aromatic ring.
Journal ArticleDOI
Synthesis with immobilized enzyme of the most important sialic acid.
TL;DR: In this paper, a crude mixture of N-acetylmannosamine and Nacetylglucosamine has been used to synthesize N -acetylneuraminic acid on 2.8 millimoles scale.
Journal ArticleDOI
The use of immobilised glycosyltransferases in the synthesis of sialyloligosaccharides.
TL;DR: CMP-NeuAc was used as a donor of N-acetylneuraminic acid in the reaction catalysed by immobilised porcine liver, and alpha-D-Neup5Ac-(2----6)-beta- D-Galp-(1----4)-beta -D-GlcpNAc-(1--- -2)-alpha-D - Man-OMe (5) was obtained on a 0.1-mmol scale.
Journal ArticleDOI
Non-isosteric C-glycosyl analogues of natural nucleotide diphosphate sugars as glycosyltransferase inhibitors.
TL;DR: In vitro inhibitory activity of UDP-Gal, GDP-Fuc and UDP-GlcNAc analogues towards glycosyltransferases (beta-1,4-GalT, FUT3 and LgtA) was evaluated through a competition fluorescence assay and IC(50) values of 40 microM, 2 mM and 35 mM were obtained.
Journal ArticleDOI
Synthesis with immobilized enzymes of two trisaccharides, one of them active as the determinant of a stage antigen.
TL;DR: Glycosidation of disaccharides I and III with a system of five immobilized enzymes gave excellent yields of trisaccharide β-D -Gal-(1→4)- β -D -GlcNAc-( 1→6)- D -Gal, on the milimole scale as mentioned in this paper.
