2,3-Pyrrolidinedicarboxylates as Neurotransmitter Conformer Mimics: Enantioselective Synthesis via Chelation-Controlled Enolate Alkylation

TL;DR: A diastereo- and enantioselective synthesis of the four stereoisomers of 2,3-pyrrolidinedicarboxylic acid (an analog of aspartic acid) is described, with a Rapoport-type aspartate alkylation that can be controlled to give good yields of either diastereomer as a function of enolate geometry.

TL;DR: In vitro and in vivo excitotoxic properties of a novel conformationally constrained analogue of L-glutamate, L-trans-2,3-pyrrolidine dicarboxylate were investigated and damage could be significantly reduced by co-administration of MK-801, but not by NBQX.

Efficient syntheses of diastereoselectively labeled (3S)-[4-13C]-l-valine, and regioselectively labeled [3-13CH3]-l-isoleucine hydrochlorides

TL;DR: Diastereoselectically labeled (3S)-[4-13C]-l-valine and regioselectively labeled [3-13CH3]- l-isoleucine hydrochlorides were synthesized in 54% and 62% respective overall yields from l-aspartic acid.
