GABA(A) (gamma-aminobutyric acid type A) receptors mediate most of the 'fast' synaptic inhibition in the mammalian brain and are targeted by many clinically important drugs. Certain naturally occurring pregnane steroids can potently and specifically enhance GABA(A) receptor function in a nongenomic (direct) manner, and consequently have anxiolytic, analgesic, anticonvulsant, sedative, hypnotic and anaesthetic properties. These steroids not only act as remote endocrine messengers, but also can be synthesized in the brain, where they modify neuronal activity locally by modulating GABA(A) receptor function. Such 'neurosteroids' can influence mood and behaviour in various physiological and pathophysiological situations, and might contribute to the behavioural effects of psychoactive drugs.

Neurosteroids: endogenous regulators of the GABAA receptor

The pathway leading from soluble and monomeric to hyperphosphorylated, insoluble and filamentous tau protein is at the centre of many human neurodegenerative diseases, collectively referred to as tauopathies. Dominantly inherited mutations in MAPT, the gene that encodes tau, cause forms of frontotemporal dementia and parkinsonism, proving that dysfunction of tau is sufficient to cause neurodegeneration and dementia. However, most cases of tauopathy are not inherited in a dominant manner. The first tau aggregates form in a few nerve cells in discrete brain areas. These become self propagating and spread to distant brain regions in a prion-like manner. The prevention of tau aggregation and propagation is the focus of attempts to develop mechanism-based treatments for tauopathies.

The many faces of tau.

Chronic diseases like those related to the cardiovascular system are among the most common causes of death worldwide, as are chronic obstructive diseases of the lung (COPD: chronic obstructive pulmonary disease). In most cases, the disease progression can be slowed down, yet there is no way to stop it. To this end, strict therapy compliance, along with the continuous monitoring of vital parameters, appears crucial. With the development of connected terminal devices and progress made in information technologies over the past years, continuous patient monitoring is now possible without requiring costly hospitalizations. In the best-case scenario, this translates into improved, or at least not deteriorated, quality of life for the patients, with only a minor impact on their daily life. The COMPASS 1 (COntinuous multi-variate Monitoring of Patients Affected by chronic obstructive pulmonary diSeaSe) project seeks to develop a personal health system (PHS) that is capable of continuously monitoring vital parameters and compressing patient data, while ensuring interoperability and patient safety, in addition to providing prognostic information. This article presents a novel approach to combining interoperability with HL7 standards whilst simultaneously ensuring data compression via compressive sensing.

Leistungssteigerung von Patienten-Monitoring durch Compressive Sensing

Patients suffering from diabetes often develop several comorbidities such as hypertension and dyslipidemia. The presence of the comorbidities leads to more complex patient profiles associated with specific patient treatments. In this paper we present a novel algorithm to help physicians, given a new case, in retrieving similar past patient cases. This novel algorithm is based on the bag-of-words (BoW) model to encode as features, the occurrence of each pre-computed cluster, for each patient, according to the approach of document classification. We then evaluate the algorithm on a real de-identified dataset of 3201 diabetic patients, demonstrating the advantage of our approach.

Case-based retrieval of similar diabetic patients

The dynamic nature of startups is linked to both high risks in investments as well as potentially important financial benefits. A key aspect to manage interactions among investors, experts, and startups, is the establishment of trust guarantees. This paper presents the formalization and implementation of a system enforcing trust in the startup assessment domain. To do so, an existing architecture has been extended, incorporating a multi-agent community and related interactions via private blockchain technology. The developed system enables a trust-based community, immutably storing, tracking, and monitoring the agents’ interactions and reputations.

A Startup Assessment Approach Based on Multi-Agent and Blockchain Technologies

In this article we present our work on the MAGPIE project. In this project we are developing an agent platform for Android that will help with the development of mobile applications for PHS. Moreover, with this work we aim to improve the scalability of current PHS. We plan to use this platform in the future with real patients.
