Analytical Protein A Chromatography as a Quantitative Tool for the Screening of Methionine Oxidation in Monoclonal Antibodies
Caroline Loew,Constanze Knoblich,Jürgen Fichtl,Nadja Alt,Katharina Diepold,Patrick Bulau,Pierre Goldbach,Michael Adler,Hanns-Christian Mahler,Ulla Grauschopf +9 more
TL;DR: Analytical protein A chromatography is established as a method of choice for fast and quantitative screening of total Fc methionine oxidation in therapeutic antibodies during formulation and process development and it is demonstrated that highly conserved Fc methamphetamineionines situated close to the binding site to protein A can serve as marker for the oxidation of other surface-exposed methamphetamineionine residues.
Journal ArticleDOI
Multiattribute Monitoring of Antibody Charge Variants by Cation-Exchange Chromatography Coupled to Native Mass Spectrometry.
Markus Haberger,Anna-Katharina Heidenreich,Michaela Hook,Jürgen Fichtl,Rainer Lang,Florian Cymer,Mahdi Adibzadeh,Felix Kuhne,Harald Wegele,Dietmar Reusch,Lea Bonnington,Patrick Bulau +11 more
TL;DR: In this article, an online cation-exchange chromatography native electrospray mass spectrometry method (CEC-UV-MS) was developed for direct TCB Mab charge variant monitoring during bioprocess and formulation development.
Journal ArticleDOI
Identification of Hetero-aggregates in Antibody Co-formulations by Multi-dimensional Liquid Chromatography Coupled to Mass Spectrometry
Felix Kuhne,Katrin Heinrich,Martin Winter,Jürgen Fichtl,Gabriel Hoffmann,Franziska Zähringer,Katharina Spitzauer,Monika M. Meier,Tarik A. Khan,Lea Bonnington,Katharina Wagner,Jan Olaf Stracke,Dietmar Reusch,Harald Wegele,Michael Mormann,Patrick Bulau +15 more
TL;DR: In this article , a multi-dimensional liquid chromatography coupled to tandem mass spectrometry method for antibody homo- versus hetero-aggregate characterization is described, and the results demonstrate the high stability of the co-formulation, where only minor aggregate formation is observed upon product storage and accelerated temperature or light-stress conditions.
