Pharmacokinetic comparisons of rutaecarpine and evodiamine after oral administration of Wu-Chu-Yu extracts with different purities to rats.

The invention relates to a method for extracting specnuezhenide crude products (higher than 80 percent) and high-purity specnuezhenide (higher than 98 percent) from fructus ligustri lucidi, which belongs to the field of natural medicine chemistry. The fructus ligustri lucidi is used as a raw material, medicine materials are firstly crushed, a water decoction extraction method is selected, extracting solution is rarefied by macroporous resin columns after being slightly concentrated, then, the alcohol deposition treatment is carried out, and finally, the positive silica gel column purification and the opposite silica gel column purification are selected for preparing the specnuezhenide with different content requirements. The method can be used for preparing the specnuezhenide with lower content in a scale way, the yield is high, the operation is simple, and the cost is low; and the specnuezhenide with higher content can also be prepared, the yield is lower, the cost is higher, the selection can be carried out according to the content requirement, raw material parts and reagent parts during the preparation by the two method can be repeatedly utilized, and the preparation cost is lower than that of the existing preparation method.

Method for preparing specnuezhenide

The invention discloses an iridoid glycoside compound as shown in a formula I and a preparation method and application thereof. The preparation method comprises the following steps: (1) respectively extracting a Reduning injection with ethyl acetate and n-butanol so as to obtain n-butanol extract; (2) subjecting the n-butanol extract to HP-20 macroporous adsorption resin column chromatographic separation and ethanol-water gradient elution so as to obtain a 25-35% ethanol elution part; and (3) successively subjecting the 25-35% ethanol elution part to silica gel column chromatographic separation, ODS column chromatographic separation, HW-40 column chromatographic separation and preparative liquid phase HPLC separation so as to obtain the compound. The compound provided by the invention can be used for treating hand-foot-and-mouth disease.

Iridoid glycoside compound, and preparation method and application thereof

The invention provides a method for preparing a specnuezhenide reference substance. The preparation method specifically comprises the following steps: (i) providing specnuezhenide alcohol extract; (ii), mixing the specnuezhenide alcohol extract and a solvent to obtain a first mixture, and performing column chromatography separation on the first mixture by virtue of an intermediate-pressure preparation column, thereby obtaining a crude specnuezhenide product; (iii) performing recrystallization on the crude specnuezhenide product by virtue of purified water; (iv) separating and drying crystals obtained in step (iii), so as to obtain the specnuezhenide reference substance. The method is stable in process, convenient to operate, high in separation efficiency, low in cost, economical and environment-friendly, and has relatively high economic value and academic value.

Method for preparing specnuezhenide reference substance



A sensitive, reliable liquid chromatography-tandem mass spectrometry (LCMS/
MS) method has been developed and applied to detect the evodiamine (EVO) in rat plasma after
animals were given EVO directly. However there is almost no research on the detection of EVO after
animals were given EVO-loaded solid lipid nanoparticles (EVO-SLN).



In this study, a more sensitive and rapid modified LC-MS/MS method for the quantification
of EVO in rat blood was developed and validated to evaluate the role of SLN in vivo.



Plasma samples were taken from animals orally administered EVO-SLN or free EVO, proteins
were extracted using diethyl ether containing the internal standards (IS) arbidol hydrochloride, and
the mixture was fractionated by liquid chromatography. Quantitative detection of EVO was based on
gradient elution in a mobile phase of acetonitrile-0.2% formic acid in water (70:30, v/v).



The calibration curve was linear (r2>0.999, n=9) over the concentration range from 0.1 to 250
ng/mL. Peaks in triple-quadrupole MS were detected for EVO at m/z 304.2→134.1 and for IS at m/z
479.1→343.0. Mean recovery of EVO was more than 93%. Intra and inter-day precision were within
2.7%. In pharmacokinetics studies, EVO-SLN exhibited much higher bioavailability and absorption
than free EVO.



