Journal ArticleDOI
Oral administration of a potent and selective non-peptidic BACE-1 inhibitor decreases beta-cleavage of amyloid precursor protein and amyloid-beta production in vivo.
Ishrut Hussain,Julie Hawkins,David G. Harrison,Christopher J. Hille,Gareth Wayne,Leanne Cutler,Tania Buck,Daryl S. Walter,Emmanuel Hubert Demont,C. Howes,Alan Naylor,Philip Jeffrey,Maria I. Gonzalez,Colin Dingwall,Anton D. Michel,Sally Redshaw,John B. Davis +16 more
TL;DR: A selective non‐peptidic BACE‐1 inhibitor is designed, GSK188909, that potently inhibits β‐cleavage of APP and reduces levels of secreted and intracellular Aβ in SHSY5Y cells expressing APP and can effectively lower brain Aβin vivo.
Journal ArticleDOI
BACE1 (β-secretase) transgenic and knockout mice: identification of neurochemical deficits and behavioral changes
Steve Mark Harrison,Alex J. Harper,Julie Hawkins,Graham Duddy,Evelyn Grau,Pippa L Pugh,Panida Winter,Claire S. Shilliam,Zoe A. Hughes,Lee A. Dawson,M. Isabel Gonzalez,Neil Upton,Menelas N. Pangalos,Colin Dingwall +13 more
TL;DR: The results suggest an unexpected role for BACE1 in neurotransmission, perhaps through changes in amyloid precursor protein processing and Abeta levels.
Journal ArticleDOI
Synthesis and muscarinic activities of quinuclidin-3-yltriazole and -tetrazole derivatives.
Harry John Wadsworth,Sarah Margaret Jenkins,Barry Sidney Orlek,Frederick Cassidy,Michael S. G. Clark,Frank Brown,Graham J. Riley,Diane Graves,Julie Hawkins,Christopher B. Naylor +9 more
TL;DR: A relationship between position and depth of the electrostatic minima relative to the azabicyclic ring and the potency and efficacy of the compounds was determined.
Journal ArticleDOI
Substituent variation in azabicyclic triazole- and tetrazole-based muscarinic receptor ligands.
Sarah Margaret Jenkins,Harry John Wadsworth,Steven Mark Bromidge,Barry Sidney Orlek,Wyman Paul Adrian,Graham J. Riley,Julie Hawkins +6 more
TL;DR: These compounds provide muscarinic ligands with high potency and a range of efficacies suitable for testing as candidate drugs in the treatment of Alzheimer's disease.
Journal ArticleDOI
Characterization of the Ectodomain Shedding of the β-Site Amyloid Precursor Protein-cleaving Enzyme 1 (BACE1)
TL;DR: This work confirms that the ectodomain of BACE1 is secreted from cells and that this processing is up-regulated by agents that activate protein kinase C, and shows that ADAM10 is a strong candidate for the Bace1 sheddase.
