Bioremediation of polluted temperate and cold temperature environments may require the activity of psychrotrophic bacteria, because their low temperature growth range parallels the ambient temperatures encountered in these environments. In the present study, 135 psychrotrophic microorganisms isolated from a variety of ecosystems in Canada were examined for their ability to mineralize 14C-labelled toluene, naphthalene, dodecane, hexadecane, 2-chlorobiphenyl, and pentachlorophenol. A number of the psychrotrophic strains mineralized toluene, naphthalene, dodecane, and hexadecane. None of the psychrotrophs were capable of mineralizing 2-chlorobiphenyl or pentachlorophenol. Assessment of the biodegradation potential of psychrotrophic microorganisms.

Chloroaromatics are xenobiotic compounds of environmental concern. They can be removed from the environment by (bio)degradation or by (bio)transformation. Recognition of the mechanisms and requirements of their biodegradation is of cardinal importance for understanding the fate of these chemicals in the environment, and for developing methods for biological treatment of wastes containing compounds of this type. Cleavage of the carbon-halogen bond is the critical step in degradation of chloroaromatics. As exemplified with chlorophenols, chlorobenzoates and chlorobenzenes in this review, two distinct strategies are employed by bacteria for degradation of chlorinated aromatic compounds: the particular chlorine substituents are removed either directly from the aromatic ring (as an initial step in degradation) or after oxygenative ring cleavage (from chlorinated aliphatic intermediates). Direct elimination of chlorine substituents from the aromatic ring occurs by displacement with either hydroxyl groups (hydrolytically or oxygenolytically) or hydrogen atoms (reductive dechlorination). Dechlorinations of the latter type require reducing power and are significant in anaerobic environments, but have also been observed with strictly aerobic bacteria. Various biotransformation reactions, with only minor alteration of the parent compound, are an alternative to biogradation. Two environmentally significant transformation reactions discussed here are O-methylation and O-demethylation. The capability to O-methylate chlorinated hydroxybenzenes seems to be widespread in bacteria. O-Methylation is an environmentally important transformation reaction, since methylation increases the lipophilicity of the compound and thus the potential for bioaccumulation. Bacterial O-demethylation of chlorinated methoxylated compounds has been observed under both aerobic and anaerobic conditions.

Mechanisms of bacterial degradation and transformation of chlorinated monoaromatic compounds.

The steady-state growth of a Flavobacterium strain known to utilize pentachlorophenol (PCP) was examined when cellobiose and PCP simultaneously limited its growth rate in continuous culture. A concentration of 600 mg of PCP per liter in influent medium could be continuously degraded without affecting steady-state growth. We measured specific rates of PCP carbon degradation as high as 0.15 +/- 0.01 g (dry weight) of C per h at a growth rate of 0.045 h-1. Comparable specific rates of PCP degradation were obtained and maintained by PCP-adapted, natural consortia of epilithic microorganisms. The consortium results suggest that a fixed-film bioreactor containing a PCP-adapted natural microbial population could be used to treat PCP-contaminated water.

Pentachlorophenol degradation: a pure bacterial culture and an epilithic microbial consortium.

Two toluene-degrading strains, T103 and T104, were isolated from rock surface biomass in a freshwater stream contaminated with toluene. The strains exhibit different capacities for degradation of toluene and other aromatic compounds and have characteristics of the genus Mycobacterium. Both are aerobic, rod-shaped, gram-positive, nonmotile, and acid-alcohol fast and produce yellow pigments. They have mainly straight-chain saturated and monounsaturated fatty acids with 10 to 20 carbon atoms and large amounts of tuberculostearic acid that are typical of mycobacteria. Fatty acid analyses indicate that T103 and T104 are different mycobacterial strains that are related at the subspecies level. Their identical 16S rDNA sequences are most similar to Mycobacterium aurum and Mycobacterium komossense, and they constitute a new species of fast-growing mycobacteria. Ecological studies reveal that toluene contamination has enriched for toluene-degrading bacteria in the epilithic microbial community. Strains T103 and T104 play only a small role in toluene degradation in the stream, although they are present in the habitat and can degrade toluene. Other microorganisms are consequently implicated in the biodegradation.