The developed method in this work can provide a sensitive, effective and rapid process for
the analysis of EVO in whole blood samples. The pharmacokinetics results suggest that the usefulness
of SLN for improving oral bioavailability of poorly soluble drugs.


LC-MS/MS Determination of Evodiamine in Plasma and Its Pharmacokinetics in Rats after Administration in the Form of Solid Lipid Nanoparticles

The invention relates to a process for separating and purifying a special component specnuezhenide of glossy privet fruit, which is a dry ripe fruit of Ligustum lucidum Ait recorded in the books of Chinese Pharmacopoeia in all ages. The current Chinese Pharmacopoeias shows that the component under the item of the traditional Chinese medicament glossy privet fruit is oleanolic acid after identifying the film and measuring the content, and the component is contained in different medical materials, so it is not special. Through research, the specnuezhenide belongs to secoiridoid glycoside compounds, and is the special component of the glossy privet fruit, so the specnuezhenide can be used as an important index for controlling the quality of the glossy privet fruit. The special component has been recorded under the item of the traditional Chinese medicament glossy privet fruit in Chinese Pharmacopoeias 2010 Edition, and is about to be issued and implemented as the content of Chinese Pharmacopoeia. The process for separating and purifying the specnuezhenide is invented by researchers of Institute of Chinese Materia Medica, China Academy of Chinese Academy of Sciences, and is prepared by ethanol extraction, macroporous absorbent resin separation, and reverse silica gel column purification.

Process for separating and purifying special component specnuezhenide of glossy privet fruit

Indian trumpetflower seed is a common traditional Chinese medicine and contains main chemical components of flavonoid compounds, such as baicalein, chrysin, Oroxin A, Oroxin B, baicalin and the like, wherein the Oroxin B is a special component of the Indian trumpetflower seed and can serve as an important index for quality control of the Indian trumpetflower seed medicinal material. An identification method and a contrast for the special component have not been present under the item of Indian trumpetflower seed in the first part of 2005 version Chinese Pharmacopoeia. The Oroxin B prepared by separation serving as a contrast in a thin-layer identification method for the specificity of the traditional Chinese medicine Indian trumpetflower seed, has been written under the item of the traditional Chinese medicine Indian trumpetflower seed in the first part of 2010 version Chinese Pharmacopoeia, and is about to serve as the content of Chinese Pharmacopoeia to be published and implemented. A method for preparing the special component of traditional Chinese medicine Indian trumpetflower seed, namely Oroxin B is invented by scientific research personnel of Institute of Chinese Materia Medica, China Academy of Chinese Medical Sciences through years of experiments. The Oroxin B is prepared by ethanol extraction, polyamide separation, and glucan gel purification.

Method for preparing special component of traditional Chinese medicine Indian trumpetflower seed, namely Oroxin B

OBJECTIVE To establish a SPE-HPLC method for the determination and pharmacokinetic study of evodiamine and rutacarpine in rat plasma. METHOD A Kromasil C18 column (4.6 mm x 250 mm, 5 microm) was used with acetonitrile-water-tetrahydrofuran-acetic acid (51:48:1:0.1) as a mobile phase and at a flow rate of 1 mL x min(-1), and the UV detection was at 225 nm. The column temperature was 35 degrees C. After the analytes were extracted from the plasma of rats by solid phase extraction (SPE), the content of evodiamine and rutaecarpine was measuared by HPLC method using halcinonide as an internal standard solution. RESULT After transdermal administration to rats, the pharmacokinetic behavior of evodiamine and rutaecarpine belongs to the one-compartment model. The main pharmacokinetic parameters was as follows: K(a) 0.224 h(-1) and 0.220 h(-1), K(e) 0.114 h(-1) and 0.118 h(-1), C(max) 0.211 mg x L(-1) and 0.272 mg x L(-1), T(peak) 6.132 h and 6.102 h, respectively. CONCLUSION The method in this study is simple, rapid and sensitive. It is proved to be suitable for pharmacokinetic study of evodiamine and rutaecarpine.

