A simplified and efficient route to 2'-O, 4'-C-methylene-linked bicyclic ribonucleosides (locked nucleic acid).

TL;DR: A novel efficient method for the synthesis of locked nucleic acid (LNA) monomers is described that easily can be prepared from D-glucose in multigram scale and can be directly converted into LNA-phosphoramidites for automated chemical synthesis of LNA containing oligonucleotides.
Patent

Synthesis of [2.2.1]bicyclo nucleosides

TL;DR: In this paper, a short route to each LNA (Locked Nucleic Acid) derivatives of adenosine, cytosine, uridine, thymidine, and guanidine is demonstrated.

Improved synthesis of [2.2.1]bicyclo nucleosides

TL;DR: In this article, a short route to each LNA (Locked Nucleic Acid) derivatives of adenosine, cytosine, uridine, thymidine and guanidine is demonstrated.
Journal ArticleDOI

Synthesis of 2'-o,3'-c-linked bicyclic nucleosides and bicyclic oligonucleotides

TL;DR: The 3′-C-allyl furanose 4 has been used as a precursor for synthesis of the novel 2′-O,3′- C-linked bicyclic thymine nucleosides 15, 16, 20 and 25 and is incorporated into oligodeoxynucleotides.
Journal ArticleDOI

A novel class of conformationally restricted oligonucleotide analogues: synthesis of 2',3'-bridged monomers and RNA-selective hybridisation

TL;DR: A novel 2′,3′-bicyclic nucleoside 5 has been synthesised and incorporated into oligonucleotide analogues resulting in strong and selective binding to an RNA complement.
