Wood is a renewable, versatile material with multiple applications and the largest terrestrial pool of sequestered carbon. However, it is susceptible to degradation, mainly caused by wood-decaying fungi. Since several traditional wood preservatives have been banned owing to their detrimental effects on humans and the environment, extending the lifespan of wood products using new generation natural preservatives is an imperative from the perspectives of human health and environmental protection. Several natural compounds of plant and animal origin have been tested for their fungicidal properties, including essential oils, tannins, wood extractives, alkaloids, propolis or chitosan; and their enormous potential in wood protection has been shown. Although they are not free of limitations, the potential methods to overcome their drawbacks and enhance their bioactivity already exist, such as co-impregnation with different polymers, cross-linkers, metal chelators or antioxidants. The presence of the discrepancies between laboratory tests and the field performance, as well as legislation-related problems resulting from the lack of standards defining the quality and performance of natural protective formulations, however, create an urgent need for further thorough research and arrangements. The collaboration with other industries interested in the utilisation of natural active compounds will reduce the associated costs, thus, will facilitate the successful implementation of alternative antifungal agents.

Natural Compounds for Wood Protection against Fungi-A Review.

The biological properties and main phenolic compounds of the O. vulgare L. ssp. vulgare extract are described in the present paper. The polyphenolic compounds were analyzed by chromatographic and spectrophotometric techniques. The antioxidant potential was evaluated using several methods: CUPRAC (cupric ion reducing antioxidant capacity), FRAP (ferric reducing ability of plasma), inhibition of lipid peroxidation catalyzed by cytochrome c, and superoxide (SO) scavenging assays. The antimicrobial activity of the oregano extract was evaluated by means of agar-well diffusion assay. The hepatoprotective effect of the O. vulgare extract on CCl4-induced hepatotoxicity was evaluated in rats. Liver injury was estimated by determination of alanine aminotransaminase (ALT), aspartate aminotransaminase (AST), gamma-glutamyl transferase GGT, total protein and albumin concentrations, glutathione peroxidase (GPx), catalase (CAT), superoxide dismutase (SOD) and malondialdehyde (MDA). These values were improved by the administration of oregano extract. A specific phenolic profile was evidenced by these data, with large amounts of rosmarinic and chlorogenic acids. The oregano extract showed very strong antioxidant activity in good agreement with the phenolic content. Antimicrobial activity was good, especially against Salmonella enteritidis and Aspergillus niger strains. The high hepatoprotective, antioxidant and antimicrobial activity, along with polyphenol-rich content, can support the use of O. vulgare in therapy. We also expect our results to open new research directions for designing important new drug products, using indigenous plant material.

Background Gallic acid (GA-3,4,5-trihydroxybenzoic acid), a phenolic phytochemical, is a ubiquitous secondary metabolite found in most plants, with appreciable concentrations in grapes seed, rose flowers, sumac, oak and witch hazel. GA often results from the hydrolysis of terpenes and the polyphenol tannic acid. Applications It exhibits powerful antioxidant, anti-inflammatory, antimicrobial, and anti-cancer activities. Most intriguing benefit has been reported to be on the skin. Due to these beneficial properties, GA and its derivatives (e.g. lipid-soluble phenols such as synthetic gallic esters aka gallates) have been extensively used as an adjuvant in a number of therapeutic formulations, as a substitute of hydrocortisone in children with atopic dermatitis (AD) and other skin conditions (hyperpigmentation, wound healing), and as a cosmetic ingredient. GA has a USFDA GRAS status (generally recognized as safe), exhibiting fairly low systemic toxicity and associated mortality at acute doses in many experimental models. Despite anti-skin aging benefits obtained with relatively safe GA formulations, few cases of gallate-induced skin allergic have been reported in humans. Therefore, approaches to improve the bioavailability and biodegradability of this poor-water soluble and non-biodegradable phenolic compound are warranted. Purpose This review has focused on the recently reported biological activities pertaining to the skin as well as the pharmacological properties of GA and its derivatives with special emphasis on its use in (nano-) cosmetic formulations. Since this is an evolving area of research, an adequate emphasis has been placed upon advantages and disadvantages of various nanoformulations.

Hepatitis B virus (HBV) is a human pathogen causing severe liver disease and eventually death. Despite important progress in deciphering HBV internalization, the early virus-cell interactions leading to infection are not known. HepaRG is a human bipotent liver cell line bearing the unique ability to differentiate towards a mixture of hepatocyte- and biliary-like cells. In addition to expressing metabolic functions normally found in liver, differentiated HepaRG cells support HBV infection in vitro, thus resembling cultured primary hepatocytes more than other hepatoma cells. Therefore, extensive characterization of the plasma membrane proteome from HepaRG cells would allow the identification of new cellular factors potentially involved in infection. Here we analyzed the plasma membranes of non-differentiated and differentiated HepaRG cells using nanoliquid chromatography-tandem mass spectrometry to identify the differences between the proteomes and the changes that lead to differentiation of these cells. We followed up on differentially-regulated proteins in hepatocytes- and biliary-like cells, focusing on Cathepsins D and K, Cyclophilin A, Annexin 1/A1, PDI and PDI A4/ERp72. Major differences between the two proteomes were found, including differentially regulated proteins, protein-protein interactions and intracellular localizations following differentiation. The results advance our current understanding of HepaRG differentiation and the unique properties of these cells.

Comparative proteomics reveals novel components at the plasma membrane of differentiated HepaRG cells and different distribution in hepatocyte- and biliary-like cells.

The tendency of using herbs extracts or natural compounds extracted from herbs for preventing or treating different illnesses, including cancer, continues to be an alternative to drug use. Many studies of plant compounds aimed at finding substances with selective cytotoxicity on abnormal cells. Phenolic compounds, as important secondary metabolites from plants, are one of them. In this review, the recent literature data from the past five years about anticancer/antitumor effect of flavonoids and tannins extracted from medicinal plants are surveyed. The cytostatic/antitumor effects of the individual compounds extracted from plants and/or of the plants' polyphenolic extracts are considered, in order to point out the most significant constituents or plants with anticancer potential. The most important results concerning these compounds and their derivatives in cancer prevention and treatment, the importance of their chemical structure, their mechanism of action in vitro and in vivo, and some bioavailability aspects are discussed.

The Potential of Flavonoids and Tannins from Medicinal Plants as Anticancer Agents.

In this paper, a liquid chromatography-atmospheric pressure chemical ionization-mass spectrometry in negative mode method was developed for the identification and quantitative determination of 13 individual phenolics (chlorogenic acid, caffeic acid, coumaric acid, ferulic acid, (1)-catechin, ( –)-epicatechin, rutin, quercitrin, isoquercitrin, fisetin, isorhamnetin, hesperidin and chrysin) from ethanolic extracts [30, 50 and 70% (w/v)] of Calendula officinalis, Hypericum perforatum, Galium verum and Origanum vulgare and some commercial extracts of these medicinal herbs. Correlation coefficients (r 2 ) from calibration curves for all the compounds were between 0.9971 and 0.9996. Limit of detection was in the range of 0.070 – 0.280 mg/mL and limit of quantification was from 0.233 to 0.932 mg/mL. The method was partially validated and the results obtained are: the intra- and interday relative standard deviation values were within 0.086 and 2.821% and recovery values vary from 95.84% (coumaric acid) to 103.20% (rutin).
