Journal ArticleDOI
Endogenous siRNAs from naturally formed dsRNAs regulate transcripts in mouse oocytes
Toshiaki Watanabe,Toshiaki Watanabe,Yasushi Totoki,Atsushi Toyoda,Masahiro Kaneda,Masahiro Kaneda,Satomi Kuramochi-Miyagawa,Yayoi Obata,Hatsune Chiba,Hatsune Chiba,Yuji Kohara,Yuji Kohara,Tomohiro Kono,Toru Nakano,M. Azim Surani,Yoshiyuki Sakaki,Hiroyuki Sasaki,Hiroyuki Sasaki +17 more
TL;DR: The results reveal a role for endogenous siRNAs in mammalian oocytes and show that organisms lacking RdRP activity can produce functional endogenous si RNAs from naturally occurring dsRNAs.
Journal ArticleDOI
Targets and dynamics of promoter DNA methylation during early mouse development
Julie Borgel,Sylvain Guibert,Yufeng Li,Yufeng Li,Hatsune Chiba,Dirk Schübeler,Hiroyuki Sasaki,Thierry Forné,Michael Weber +8 more
TL;DR: This work optimized methylated DNA immunoprecipitation for low numbers of cells and profiled DNA methylation during early development of the mouse embryonic lineage in vivo, suggesting that escape of post-fertilizationDNA methylation reprogramming is prevalent in the mouse genome.
Journal ArticleDOI
Maternal and zygotic Dnmt1 are necessary and sufficient for the maintenance of DNA methylation imprints during preimplantation development
Ryutaro Hirasawa,Hatsune Chiba,Hatsune Chiba,Masahiro Kaneda,Masahiro Kaneda,Shoji Tajima,En Li,Rudolf Jaenisch,Hiroyuki Sasaki,Hiroyuki Sasaki +9 more
TL;DR: It is demonstrated, by using conditional knockout mice, that the other known DNA methyltransferases Dnmt3a and DnMT3b are dispensable for the maintenance of the methylation marks at most imprinted loci and that zygotic DNmt1 is expressed in the preimplantation embryo.
Journal ArticleDOI
Mouse oocyte methylomes at base resolution reveal genome-wide accumulation of non-CpG methylation and role of DNA methyltransferases.
Kenjiro Shirane,Hidehiro Toh,Hisato Kobayashi,Fumihito Miura,Hatsune Chiba,Hatsune Chiba,Takashi Ito,Tomohiro Kono,Hiroyuki Sasaki +8 more
TL;DR: It is found that nearly two-thirds of all methylcytosines occur in a non-CG context in GVOs, which indicates that non- CG methylation accumulates during oocyte growth and provides a basis for understanding the mechanisms and significance ofNon-CG methylation in mammalian oocytes.
Journal ArticleDOI
Genome-wide analysis of DNA methylation dynamics during early human development.
Hiroaki Okae,Hatsune Chiba,Hitoshi Hiura,Hirotaka Hamada,Akiko Sato,Takafumi Utsunomiya,Hiroyuki Kikuchi,Hiroaki Yoshida,Atsushi Tanaka,Mikita Suyama,Takahiro Arima +10 more
TL;DR: It was found that the maternal genome was demethylated to a much lesser extent in human blastocysts than in mouse Blastocysts, which could contribute to an increased number of imprinted differentially methylated regions in the human genome.
