The relationship between antioxidant activity and antimutagenicity of various tea extracts (green tea, pouchong tea, oolong tea, and black tea) was investigated. All tea extracts exhibited markedly antioxidant activity and reducing power, especially oolong tea, which inhibited 73.6% peroxidation of linoleic acid. Tea extracts exhibited a 65-75% scavenging effect on superoxide at a dose of 1 mg and 30 - 60% scavenging effect on hydrogen peroxide at a dose of 400 microgram. They scavenged 100% hydroxyl radical at a dosage of 4 mg except the black tea. Tea extracts also showed 50 - 70% scavenging effect on alpha, alpha-diphenyl-beta-picrylhydrazyl radical. The antioxidant activity and the scavenging effects on active oxygen decreased in the order semifermented tea > nonfermented tea > fermented tea. Tea extracts showed strong antimutagenic action against five indirect mutagens, i.e., AFB1, Trp-P-1, Glu-P-1, B[a]P, and IQ, especially oolong and pouchong teas. The antioxidant effect of tea extracts was well correlated to their antimutagenicity in some cases but varied with the mutagen and antioxidative properties.

Antioxidant activity of various tea extracts in relation to their antimutagenicity

Epidemiologic studies have demonstrated an inverse association between consumption of fruits and vegetables and morbidity and mortality from degenerative diseases. The antioxidant content of fruits and vegetables may contribute to the protection they offer from disease. Because plant foods contain many different classes and types of antioxidants, knowledge of their total antioxidant capacity (TAC), which is the cumulative capacity of food components to scavenge free radicals, would be useful for epidemiologic purposes. To accomplish this, a variety of foods commonly consumed in Italy, including 34 vegetables, 30 fruits, 34 beverages and 6 vegetable oils, were analyzed using three different assays, i.e., Trolox equivalent antioxidant capacity (TEAC), total radical-trapping antioxidant parameter (TRAP) and ferric reducing-antioxidant power (FRAP). These assays, based on different chemical mechanisms, were selected to take into account the wide variety and range of action of antioxidant compounds present in actual foods. Among vegetables, spinach had the highest antioxidant capacity in the TEAC and FRAP assays followed by peppers, whereas asparagus had the greatest antioxidant capacity in the TRAP assay. Among fruits, the highest antioxidant activities were found in berries (i.e., blackberry, redcurrant and raspberry) regardless of the assay used. Among beverages, coffee had the greatest TAC, regardless of the method of preparation or analysis, followed by citrus juices, which exhibited the highest value among soft beverages. Finally, of the oils, soybean oil had the highest antioxidant capacity, followed by extra virgin olive oil, whereas peanut oil was less effective. Such data, coupled with an appropriate questionnaire to estimate antioxidant intake, will allow the investigation of the relation between dietary antioxidants and oxidative stress-induced diseases.

Total Antioxidant Capacity of Plant Foods, Beverages and Oils Consumed in Italy Assessed by Three Different In Vitro Assays

Free-radical scavenging capacity and antioxidant activity of selected plant species from the Canadian prairies

The antioxidant activity of water extracts of Harng Jyur (Chrysanthemum morifolium Ramat) from four varieties, including Huang Harng Jyur (HHJ), Bai Harng Jyur (BHJ), Gan Harng Jyur (GHJ), and Kung Harng Jyur (KHJ), was investigated. The water extracts of the four Harng Jyur varieties showed strong antioxidant activity in linoleic acid and liposome model systems. Each extract at 20 mg exhibited 65.3–71.8% inhibition on malondialdehyde formation of rat liver homogenate in vitro, greater than 20 mg of tocopherol. A correlation established between the total phenolic compounds and antioxidant activity, yielded a coefficient of r2=0.934, 0.997, 0.758 and 0.983 for HHJ, BHJ, GHJ and KHJ respectively. Each extract of the four Harng Jyur varieties had effective activities as radical scavengers, metal chelators, and active oxygen scavengers. These properties may be the major reasons for the inhibition of lipid peroxidation. Of the four varieties, BHJ was the most significant antioxidative material. No mutagenicity in the four varieties was found in Salmonella typhimurium TA 98, either with or without S9 mix.

Antioxidant activity of water extract of Harng Jyur (Chrysanthemum morifolium Ramat).

The antioxidant activity and free-radical and active oxygen-scavenging activity of burdock extracts were investigated. Of the solvents used for extraction, water yielded the greatest amount of extract that exhibited the strongest antioxidant activity. Water extracts of burdock (WEB) and hot water extracts of burdock (HWEB) exhibited comparable and marked activity on inhibition of linoleic acid peroxidation, indicating that heat treatment did not alter the antioxidant activity of WEB. WEB and HWEB produced significantly lower (P<0.05) malondialdehyde (MDA) in both linoleic acid and liposome model systems than did the control. Moreover, mixtures of tocopherol (Toc), WEB, and HWEB exhibited a remarkable synergistic antioxidant effect in a liposome system; WEB and HWEB thus potentiated the action of Toc. Furthermore, WEB and HWEB displayed a marked inhibitory effect on lipid peroxidation of rat liver homogenate in vitro. WEB and HWEB exhibited an 80% scavenging effect on α,α-diphenyl-β-picrylhydrazyl radical and marked reducing power, indicating that WEB and HWEB act as primary antioxidants. Both extracts at a dose of 1.0 mg exhibited a 60.4–65.0% scavenging effect on superoxide and an 80.5% scavenging effect on hydrogen peroxide. They also showed a marked scavenging effect on the hydroxyl radical. These results revealed that WEB and HWEB are also active as oxygen scavengers and as secondary antioxidants. Based on these results, termination of free-radical reactions and quenching of reactive oxygen species in burdock extracts are suggested to be, in part, responsible for the antioxidant activity of burdock extracts.
