Journal ArticleDOI
Evidence for altered thiamine metabolism in diabetes: Is there a potential to oppose gluco- and lipotoxicity by rational supplementation?
TL;DR: It could be concluded that although experimental studies mostly resulted in beneficial effects, clinical studies of appropriate size and duration focusing on the effect of thiamine supplementation/therapy on hard endpoints are missing at present.
Journal ArticleDOI
Hyperuricemia contributes to the faster progression of diabetic kidney disease in type 2 diabetes mellitus
Vendula Bartáková,Katarína Kuricová,Lukáš Pácal,Zuzana Nová,Veronika Dvořáková,Martina Švrčková,Denisa Malúšková,Ivana Svobodová,Jitka Řehořová,Jan Svojanovský,Jindřich Olšovský,Jana Bělobrádková,Kateřina Kaňková +12 more
TL;DR: It is demonstrated that initial hyperuricemia or need for allopurinol is an independent risk factor for diabetic kidney disease progression and that SUA levels in diabetic subjects conferring protection against DKD progression might be lower than current cut-offs for general population.
Journal ArticleDOI
Role of thiamine status and genetic variability in transketolase and other pentose phosphate cycle enzymes in the progression of diabetic nephropathy
Lukáš Pácal,Josef Tomandl,Jan Svojanovský,Darja Krusová,Soňa Štěpánková,Jitka Řehořová,Jindřich Olšovský,Jana Bělobrádková,Veronika Tanhäuserová,Marie Tomandlová,Jan Mužík,Kateřina Kaňková +11 more
TL;DR: The results support the role of 'functional' thiamine deficiency in the development of hyperglycaemia-related pathology and limited intracellular availability of active TKT co-factor seems to be a dominant abnormality.
Journal ArticleDOI
Parameters of oxidative stress, DNA damage and DNA repair in type 1 and type 2 diabetes mellitus
Lukáš Pácal,Jana Varvařovská,Zdeněk Rušavý,Silva Lacigová,Rudolf Štětina,Jaroslav Racek,Renata Pomahačová,Veronika Tanhäuserová,Kateřina Kaňková +8 more
TL;DR: Significant age- and DM type-related changes of oxidative DNA modification and capacity for its repair in subjects with T1DM and T2DM are demonstrated.
Journal ArticleDOI
Genetic risk factors for diabetic nephropathy on chromosomes 6p and 7q identified by the set-association approach.
Kateřina Kaňková,Andrea Stejskalová,Lukáš Pácal,Svatava Tschöplová,Miluše Hertlová,Darja Krusová,Lydie IzakovičováHollá,Michal Beránek,Anna Vašků,Sandra Barral,Jurg Ott +10 more
TL;DR: Using the set-association approach, this study identified significant associations of several SNPs on chromosomes 6 and 7 with DN, and identified SNPs AGER 2184A/G and NOS3 774C/T, together with diabetes duration and HbA1c, as significant predictors of DN.
