TL;DR: It is found that tumors are rigid because they have a stiff stroma and elevated Rho-dependent cytoskeletal tension that drives focal adhesions, disrupts adherens junctions, perturbs tissue polarity, enhances growth, and hinders lumen formation.

Demystifying the Effects of a Three‐Dimensional Microenvironment in Tissue Morphogenesis
TL;DR: 3D organotypic models have been developed to clarify the mechanisms of epithelial morphogenesis and the subsequent maintenance of tissue homeostasis and the application of these 3D culture models are described to illustrate how the microenvironment plays a critical role in regulating mammary tissue function and signaling.
