Journal ArticleDOI
Ebola virus entry requires the host-programmed recognition of an intracellular receptor
Emily Happy Miller,Gregor Obernosterer,Matthijs Raaben,Andrew S. Herbert,Maika S. Deffieu,Anuja Krishnan,Esther Ndungo,Rohini G. Sandesara,Jan E. Carette,Ana I. Kuehne,Gordon Ruthel,Suzanne R. Pfeffer,John M. Dye,Sean P. J. Whelan,Thijn R. Brummelkamp,Kartik Chandran +15 more
TL;DR: These findings support a model in which GP cleavage by endosomal cysteine proteases unmasks the binding site for NPC1, and GP–NPC1 engagement within lysosomes promotes a late step in entry proximal to viral escape into the host cytoplasm.
Journal ArticleDOI
Niemann–Pick type C 1 function requires lumenal domain residues that mediate cholesterol-dependent NPC2 binding
TL;DR: It is shown that the second lumenal domain of NPC1 binds directly to NPC2 protein, a finding that supports directional transfer of cholesterol from NPC2 onto NPC1’s N-terminal domain, and human disease-causing mutations in NPC1 domain 2 decrease NPC2 binding, suggesting that NPC1 binding is necessary for NPC1 function in humans.
Journal ArticleDOI
Glycosylation inhibition reduces cholesterol accumulation in NPC1 protein-deficient cells
TL;DR: It is shown here that the requirement for NPC1 is decreased in cells containing lower levels of protein glycosylation, which supports a model in which NPC1 protein helps transfer cholesterol past the glycocalyx barrier that lines the interior of lysosomes.
