Abstract: The application of the conventional double-fixation method (glutaraldehyde and osmium tetroxide) to whole yeast cells is difficult because the thick cell wall of the yeast prevents the penetration of osmium tetroxide. However, this problem was solved by using the freeze-substitution fixation method. Therefore, it was possible to examine the intracellular structures of the yeast cells without digestion of the cell wall. In the present method, specimens for transmission electron microscopy and for scanning electron microscopy were prepared simultaneously. By scanning electron microscopic observation, three-dimensional information about internal structures was obtained. In the cytological analysis of the yeasts, intracellular structures were well preserved by using the freeze-substitution fixation method. On the outer leaflet of the nuclear envelope, many ribosomes were attached. The rough endoplasmic reticulum and Golgi apparatus were clearly seen in the yeast cytoplasm. The Golgi stack appeared to consist of smooth membranes, and small vesicles were present beside it. The details of other structures such as the nuclear division apparatus, actinlike filaments, and viruslike particles were also revealed. The present technique can be applied to most species of yeast cells. With this new information, the previous model of a yeast cell was modified.

Abstract: Protoplasts of Candida albicans were prepared by digestion with Zymolyase and the effect of aculeacin A, a wall-active antibiotic, on the synthesis of microfibrillar structures by the protoplasts incubated for 5 hr in an osmotically stabilized medium was studied using several electron microscopical techniques. Chemical analyses of the protoplasts before and after reversion with or without the antibiotic were also performed. Aculeacin A not only inhibited synthesis of microfibrils mainly composed of alkali-insoluble beta-glucan, but also their assembly to form thicker bundles. The antibiotic appeared to have no effect on other wall components constituting the surface structure of reverting protoplasts. These data confirmed our previous postulation that aculeacin A is a specific and potent inhibitor of beta-glucan synthesis as well as biogenesis of cell walls including beta-glucans in susceptible yeasts.

Abstract: Recently, the reliability of field-emission electron guns has increased. In addition, the cost of computer systems for on-line processing has dropped. Hence, we should now consider the use of scanning transmission electron microscopy (STEM) for routine work, especially, in the field of biology where one may expect to utilize digital image processing techniques. An STEM has been constructed, without disturbing the original functions, by converting a commercial scanning electron microscope equipped with a fieldemission gun. The STEM is generally operated at accelerating voltage 30 kV, focal length 7.5 mm, and beam current 1−2 × 10−10 A. Several improvements have been incorporated for removing the effects of vibration, contamination, and stray magnetic fields. Also, an adjustable detector aperture was utilized. The modified instrument was connected to an on-line digital image processing system for utilizing the information obtained from STEM images. The advantages of the modified system were studied from various viewpoints.