[Determination of evodiamine and rutaecarpine of compound Wuzhuyu cataplasm in plasma by SPE-HPLC: application to its pharmacokinetics].

Abstract Background The proportion of patients with obstructive sleep apnea–hypopnea syndrome (OSAHS) is increasing year by year in China, which has become a major public health problem. Self-management of OSAHS and multiple support from caregivers are key to low hospital admissions and high quality of life for patients with OSAHS. Social support and health literacy are the main promoters of self-management behavior. However, their contributions have not been adequately studied. The purpose of this study is to investigate the level of self-management among patients with OSAHS and its relationship with general demographics, social support, and health literacy. Methods A total of 280 patients with OSAHS treated in two Classiii Grade A hospitals in Jinzhou City, Liaoning Province from October 2020 to July 2021 were selected as the study subjects. Patients were investigated by General Characteristics Questionnaire, Social Support Rating Scale (SSRS), Health Literacy Scale for Chronic Patients (HLSCP), and OSAHS Self-management Behavior Questionnaire, and the influencing factors of self-management of patients with OSAHS were analyzed. Results The average score of OSAHS self-management was 74.49(SD = 8.06), SSRS and HLSCP scores were positively correlated with total scores of self-management behavior. Furthermore, we found that disease duration, SSRS, and HLSCP scores were the main predictors of self-management behavior (R 2 = 0.390, P &lt; 0.001). Conclusion This study found that OSAHS patients with a longer duration of disease and higher SSRS or HLSCP scores also had higher levels of self-management. The factors discussed in this study may be helpful in developing individualized interventions in self-management for patients with OSAHS.

Self-management behavior, associated factors and its relationship with social support and health literacy in patients with obstructive sleep apnea–hypopnea syndrome

Abstract Background Obstructive sleep apnoea (OSA) is the most common sleep-related breathing disorder, yet it remains undiagnosed in a large proportion of adults. Objective This study aims to investigate the status of patient delay and provider delay in OSA patients and examine related factors affecting patient delay in OSA individuals in China. Methods A cross-sectional design was conducted on a sample of 309 OSA patients (aged from 18 to 76, median age of 47 years, 84.8% male) in Northeast China. Participants were required to complete the sociodemographic questionnaire, the symptom characteristics questionnaire, the help-seeking attitude scale (HSAS), the social support rating scale and the chronic disease self-efficacy scales (CDSES) to test the hypothesis. Binary logistic regression analysis was conducted to explore factors that account for the patient delay. Results The median patient delay among OAS patients in this study was 22 months, the median provider delay was one month, and the median total delay was 26 months. As shown by multivariate analysis results, patients who have snored for over 6 years (OR = 3.377, 95%CI: 1.175–9.702) were more likely to experience prolonged patient delays. Per capita monthly family income above 3000 RMB (OR = 0.172, 95%CI: 0.052–0.571), taking up residence in cities or towns (OR = 0.484, 95%CI: 0.248–0.946), higher self-recognition of the disease (OR = 0.793, 95%CI: 0.647–0.972), higher objective support (OR = 0.825, 95%CI: 0.739–0.921) and stronger self-efficacy (OR = 0.674, 95%CI: 0.525–0.867) were significantly associated with shorter patient delays. Conclusion Patient delay is common in Chinese OSA patients. The upstream factors affecting the patient delay in individuals with OSA include income, place of residence, and objective support; midstream factors include self-recognition of the disease and self-efficacy; downstream factors include years of snoring. KEY MESSAGES Despite being a high-prevalence disease, many obstructive sleep apnoea (OSA) patients are not clearly diagnosed and treated. The factors affecting the delay in seeking medical treatment in individuals with OSA included income, place of residence, objective support, self-recognition of the disease, self-efficacy and years of snoring. Investigations into OSA patients’ care-seeking behaviours can better reflect the secondary prevention of OSA, and it is crucial to pay attention to the delayed phase of patients.
