Journal ArticleDOI
Tensional homeostasis and the malignant phenotype.
Matthew J. Paszek,Nastaran Zahir,Kandice R. Johnson,Johnathon N. Lakins,Gabriela I. Rozenberg,Amit Gefen,Cynthia A. Reinhart-King,Susan S. Margulies,Micah Dembo,David Boettiger,Daniel A. Hammer,Valerie M. Weaver +11 more
TL;DR: It is found that tumors are rigid because they have a stiff stroma and elevated Rho-dependent cytoskeletal tension that drives focal adhesions, disrupts adherens junctions, perturbs tissue polarity, enhances growth, and hinders lumen formation.
Journal ArticleDOI
Polymersomes: tough vesicles made from diblock copolymers.
Bohdana M. Discher,You-Yeon Won,David S. Ege,James C. Lee,Frank S. Bates,Dennis E. Discher,Daniel A. Hammer +6 more
TL;DR: The results suggest a new class of synthetic thin-shelled capsules based on block copolymer chemistry, and both the membrane bending and area expansion moduli of electroformed polymersomes (polymer-based liposomes) fell within the range of lipid membrane measurements.
Journal ArticleDOI
Lifetime of the P-selectin-carbohydrate bond and its response to tensile force in hydrodynamic flow.
TL;DR: It is reported here that transient tethers had first-order kinetics and other characteristics suggesting a unimolecular interaction between P-selectin and its glycoprotein ligand (PSGL-1) and fast on and off rates appear necessary to support rolling at physiological shear stresses.
Journal ArticleDOI
Self-Assembly of Janus Dendrimers into Uniform Dendrimersomes and Other Complex Architectures
Virgil Percec,Daniela A. Wilson,Pawaret Leowanawat,Christopher J. Wilson,Andrew D. Hughes,Mark S. Kaucher,Daniel A. Hammer,Dalia H. Levine,Anthony J. Kim,Frank S. Bates,Kevin P. Davis,Timothy P. Lodge,Michael L. Klein,Russell DeVane,Emad Aqad,Brad M. Rosen,Andreea O. Argintaru,Monika J. Sienkowska,Kari Rissanen,Sami Nummelin,Sami Nummelin,Jarmo Ropponen +21 more
TL;DR: Dendrimersomes marry the stability and mechanical strength obtainable from polymersomes with the biological function of stabilized phospholipid liposomes, plus superior uniformity of size, ease of formation, and chemical functionalization, providing access to systematic tuning of molecular structure and of self-assembled architecture.
Journal ArticleDOI
The cancer glycocalyx mechanically primes integrin-mediated growth and survival
Matthew J. Paszek,Christopher C. DuFort,Olivier Rossier,Russell Bainer,Janna K. Mouw,Kamil Godula,Jason E. Hudak,Jonathon N. Lakins,Amanda C. Wijekoon,Luke Cassereau,Matthew G. Rubashkin,Mark Jesus M. Magbanua,Kurt S. Thorn,Michael W. Davidson,Hope S. Rugo,John W. Park,Daniel A. Hammer,Grégory Giannone,Carolyn R. Bertozzi,Valerie M. Weaver +19 more
TL;DR: A bulky glycocalyx is a feature of tumour cells that could foster metastasis by mechanically enhancing cell-surface receptor function by increasing integrin adhesion and signalling.
