Host-Viral Infection Maps Reveal Signatures of Severe COVID-19 Patients.

The NLRP3 inflammasome is cytosolic multi-protein complex that induces inflammation and pyroptotic cell death in response to both pathogen (PAMPs) and endogenous activators (DAMPs). Recognition of PAMPs or DAMPs leads to formation of the inflammasome complex, which results in activation of caspase-1, followed by cleavage and release of pro-inflammatory cytokines. Excessive activation of NLRP3 inflammasome can contribute to development of inflammatory diseases and cancer. Autophagy is vital intracellular process for recycling and removal of damaged proteins and organelles, as well as destruction of intracellular pathogens. Cytosolic components are sequestered in a double-membrane vesicle-autophagosome, which then fuses with lysosome resulting in degradation of the cargo. The autophagy dysfunction can lead to diseases with hyperinflammation and excessive activation of NLRP3 inflammasome and thus acts as a major regulator of inflammasomes. Autophagic removal of NLRP3 inflammasome activators, such as intracellular DAMPs, NLRP3 inflammasome components, and cytokines can reduce inflammasome activation and inflammatory response. Likewise, inflammasome signaling pathways can regulate autophagic process necessary for balance between required host defense inflammatory response and prevention of excessive and detrimental inflammation. Autophagy has a protective role in some inflammatory diseases associated with NLRP3 inflammasome, including gouty arthritis, familial Mediterranean fever (FMF), and sepsis. Understanding the interregulation between these two essential biological processes is necessary to comprehend the biological mechanisms and designing possible treatments for multiple inflammatory diseases.

Interplay Between NLRP3 Inflammasome and Autophagy.

Type VI CRISPR enzymes are RNA-targeting proteins with nuclease activity that enable specific and robust target gene knockdown without altering the genome. To define rules for the design of Cas13d guide RNAs (gRNAs), we conducted massively parallel screens targeting messenger RNAs (mRNAs) of a green fluorescent protein transgene, and CD46, CD55 and CD71 cell-surface proteins in human cells. In total, we measured the activity of 24,460 gRNAs with and without mismatches relative to the target sequences. Knockdown efficacy is driven by gRNA-specific features and target site context. Single mismatches generally reduce knockdown to a modest degree, but spacer nucleotides 15-21 are largely intolerant of target site mismatches. We developed a computational model to identify optimal gRNAs and confirm their generalizability, testing 3,979 guides targeting mRNAs of 48 endogenous genes. We show that Cas13 can be used in forward transcriptomic pooled screens and, using our model, predict optimized Cas13 gRNAs for all protein-coding transcripts in the human genome.

Massively parallel Cas13 screens reveal principles for guide RNA design

The emergence of high-entropy materials (HEMs) with their excellent mechanical properties, stability at high temperatures, and high chemical stability is poised to yield new advancement in the performance of energy storage and conversion technologies. This review covers the recent developments in catalysis, water splitting, fuel cells, batteries, supercapacitors, and hydrogen storage enabled by HEMs covering metallic, oxide, and non-oxide alloys. Here, first, the primary rules for the proper selection of the elements and the formation of a favorable single solid solution phase in HEMs are defined. Furthermore, recent developments in different fields of energy conversion and storage achieved by HEMs are discussed. Higher electrocatalytic and catalytic activities with longer cycling stability and durability compared to conventional noble metal-based catalysts are reported for high-entropy materials. In electrochemical energy storage systems, high-entropy oxides and alloys have shown superior performance as anode and cathode materials with long cycling stability and high capacity retention. Also, when used as metal hydrides for hydrogen storage, remarkably high hydrogen storage capacity and structural stability are observed for HEMs. In the end, future directions and new energy-related technologies that can be enabled by the application of HEMs are outlined.

Recent progress of high-entropy materials for energy storage and conversion

Nanostructured materials premise to revolutionize the label-free biosensing of analytes for clinical applications, leveraging the deeper interaction between materials and analytes with comparable size. However, when the characteristic dimension of the materials reduces to the nanoscale, the surface functionalization for the binding of bioreceptors becomes a complex issue that can affect the performance of label-free biosensors. Here we report on an effective and robust route for surface biofunctionalization of nanostructured materials based on the layer-by-layer (LbL) electrostatic nano-assembly of oppositely-charged polyelectrolytes, which are engineered with bioreceptors to enable label-free detection of target analytes. LbL biofunctionalization is demonstrated using nanostructured porous silicon (PSi) interferometers for affinity detection of streptavidin in saliva, through LbL nano-assembly of a bi-layer of positively-charged poly(allylamine hydrochloride) (PAH) and negatively-charged biotinylated poly(methacrylic acid) (b-PMAA). High sensitivity in streptavidin detection is achieved, with high selectivity and stability, down to a detection limit of 600 fM. In label-free biosensing surface functionalisation is a complex issue that can affect sensing performance. Here, the authors report on an electrostatic layer-by-layer technique to functionalize a surface and demonstrate this technique using biotinylated polymer for streptavidin detection in saliva.
