Antibacterial approaches in tissue engineering using metal ions and nanoparticles: From mechanisms to applications

The inappropriate use of antimicrobials has resulted in the selection of resistant strains. Thus, a great number of studies have focused on the investigation of new antimicrobial agents. The use of zinc oxide nanoparticles (ZnO NPs) to optimise the fight against microbial resistance has been receiving increased attention due to the non-specific activity of inorganic antimicrobial agents. The small particle size and the high surface area of ZnO NPs can enhance antimicrobial activity, causing an improvement in surface reactivity. In addition, surface modifiers covering ZnO NPs can play a role in mediating antimicrobial activity since the surface properties of nanomaterials alter their interactions with cells; this may interfere with the antimicrobial effect of ZnO NPs. The possibility of using surface modifiers with groups toxic to microorganisms can improve the antimicrobial activity of ZnO NPs. Understanding the exact toxicity mechanisms is crucial to elucidating the antimicrobial activity of ZnO NPs in bacteria and fungi. Therefore, this review aims to describe the mechanisms of ZnO NPs toxicity against fungi and bacteria and how the different structural and physical-chemical characteristics of ZnO NPs can interfere in their antimicrobial activity.

Relationship Between Structure And Antimicrobial Activity Of Zinc Oxide Nanoparticles: An Overview.

One of the promising ways to overcome microbial antibiotic resistance is the use of nanoparticles of metal oxides. As you know, a wide range of metals has antimicrobial action: Ag, Al, Cu, Fe, Mo, Mn, etc. The mechanisms of the antimicrobial action of metals are impaired functioning of microbial enzymes, generation of reactive oxygen species, damage to the cell membrane, and genotoxic action. ZnO nanoparticles have bacteriostatic and bactericidal action against gram-positive and gram-negative bacteria, as well as fungicidal activity. The mechanism of action is based on the release of Zn2+, generation of reactive oxygen species, damage to the cell membrane, binding to microbial proteins and nucleic acids. NPs based on ZnO have a number of advantages: high antibacterial efficiency at low concentrations (0.16-5.00 mM/l), action on a wide range of microbial strains, and relatively low production costs. Methods have now been found to improve the antimicrobial properties of nanoparticles. The use of several methods at the same time in some cases gives a synergistic effect. All of the above allows us to predict the use of ZnO nanoparticles as a new generation of antimicrobial agents in the near future.

A Mini Review of Antibacterial Properties of ZnO Nanoparticles

In the present study, zinc oxide nanoparticles (ZnO NPs) were synthesized using leaf extract of Catharanthus roseus (C. roseus) under different physical parameters. Biosynthesis of ZnO NPs was confirmed by UV-Visible spectrophotometer and further, characterized by X-Ray Diffraction (XRD), Fourier Transform Infrared spectroscopy (FTIR), Scanning Electron Microscopy (SEM), Transmission Electron Microscopy (TEM), Energy-Dispersive X-ray spectroscopy (EDX), Atomic Force Microscopy (AFM), Photoluminescence study and Dynamic Light Scattering (DLS). We have also confirmed that several physical parameters such as pH, temperature, concentration of metal ions and reaction time were able to regulate shape and size of synthesized ZnO NPs. XRD and TEM analysis provided the information about the average size and hexagonal morphology of ZnO NPs. FTIR spectra analysis suggested that phenolic compounds played crucial role in the biosynthesis of ZnO NPs. The significant antibacterial activity of ZnO NPs was observed against Staphylococcus aureus MTCC 9760 (S. aureus), Streptococcus pyogenes MTCC 1926 (S. pyogenes), Bacillus cereus MTCC 430 (B. cereus), Pseudomonas aeruginosa MTCC 424 (P. aeruginosa), Proteus mirabilis MTCC 3310 (P. mirabilis) and Escherichia coli MTCC 40 (E. coli). The synthesized ZnO NPs have shown antibacterial efficacy against both Gram-positive and Gram-negative pathogens. Synergistic effects of ZnO NPs and streptomycin showed increased efficacy as indicated by the increased zone of clearance in comparison to their individual effects (either ZnO NPs or streptomycin). Overall, the results elucidated a rapid, cost-effective, environmentally friendly and convenient method for ZnO NPs synthesis, which could be used as a potential antimicrobial agent against drug resistant microbes.

Effective Antimicrobial Activity of Green ZnO Nano Particles of Catharanthus roseus.

Therapeutic and diagnostic potential of nanomaterials for enhanced biomedical applications.

Acinetobacter baumannii is a multi-drug resistant opportunistic pathogen, which causes respiratory and urinary tract infections. Its prevalence increases gradually in the clinical setup. Carbapenems (beta-lactam) are most effective antibiotics till now against A. baumannii, but the development of resistance against it may lead to high mortality. Therefore, it is of utmost importance to develop an alternative drug against A. baumannii. In the present study, we have synthesized ZnO nanoparticle (ZnO-NP) and characterized by X-ray diffraction, Fourier transform infrared (FTIR) spectroscopy and UV-Visible spectroscopy. Prepared ZnO-NPs have the size of 30 nm and have different characteristics of ZnO-NPs. Growth kinetics and disk diffusion assay showed that ZnO-NP demonstrated good antibacterial activity against carbapenem resistant A. baumannii. We have also investigated the mechanism of action of ZnO-NPs on the carbapenem resistant strain of A. baumannii. The proposed mechanism of action of ZnO involves the production of reactive oxygen species, which elevates membrane lipid peroxidation that causes membrane leakage of reducing sugars, DNA, proteins, and reduces cell viability. These results demonstrate that ZnO-NP could be developed as alternative therapeutics against A. baumannii.
