Lack of the bone morphogenetic protein BMP6 induces massive iron overload
Delphine Meynard,Delphine Meynard,Léon Kautz,Léon Kautz,Valérie Darnaud,Valérie Darnaud,François Canonne-Hergaux,Hélène Coppin,Hélène Coppin,Marie-Paule Roth,Marie-Paule Roth +10 more
TL;DR: It is shown that targeted disruption of Bmp6 in mice causes a rapid and massive accumulation of iron in the liver, the acinar cells of the exocrine pancreas, the heart and the renal convoluted tubules, and this indicates that BMP6 is critical for iron homeostasis and that it is functionally nonredundant with other members of the Bmp subfamily.
Journal ArticleDOI
Iron regulates phosphorylation of Smad1/5/8 and gene expression of Bmp6, Smad7, Id1, and Atoh8 in the mouse liver.
Léon Kautz,Léon Kautz,Delphine Meynard,Delphine Meynard,Annabelle Monnier,Annabelle Monnier,Valérie Darnaud,Valérie Darnaud,Régis Bouvet,Rui-Hong Wang,Chiuxia Deng,Sophie Vaulont,Sophie Vaulont,Jean Mosser,Hélène Coppin,Hélène Coppin,Marie-Paule Roth,Marie-Paule Roth +17 more
TL;DR: This study points out molecules that appear to play a critical role in the control of systemic iron balance, including Smad4, which is required for activation of Smad7, Id1, and Atoh8 transcription by iron, and 4 which are related to the BMP/Smad pathway.
Journal ArticleDOI
Serum and liver iron differently regulate the bone morphogenetic protein 6 (BMP6)-SMAD signaling pathway in mice
Elena Corradini,Delphine Meynard,Qifang Wu,Shan Chen,Paolo Ventura,Antonello Pietrangelo,Jodie L. Babitt +6 more
TL;DR: It is demonstrated that both transferrin saturation and liver iron content independently influence hepcidin expression, whereas the hepatic Erk1/2 signaling pathway is not activated by iron in vivo.
Journal ArticleDOI
The liver: conductor of systemic iron balance
TL;DR: How the liver is the central conductor of systemic iron balance is reviewed and shown that this central role is related to the secretion of a peptide hormone hepcidin by hepatocytes.
Journal ArticleDOI
BMP/Smad signaling is not enhanced in Hfe-deficient mice despite increased Bmp6 expression.
Léon Kautz,Delphine Meynard,Delphine Meynard,Céline Besson-Fournier,Céline Besson-Fournier,Valérie Darnaud,Valérie Darnaud,Talal Al Saati,Talal Al Saati,Hélène Coppin,Hélène Coppin,Marie-Paule Roth,Marie-Paule Roth +12 more
TL;DR: The inappropriately low levels of Id1 and hepcidin mRNA observed at weaning suggest that Hfe deficiency triggers iron overload by impairing hepatic Bmp/Smad signaling, and HFE appears to facilitate signal transduction induced by the BMP6 ligand.
