The first prior: From co-embodiment to co-homeostasis in early life.

Various studies demonstrate a special role of the right compared to the left anterior insula in mediating our self. However, the neural features of the right insula that allow for its special role remain unclear. Presupposing a spatiotemporal model of self—“Basis model of self-specificity” (BMSS)—we here address the following question: what spatial-topographic and temporal-dynamic features render neural activity in the right insula to be more suitable in mediating self-specificity than the left insula? First, applying fMRI, we demonstrate that the right insula (i) exhibits higher degrees of centrality in rest, and (ii) higher context-dependent functional connectivity in a self-specific task among regions of distinct layers of self (intero-, extero-proprioceptive, and mental). Second, using EEG in rest and task, we show that the right insula shows longer autocorrelation window (ACW) in its neural activity than both left insula and other regions of the different layers of self. Together, we demonstrate special topographic, i.e., high functional connectivity, and dynamic, i.e., long ACW, neural features of the right insula compared to both left insula and other regions of the distinct layers of self. This suits neural activity in the right insula ideally for high functional integration and temporal continuity as key features of the self including its intero-, extero-proprioceptive, and mental layers.

The Self and Its Right Insula—Differential Topography and Dynamic of Right vs. Left Insula

The COVID-19 crisis is affecting our sense of self and touches upon our existential fears. This extends to the self-other relationship, as there is both being infected and infecting the other. What does this pandemic crisis tell us about our self and relatedness, its cultural differences, and how these are rooted in the brain's relation to the world? First, we discuss the psychological and neuronal features of self and self-other relation and how they are rooted in a deeper layer of the brain's neural activity complementing its cognitive surface layer. Second, we demonstrate cultural differences of Eastern and Western concepts of the self (i.e., independency and interdependency) and how these reflect the manifestation of the brain's neuro-social and neuro-ecological alignment. Finally, we highlight the intersubjective and cultural nature of the self and its surface in the COVID-19 crisis. Discussing various lines of empirical data showing the brain's intimate alignment to both social and ecological environmental contexts, our results support the assumption of the brain's deep layer features by laying bare a continuum of different degrees of neuro-social and neuro-ecological alignment. This entails a two-stage model of self with neuro-social-ecological and psychological levels that extends the previously suggested basis model of self-specificity. We conclude that the current pandemic shows the importance of the deeper intersubjective and cultural layers of both the self and brain; their neglect can be life-threatening for the self and others and, paradoxically, might reduce, rather than enlarge, the self's sense of freedom and independence.

What COVID-19 tells us about the self: The deep intersubjective and cultural layers of our brain

LSD is the prototypical psychedelic. Despite a clear central role of the 5HT2a receptor in its mechanism of action, the contributions of additional receptors for which it shows affinity and agonist activity remain unclear.We employed receptor-enriched analysis of functional connectivity by targets (REACT) to explore differences in functional connectivity (FC) associated with the distributions of the primary targets of LSD-the 5HT1a, 5HT1b, 5HT2a, D1 and D2 receptors.We performed secondary analyses of an openly available dataset (N = 15) to estimate the LSD-induced alterations in receptor-enriched FC maps associated with these systems. Principal component analysis (PCA) was employed as a dimension reduction strategy for subjective experiences associated with LSD captured by the Altered States of Consciousness (ASC) questionnaire. Correlations between these principal components as well as VAS ratings of subjective effects with receptor-enriched FC were explored.Compared to placebo, LSD produced differences in FC when the analysis was enriched with each of the primary serotonergic and dopaminergic receptors. Altered receptor-enriched FC showed relationships with the subjective effects of LSD on conscious experience, with serotonergic and dopaminergic systems being predominantly associated with perceptual effects and perceived selfhood as well as cognition respectively. These relationships were dissociable, with different receptors showing the same relationships within, but not between, the serotonergic and dopaminergic systems.These exploratory findings provide new insights into the pharmacology of LSD and highlight the need for additional investigation of non-5HT2a-mediated mechanisms.

Diminished self-monitoring in hallucinations – Aberrant anterior insula connectivity differentiates auditory hallucinations in schizophrenia from subjective tinnitus

Chronic tinnitus is a prevalent condition that could cause severe negative impact on an individual's life. However, there has not been an established treatment due to a limited understanding of the pathophysiology of this multifarious disorder. In this study, we tested the efficacy of an integrative treatment, combining music therapy with cognitive-behavioral therapy (CBT). We collected three groups of patients receiving three different treatments: Music-CBT, music therapy and CBT. We used both subjective (i.e., questionnaires) and objective (i.e., resting-state EEG data) measurements to assess the behavioral and neural changes brought upon by the treatments. Analyses of the subjective measurements found a significant improvement of scale scores in Music-CBT and CBT, but not in the Music group. Analysis of the EEG data further showed increased powers in alpha and theta band after the Music-CBT treatment, and increased gamma power after CBT, whereas no significant difference was found for the music therapy. Further source localization analysis of alpha and theta changes in the Music-CBT group found that primary sources of the changes were located at auditory processing regions such as superior temporal gyrus, and higher emotional and cognitive processing regions such as ventromedial prefrontal cortex (vMPFC), lateral prefrontal cortex and parahippocampus. These results indicated that Music-CBT was effective in improving tinnitus symptoms on both a behavioral and neural level, which is more robust than the music therapy or CBT alone.
