The halogen bond occurs when there is evidence of a net attractive interaction between an electrophilic region associated with a halogen atom in a molecular entity and a nucleophilic region in another, or the same, molecular entity. In this fairly extensive review, after a brief history of the interaction, we will provide the reader with a snapshot of where the research on the halogen bond is now, and, perhaps, where it is going. The specific advantages brought up by a design based on the use of the halogen bond will be demonstrated in quite different fields spanning from material sciences to biomolecular recognition and drug design.

The Halogen Bond

Just over a century ago, Lewis published his seminal work on what became known as the covalent bond, which has since occupied a central role in the theory of making organic molecules. With the advent of covalent organic frameworks (COFs), the chemistry of the covalent bond was extended to two- and three-dimensional frameworks. Here, organic molecules are linked by covalent bonds to yield crystalline, porous COFs from light elements (boron, carbon, nitrogen, oxygen, and silicon) that are characterized by high architectural and chemical robustness. This discovery paved the way for carrying out chemistry on frameworks without losing their porosity or crystallinity, and in turn achieving designed properties in materials. The recent union of the covalent and the mechanical bond in the COF provides the opportunity for making woven structures that incorporate flexibility and dynamics into frameworks.

Halogen Bonding in Supramolecular Chemistry.

Halogen bonding has been known in material science for decades, but until recently, halogen bonds in protein–ligand interactions were largely the result of serendipitous discovery rather than rational design. In this Perspective, we provide insights into the phenomenon of halogen bonding, with special focus on its role in drug discovery. We summarize the theoretical background defining its strength and directionality, provide a systematic analysis of its occurrence and interaction geometries in protein–ligand complexes, and give recent examples where halogen bonding has been successfully harnessed for lead identification and optimization. In light of these data, we discuss the potential and limitations of exploiting halogen bonds for molecular recognition and rational drug design.

Iodotriazole and triazole anion recognition groups have been integrated into a picket-fence zinc(II)-metalloporphyrin scaffold to produce receptors for anion recognition and sensing applications. 1H NMR and UV/visible spectroscopic investigations reveal both host systems exhibit strong anion binding affinities in a range of solvent media. Importantly, the halogen-bonding iodotriazole-containing porphyrin-based host displays halide binding affinities substantially larger than the protic-functionalised analogue concomitant with a reduced strength of oxoanion complexation.

Interlocked molecules have captured chemists’ imagination owing to their nontrivial topology and the promise of their potential nanotechnological uses as molecular machines[1] and in chemical sensor applications.[2] The synthesis of such sophisticated architectures is a challenge, and as a consequence this has necessitated the implementation of imaginative cation,[3] anion,[4] and neutral[5] templation methodologies, which use a combination of complementary Lewis acid–base, electrostatic, and hydrogen-bonding interactions for component assembly. Halogen bonding (XB) is the attractive highly directional, noncovalent interaction between an electron-deficient halogen atom and a Lewis base.[6] The scope of XB in solid-state crystal engineering has been intensively explored for a number of years,[7] however, in spite of the complementary analogy to ubiquitous hydrogen bonding, it is only in recent times that investigations into the application of solution-phase XB interactions to molecular recognition processes, self-assembly, and catalysis have resulted in this field developing rapidly.[8] Indeed, in conjunction with anion templation, we have used XB to assemble interpenetrated and interlocked molecular frameworks.[8b–d]

A Catenane Assembled through a Single Charge‐Assisted Halogen Bond

Triazole and triazolium groups have been integrated into a zinc(II) metalloporphyrin-based structural framework to produce two porphyrin-cages for anion sensing applications. UV/visible spectroscopic titration investigations reveal both host systems exhibit strong anion binding affinities, with the positively-charged triazolium-porphyrin cage capable of colorimetric sensing halides, fluoride and chloride, and oxoanions in acetone–water solvent mixtures.
