Journal ArticleDOI
Development of a Drug Screening Platform Based on Engineered Heart Tissue
Arne Hansen,Alexandra Eder,Marlene Bönstrup,Marianne Flato,Marco Mewe,Sebastian Schaaf,Bülent Aksehirlioglu,Alexander Schwörer,June Uebeler,Thomas Eschenhagen +9 more
TL;DR: A simple technique to construct large series of EHT and automatically evaluate contractile activity is developed and shall be useful for drug screening and disease modeling.
Journal ArticleDOI
In vitro perfusion of engineered heart tissue through endothelialized channels.
Ingra Vollert,Moritz Seiffert,Johanna Bachmair,Merle Sander,Alexandra Eder,Lenard Conradi,Alexander Vogelsang,Thomas G. Schulze,June Uebeler,Wolfgang Holnthoner,Heinz Redl,Hermann Reichenspurner,Arne Hansen,Thomas Eschenhagen +13 more
TL;DR: In this paper, a miniaturized fibrin-based EHT format was expanded to a larger six-well format with six flexible silicone posts holding each EHT (15×25×3 mm3).
Journal ArticleDOI
Contractile abnormalities and altered drug response in engineered heart tissue from Mybpc3-targeted knock-in mice
Andrea Stöhr,Felix W. Friedrich,Frederik Flenner,Birgit Geertz,Alexandra Eder,Sebastian Schaaf,Marc N. Hirt,June Uebeler,Saskia Schlossarek,Lucie Carrier,Arne Hansen,Thomas Eschenhagen +11 more
TL;DR: The present findings in auxotonically contracting EHTs support the idea that cMyBP-C's normal role is to suppress force generation at low intracellular Ca(2+) and stabilize the power-stroke step of the cross bridge cycle.
Journal ArticleDOI
Guanabenz interferes with ER stress and exerts protective effects in cardiac myocytes.
Christiane Neuber,June Uebeler,Thomas G. Schulze,Hannieh Sotoud,Ali El-Armouche,Thomas Eschenhagen +5 more
TL;DR: It is confirmed that guanabenz interferes with ER stress-signalling and has protective effects on cell survival and the modest protection in EHTs points to more complex mechanisms of force regulation in intact functional heart muscle.
Journal ArticleDOI
Effects of proarrhythmic drugs on relaxation time and beating pattern in rat engineered heart tissue
Alexandra Eder,Arne Hansen,June Uebeler,Thomas G. Schulze,Christiane Neuber,Sebastian Schaaf,Lei Yuan,Torsten Christ,Marc A. Vos,Thomas Eschenhagen +9 more
TL;DR: Insensitivity to IKr inhibitors makes rat EHTs unsuitable as general proarrhythmia screen, but favors detection of effects on Ito, Iks + Ito or IKs–+ IKr, which suggests that effects on these currents are more common than anticipated.
