Point of care testing: The impact of nanotechnology.

Electrochemistry, biosensors and microfluidics are popular research topics that have attracted widespread attention from chemists, biologists, physicists, and engineers. Here, we introduce the basic concepts and recent histories of electrochemistry, biosensors, and microfluidics, and describe how they are combining to form new application-areas, including so-called “point-of-care” systems in which measurements traditionally performed in a laboratory are moved into the field. We propose that this review can serve both as a useful starting-point for researchers who are new to these topics, as well as being a compendium of the current state-of-the art for experts in these evolving areas.

Electrochemistry, biosensors and microfluidics: a convergence of fields

A digital microfluidic platform manipulates droplets on an open surface. Magnetic digital microfluidics utilizes magnetic forces for actuation and offers unique advantages compared to other digital microfluidic platforms. First, the magnetic particles used in magnetic digital microfluidics have multiple functions. In addition to serving as actuators, they also provide a functional solid substrate for molecule binding, which enables a wide range of applications in molecular diagnostics and immunodiagnostics. Second, magnetic digital microfluidics can be manually operated in a “power-free” manner, which allows for operation in low-resource environments for point-of-care diagnostics where even batteries are considered a luxury item. This review covers research areas related to magnetic digital microfluidics. This paper first summarizes the current development of magnetic digital microfluidics. Various methods of droplet manipulation using magnetic forces are discussed, ranging from conventional magnetic particle-based actuation to the recent development of ferrofluids and magnetic liquid marbles. This paper also discusses several new approaches that use magnetically controlled flexible substrates for droplet manipulation. In addition, we emphasize applications of magnetic digital microfluidics in biosensing and medical diagnostics, and identify the current limitations of magnetic digital microfluidics. We provide a perspective on possible solutions to close these gaps. Finally, the paper discusses the future improvement of magnetic digital microfluidics to explore potential new research directions.

Magnetic digital microfluidics – a review

Spectrometrically or optically encoded microsphere based suspension array technology (SAT) is applicable to the high-throughput, simultaneous detection of multiple analytes within a small, single sample volume. Thanks to the rapid development of nanotechnology, tremendous progress has been made in the multiplexed detecting capability, sensitivity, and photostability of suspension arrays. In this review, we first focus on the current stock of nanoparticle-based barcodes as well as the manufacturing technologies required for their production. We then move on to discuss all existing barcode-based bioanalysis patterns, including the various labels used in suspension arrays, label-free platforms, signal amplification methods, and fluorescence resonance energy transfer (FRET)-based platforms. We then introduce automatic platforms for suspension arrays that use superparamagnetic nanoparticle-based microspheres. Finally, we summarize the current challenges and their proposed solutions, which are centered on improving encoding capacities, alternative probe possibilities, nonspecificity suppression, directional immobilization, and “point of care” platforms. Throughout this review, we aim to provide a comprehensive guide for the design of suspension arrays, with the goal of improving their performance in areas such as multiplexing capacity, throughput, sensitivity, and cost effectiveness. We hope that our summary on the state-of-the-art development of these arrays, our commentary on future challenges, and some proposed avenues for further advances will help drive the development of suspension array technology and its related fields.

Suspension arrays based on nanoparticle-encoded microspheres for high-throughput multiplexed detection.

We introduce an automated digital microfluidic (DMF) platform capable of performing immunoassays from sample to analysis with minimal manual intervention. This platform features (a) a 90 Pogo pin interface for digital microfluidic control, (b) an integrated (and motorized) photomultiplier tube for chemiluminescent detection, and (c) a magnetic lens assembly which focuses magnetic fields into a narrow region on the surface of the DMF device, facilitating up to eight simultaneous digital microfluidic magnetic separations. The new platform was used to implement a three-level full factorial design of experiments (DOE) optimization for thyroid-stimulating hormone immunoassays, varying (1) the analyte concentration, (2) the sample incubation time, and (3) the sample volume, resulting in an optimized protocol that reduced the detection limit and sample incubation time by up to 5-fold and 2-fold, respectively, relative to those from previous work. Automated digital microfluidic platform for magnetic-particle-based immunoassays with optimization by design of experiments.

A multi-element diffraction grating is disclosed. The multi-element diffraction grating may be configured to diffract incident radiation with high dispersion using at least two holographic optical elements. The multi-element diffraction grating may include a first volume phase grating that is one of the holographic optical elements. The multi-element diffraction grating may also include at least one additional volume phase grating that is the second holographic optical element. The first volume phase grating and each of the additional volume phase gratings may be positioned relative to each other such that the radiation propagates through and is diffracted by the first volume phase grating and each of the additional volume phase gratings.

High dispersion diffraction grating including multiple holographic optical elements

Volume phase holographic gratings (VPHGs) have gained popularity for use in astronomical spectrographs due to their high first order diffraction efficiency, angle tuning capabilities, low scatter, durability, and ability to be manufactured in large formats; gratings up to 30 cm across are routinely achievable. The nature of the VPHG manufacturing process offers optical and instrument designers a tremendous amount of flexibility to optimize performance for a specific application or need. In this talk, we will explore VPHGs from a grating designer’s perspective, aiming to give insight to spectrograph designers as they develop grating requirements for new spectrographs. We will address considerations and limitations with respect to grating size, angle of incidence (AOI), dispersion, wavelength of operation, and bandwidth. We will look at how AOI affects grating design and efficiency, stepping through several AOI regimes to explore the diffraction efficiency and spectral profiles which can be achieved, as well as the physics behind them – from required film thickness to the relative efficiency of s- and p- polarization as index modulation is varied in each regime. We will also touch upon the advantages offered by use of a grism configuration (GRating + prISM), and provide examples of both routine and unusual examples of grism implementation.
