Synthetic Lethality Targeting Polθ
TL;DR: The authors discuss the current state of knowledge on Polθ as a potential target for synthetic lethality-based anticancer therapies and suggests it is a promising target in the treatment of tumors harboring deficiencies in homologous recombination repair (HRR).
Journal ArticleDOI
Polθ Inhibition: An Anticancer Therapy for HR-Deficient Tumours
TL;DR: In this paper , the authors describe the significance of DNA polymerase theta and the Polθ-mediated TMEJ pathway and summarize the current state-of-the-art inhibitors and emphasize the promising role of Pol θ as a therapeutic target.

TL;DR: In this article , the authors found that the use of VPA in combination with talazoparib (PARP1 inhibitor) and/or Dacarbazine (DTIC) resulted in an increased rate of DNA double strand breaks (DSBs) and reduced survival (while not affecting primary melanocytes) and proliferation of melanoma cells.
