Therapeutic treatment with an oral prodrug of the remdesivir parental nucleoside is protective against SARS-CoV-2 pathogenesis in mice
Alexandra Schäfer,David R. Martinez,John J. Won,Rita M. Meganck,Fernando R. Moreira,Ariane J. Brown,Kendra Gully,M. Zweigart,William S. Conrad,S. May,Stephanie Dong,Rao Kalla,Kwon Soo Chun,Venice Du Pont,Darius Babusis,Jennifer Tang,Eisuke Murakami,Raju Subramanian,K Barrett,Blake J. Bleier,Roy Bannister,Joy Y. Feng,John P. Bilello,Tomas Cihlar,Richard L. Mackman,Stephanie A. Montgomery,Ralph S. Baric,Timothy P. Sheahan +27 more
TL;DR: The in vitro antiviral activity and in vivo therapeutic efficacy of GS-621763, an orally bioavailable prodrug ofGS-441524, the parent nucleoside of remdesivir, which targets the highly conserved virus RNA-dependent RNA polymerase of SARS-CoV-2 are demonstrated.
Journal ArticleDOI
Growth and adaptation of Zika virus in mammalian and mosquito cells.
Lindsey A. Moser,Brendan T. Boylan,Fernando R. Moreira,Laurel J. Myers,Emma L. Svenson,Nadia Fedorova,Brett E. Pickett,Kristen A. Bernard +7 more
TL;DR: The results suggest that the ability to infect and/or replicate in insect cells is limited after growth in mammalian cells, and underscore the importance of the cells used to produce viral stocks and the potential for adaptation with minimal cell passages.
Journal ArticleDOI
A deep insight into the male and female sialotranscriptome of adult Culex tarsalis mosquitoes.
TL;DR: An Illumina-based sialotranscriptome was reported that allowed finding the missing apyrase from the salivary transcriptome of C. tarsalis, to determine several full-length members of the 34-62 kDa family, when a single EST has been found previously, in addition to identifying manySalivary families with lower expression levels that were not detected previously.
Journal ArticleDOI
Facile discovery of surrogate cytokine agonists
Michelle Yen,Junming Ren,Qingxiang Liu,Caleb R. Glassman,Timothy P. Sheahan,Lora K. Picton,Fernando R. Moreira,Arjun Rustagi,Kevin Jude,Xiang Zhao,Catherine A. Blish,Ralph S. Baric,Leon Su,K. Christopher Garcia +13 more
TL;DR: In this article , the authors presented a strategy to discover cytokine surrogate agonists by using modular ligands that exploit induced proximity and receptor dimer geometry as pharmacological metrics amenable to high-throughput screening.
Journal ArticleDOI
Programmable antivirals targeting critical conserved viral RNA secondary structures from influenza A virus and SARS-CoV-2
Rachel J. Hagey,Menashe Elazar,Edward A. Pham,Siqi Tian,Lily Ben-Avi,Claire Bernardin-Souibgui,Matthew F Yee,Fernando R. Moreira,Meirav Vilan Rabinovitch,Rita M. Meganck,Benjamin Fram,Aimee Beck,Scott A. Gibson,Grace Lam,Joseph Devera,Wipapat Kladwang,Khanh Nguyen,Anming Xiong,Steven Schaffert,Talia Avisar,Ping Liu,Arjun Rustagi,Carl J. Fichtenbaum,Phillip S. Pang,Purvesh Khatri,Chien Te K. Tseng,Jeffery K. Taubenberger,Catherine A. Blish,Brett L. Hurst,Timothy P. Sheahan,Rhiju Das,Jeffrey S. Glenn +31 more
TL;DR: The authors identified and genetically validated an RNA structure (packaging stemloop 2 (PSL2)) that mediates in vitro packaging and in vivo disease and is conserved across all known IAV isolates.
