Morphine depresses the twitch and tetanus of stimulated guinea-pig ileum by reducing acetylcholine released from cholinergic nerve endings. Acetylcholine output per shock falls to roughly the same residual amount at varying stimulation rates. Since normal output per shock declines with increasing stimulus frequency, the proportionate effect of morphine diminishes as stimulus frequency rises. Acute “tolerance” to morphine and a state of “morphine-dependence” can be produced. Phenadoxone, dihydromorphinone, metopon, methadone, and heroin are more active, codeine and pethidine less active, than morphine. Nalorphine also depresses the twitch and can desensitize the gut both to itself and to morphine.

Kapazit~ts~nderungen in den Luftwegen dutch Kontraktion oder Ersehtaffung der Bronchialmuskulatur kommen bei dieser Arf. der Registrierung in einer Steigerung bzw. Senkung des intratracheal gemessenen maximalen Inspirationsdruckes (richtiger wohh Aufblasungsdruckes) zur Geltung. Bei dem anderen Verfahren wird direkt oder indirekt das Luft v o 1 u m e n ge~ messen, das die Lunge aufnimmt, wenn man den maximalen Aufblasungsdruck konstant h/~lt und dureh l~berdruekbeatmung ein gleiehbleibendes Volumen anbietet. I4apaziti~tsimderungen in den Luftwegen i~ugern sieh hier in einer Zunahme oder Abnahme der yon der Lunge unter dem gegebenen konsta.nten Aufblasungsdruck aufgenommenen Luftmenge. Die Methode der Druekmessung hat,, wie sehon Kiese ~ hervorhebt, vor allem den Naehteil, daft die Auswirkung yon Anderungen im Kontraktionszustand der Bronchialmuskulatur durch die gleichzeitigen Druckanderungen beeintriichtigt wird. Die Bronchokonstriktion ftihrt zu einer Drucksteigerung, die ihrerseits wieder der Zusammen~iehung der Bronehialmuskulatur entgegen wirken mug; umgekehrt wird eine Ersehlaffung der glatten Muskulatur in den Bronehien dureh die damit~ verbundene Druekentlastung zu einem nur unvollkommenen Erweiterungseffekt fiihren.

Die Synthese von 6 Polypeptiden aus den im Bradykinin vorkommenden Aminosauren wird beschrieben. Das Nonapeptid H-l-Arg-l-Pro-l-Pro-Gly-l-Phe-l-Ser-l-Pro-l-Phe-l-Arg-OH zeigte hohe Bradykinin-ahnliche Wirksamkeit in verschiedenen biologischen Testen.

Afin de mettre en evidence l'influence de faibles modifications de structure sur les proprietes biologiques, les auteurs comparent, a l'aide d'une serie de tableaux systematiques, les structures chimiques et les proprietes pharmacologiques des hormones post-hypophysaires avec celles de pres de quarante de leurs analogues synthetiques, qui ont ete etudies d'une maniere approfondie. Cette confrontation montre qu'il est possible de decouvrir quelques relations limitees entre la specificite biologique de ces corps et certaines particularites de leur structure.

Relationships between the chemical structures and the biological properties of the posterior pituitary hormones and their synthetic analogues

A substance apparently identical with noradrenaline has recently been found in several animal tissues (von Euler, 1946); it has also been obtained in man, from an adrenal tumour (Holton, 1948), and from the blood of an adrenal tumor patient (Vogt, 1949). Noradrenaline may transmit certain sympathetic nerve impulses chemically (Bacq, 1933; Gaddum & Goodwin, 1947; Peart, 1949; Bulbring & Burn, 1949). It is therefore important to find out its actions on different parts of the body and to compare them with those of adrenaline, especially in man. Goldenberg, Pines, Baldwin, Greene & Roh (1948) found that noradrenaline slows the human pulse, diminishes the cardiac output and increases the peripheral resistance; its action in each case being the opposite to that of adrenaline. Allen, Barcroft & Edholm (1946) investigated the action of adrenaline on the circulation in human skeletal muscle; the experiments described here were done with the object of finding out the effects of noradrenaline. The actions of noradrenaline and adrenaline on the heart rate and arterial blood pressure were also examined, as also the action of N-isopropylnoradrenaline ('isopropyl'). In the dog Konzett (1940 a, b) found that isopropyl dilates the bronchi and increases the heart rate and the cardiac output. In these respects it acts like adrenaline, only more powerfully; unlike adrenaline it does not usually cause vasoconstriction-it dilates the blood vessels in the gut and limbs (Konzett, 1940 b) as well as in the lungs (Hebb & Konzett, 1949) and in the skin (Marsh, Pelletier & Ross, 1947).

On the actions of noradrenaline, adrenaline and isopropyl noradrenaline on the arterial blood pressure, heart rate and muscle blood flow in man

Die broncholytische Wirksamkeit einiger Adrenalinderivate, die an Stelle des Methylrestes am Amino-N einen Athyl-, Propyl-, Isopropyl-, Butyl-, Isobutylrest hatten, wurde vergleichend mit Adrenalin untersucht. Isopropyladrenalin erwies sich 10fach, Athyladrenalin 3fach wirksamer als Adrenalin; Propyl- und Butyladrenalin waren ungefahr gleich wirksam wie Adrenalin, Isobutyladrenalin 10mal schwacher wirksam als dieses.
