Conjugated Microporous Poly(aryleneethynylene) Networks

The convergence of our understanding of structure-property relationships for selected biological macromolecules and our increased ability to prepare large synthetic macromolecules with a structural precision that approaches that of proteins have spawned a new area of research where chemistry and materials science join with biology. While evolution has enabled nature to perfect processes involving energy transfer or catalysis by incorporating functions such as self-replication and repair, synthetic macromolecules still depend on our synthetic skills and abilities to mesh structure and function in our designs. Clearly, we can take advantage of our understanding of natural systems to mimic the structural features that lead to optimized function. For example, numerous biological systems make use of the concept of site isolation whereby an active center or catalytic site is encapsulated, frequently within a protein, to afford properties that would not be encountered in the bulk state. The ability of the dendritic shell to encapsulate functional core moieties and to create specific site-isolated nanoenvironments, and thereby affect molecular properties, has been explored. By utilizing the distinct properties of the dendrimer architecture active sites that have either photophysical, photochemical, electrochemical, or catalytic functions have been placed at the core. Applying the general concept of site isolation to problems in materials research is likely to prove extremely fruitful in the long term, with short-term applications in areas such as the construction of improved optoelectronic devices. This review focuses on the evolution of a natural design principle that contributes to bridging the gap between biology and materials science. The recent progress in the synthesis of dendrimer-encapsulated molecules and their study by a variety of techniques is discussed. These investigations have implications that range from the preliminary design of artificial enzymes, catalysts, or light-harvesting systems to the construction of insulated molecular wires, light-emitting diodes, and fiber optics.

Dendritic Encapsulation of Function: Applying Nature's Site Isolation Principle from Biomimetics to Materials Science.

Tetrathiafulvalene (TTF) and its derivatives were originally prepared as strong electron-donor molecules for the development of electrically conducting materials. This Review emphasizes how TTF and its derivatives offer new and in some cases little-exploited possibilities at the molecular to the supramolecular levels, as well as in macromolecular aspects. TTF is a well-established molecule whose interest goes beyond the field of materials chemistry to be considered an important building block in supramolecular chemistry, crystal engineering, and in systems able to operate as machines. At the molecular level, TTF is a readily available molecule which displays a strong electron-donor ability. However, its use as a catalyst for radical-polar crossover reactions, thus mimicking samarium iodide chemistry, has only recently been addressed. Important goals have been achieved in the use of TTF at the macromolecular level where TTF-containing oligomers, polymers, and dendrimers have allowed the preparation of new materials that integrate the unique properties of TTF with the processability and stability that macromolecules display. The TTF molecule has also been successfully used in the construction of redox-active supramolecular systems. Thus, chemical sensors and redox-switchable ligands have been prepared from TTF while molecular shuttles and molecular switches have been prepared from TTF-containing rotaxanes and catenanes. A large synthetic effort has been devoted to the preparation of the so-called organic ferromagnets, many of which are derived from TTF. The main task in these systems is the introduction of ferromagnetic coupling between the conduction electrons and localized electrons. TTF has also played a prominent role in molecular electronics where TTF-containing D-sigma-A molecules have allowed the preparation of the first confirmed unimolecular rectifier. Recently, it has been confirmed that TTF can display efficient nonlinear optic (NLO) responses in the second and third harmonic generation as well as a good thermal stability. These findings can be combined with the redox ability of TTF as an external stimuli to provide a promising strategy for the molecular engineering of switchable NLO materials. Fullerenes endowed with TTF exhibit outstanding photophysical properties leading to charge-separated (CS) states that show remarkable lifetimes.

New Concepts in Tetrathiafulvalene Chemistry.

The use of photochemical transformations is a powerful strategy that allows for the formation of a high degree of molecular complexity from relatively simple building blocks in a single step. A central feature of all light-promoted transformations is the involvement of electronically excited states, generated upon absorption of photons. This produces transient reactive intermediates and significantly alters the reactivity of a chemical compound. The input of energy provided by light thus offers a means to produce strained and unique target compounds that cannot be assembled using thermal protocols. This review aims at highlighting photochemical transformations as a tool for rapidly accessing structurally and stereochemically diverse scaffolds. Synthetic designs based on photochemical transformations have the potential to afford complex polycyclic carbon skeletons with impressive efficiency, which are of high value in total synthesis.

Photochemical Approaches to Complex Chemotypes: Applications in Natural Product Synthesis

Bicyclo[1.1.1]pentanes, [n]Staffanes, [1.1.1]Propellanes, and Tricyclo[2.1.0.02,5]pentanes

Hypoxia is a hallmark of solid tumors, is associated with local invasion, metastatic spread, resistance to chemo- and radiotherapy, and is an independent, negative prognostic factor for a diverse range of malignant neoplasms. The cellular response to hypoxia is primarily mediated by a family of transcription factors, among which hypoxia-inducible factor 1 (HIF1) plays a major role. Under normoxia, the oxygen-sensitive α subunit of HIF1 is rapidly and constitutively degraded but is stabilized and accumulates under hypoxia. Upon nuclear translocation, HIF1 controls the expression of over 100 genes involved in angiogenesis, altered energy metabolism, antiapoptotic, and pro-proliferative mechanisms that promote tumor growth. A designed transcriptional antagonist, dimeric epidithiodiketopiperazine (ETP 2), selectively disrupts the interaction of HIF1α with p300/CBP coactivators and downregulates the expression of hypoxia-inducible genes. ETP 2 was synthesized via a novel homo-oxidative coupling of the aliphatic primary carbons of the dithioacetal precursor. It effectively inhibits HIF1-induced activation of VEGFA, LOX, Glut1, and c-Met genes in a panel of cell lines representing breast and lung cancers. We observed an outstanding antitumor efficacy of both (±)-ETP 2 and meso-ETP 2 in a fully established breast carcinoma model by intravital microscopy. Treatment with either form of ETP 2 (1 mg/kg) resulted in a rapid regression of tumor growth that lasted for up to 14 days. These results suggest that inhibition of HIF1 transcriptional activity by designed dimeric ETPs could offer an innovative approach to cancer therapy with the potential to overcome hypoxia-induced tumor growth and resistance.

Suppression of tumor growth by designed dimeric epidithiodiketopiperazine targeting hypoxia-inducible transcription factor complex.

Direct fluorination of dimethyl bicyclo[1.1.1]pentane-1,3-dicarboxylate, obtained from [1.1.1]propellane prepared by an improved synthetic procedure, furnished esters of 14 of the 15 possible bridge-fluorinated bicyclo[1.1.1]pentane-1,3-dicarboxylic acids, isolated by preparative GC. Calculated geometries reflect the substitution pattern in a regular fashion compatible with Bent's rules. Considerable additional strain is introduced into the bicyclo[1.1.1]pentane cage by polyfluorination; it is calculated to be as high as 33−35 kcal/mol for hexasubstitution. Three arrangements of the fluorine substituents are especially strain-rich:  geminal, proximate, and W-related. The 1H, 13C, and 19F NMR spectra exhibit a striking variety of chemical shifts and long-range coupling constants.
