Journal ArticleDOI
Incorporation of pseudouridine into mRNA yields superior nonimmunogenic vector with increased translational capacity and biological stability.
Katalin Karikó,Hiromi Muramatsu,Frank A. Welsh,Janos Ludwig,Hiroki Kato,Shizuo Akira,Drew Weissman +6 more
TL;DR: It is found that mRNAs containing pseudouridines have a higher translational capacity than unmodified m RNAs when tested in mammalian cells and lysates or administered intravenously into mice at 0.015-0.15 mg/kg doses.
Journal ArticleDOI
Generating the optimal mRNA for therapy: HPLC purification eliminates immune activation and improves translation of nucleoside-modified, protein-encoding mRNA
TL;DR: It is identified that contaminants, including double-stranded RNA, in nucleoside-modified in vitro-transcribed RNA are responsible for innate immune activation and their removal by high performance liquid chromatography results in mRNA that does not induce IFNs and inflammatory cytokines and is translated at 10- to 1000-fold greater levels in primary cells.
Journal ArticleDOI
Zika virus protection by a single low-dose nucleoside-modified mRNA vaccination
Norbert Pardi,Michael J. Hogan,Rebecca S. Pelc,Hiromi Muramatsu,Hanne Andersen,Christina R. DeMaso,Kimberly A. Dowd,Laura L. Sutherland,Richard M. Scearce,Robert Parks,Wendeline Wagner,Alex Granados,Jack Greenhouse,Michelle Walker,Elinor Willis,Jae-Sung Yu,Charles E. McGee,Gregory D. Sempowski,Barbara L. Mui,Ying K. Tam,Yan Jang Huang,Dana L. Vanlandingham,Veronica M. Holmes,Harikrishnan Balachandran,Sujata Sahu,Michelle A. Lifton,Stephen Higgs,Scott E. Hensley,Thomas D. Madden,Michael J. Hope,Katalin Karikó,Sampa Santra,Barney S. Graham,Mark G. Lewis,Theodore C. Pierson,Barton F. Haynes,Drew Weissman +36 more
TL;DR: It is demonstrated that a single low-dose intradermal immunization with nucleoside-modified mRNA–LNP elicits rapid and durable protective immunity and therefore represents a new and promising vaccine candidate for the global fight against ZIKV.
Journal ArticleDOI
Expression kinetics of nucleoside-modified mRNA delivered in lipid nanoparticles to mice by various routes.
Norbert Pardi,Steven Tuyishime,Hiromi Muramatsu,Katalin Karikó,Barbara L. Mui,Ying K. Tam,Thomas D. Madden,Michael J. Hope,Drew Weissman +8 more
TL;DR: It is demonstrated that LNPs are appropriate carriers for mRNA in vivo and have the potential to become valuable tools for delivering mRNA encoding therapeutic proteins.
Journal ArticleDOI
Incorporation of pseudouridine into mRNA enhances translation by diminishing PKR activation
Bart R. Anderson,Hiromi Muramatsu,Subba Rao Nallagatla,Philip C. Bevilacqua,Lauren H Sansing,Drew Weissman,Katalin Karikó +6 more
TL;DR: It is demonstrated that in vitro transcripts containing uridine activate RNA-dependent protein kinase (PKR), which then phosphorylates translation initiation factor 2-alpha (eIF-2α), and inhibits translation.
