Journal ArticleDOI
Premature polyadenylation-mediated loss of stathmin-2 is a hallmark of TDP-43-dependent neurodegeneration.
Ze’ev Melamed,Jone López-Erauskin,Michael Baughn,Ouyang Zhang,Kevin Drenner,Ying Sun,Fernande Freyermuth,Moira A. McMahon,Melinda S. Beccari,Jon Artates,Takuya Ohkubo,María José Cubillas Rodríguez,Nianwei Lin,Dongmei Wu,C. Frank Bennett,Frank Rigo,Sandrine Da Cruz,John Ravits,Clotilde Lagier-Tourenne,Don W. Cleveland +19 more
TL;DR: Reduction in TDP-43 is shown to inhibit axonal regeneration of iPSC-derived motor neurons, and rescue of stathmin-2 expression restores axonal regenerative capacity, a hallmark of ALS–FTD that functionally links reduced nuclear TSP function to enhanced neuronal vulnerability.
Book ChapterDOI
Gene Disruption Using Chemically Modified CRISPR-Cpf1 RNA.
Moira A. McMahon,Meghdad Rahdar +1 more
TL;DR: A detailed protocol for use of CRISPR-Cpf1 and chemically modified guide RNAs in cell lines is provided, outlining the steps from designing guideRNAs to a target gene of interest, delivery and expression in cells, and analysis of gene editing events.
