Clinical Responses and Pharmacokinetics of MCARH171, a Human-Derived Bcma Targeted CAR T Cell Therapy in Relapsed/Refractory Multiple Myeloma: Final Results of a Phase I Clinical Trial
Sham Mailankody,Arnab Ghosh,Mette Staehr,Terence J. Purdon,Mikhail Roshal,Elizabeth Halton,Claudia Diamonte,John Pineda,Pavan Anant,Yvette Bernal,Analisa Wills,Neha Korde,Nikoletta Lendvai,Alexander M. Park,Kevin J. Curran,Craig S. Sauter,Maria Lia Palomba,Brigitte Senechal,Xiuyan Wang,Ahmet Dogan,Sergio Giralt,Isabelle Riviere,Ola Landgren,Renier J. Brentjens,Eric L. Smith +36 more
TL;DR: The peak expansion and persistence of MCARH171 as well as durable clinical responses were dose dependent and Laboratory values correlating with CRS reaching grade 3 or requiring Tocilizumab, as IL6 elevation artificially increases after use were seen.

Brentjens +27 more
TL;DR: A retroviral construct for clinical use including a truncated epithelial growth factor receptor (EGFRt) elimination gene: EGFRt/hBCMA-41BBz is generated and clinical investigation of this construct is underway in a dose escalation, single institution trial.

Brentjens,Eric G. Pamer,Marcel R.M. van den Brink,Marcel R.M. van den Brink +26 more
TL;DR: It is hypothesized that the differences in outcomes of patients who receive CAR T cells are related to the composition of their intestinal microbiota at baseline, and a single-center analysis of pre-CAR T cell infusion microbiota composition demonstrates that a subset of patients had diverse microbial composition prior to receiving CAR T cell therapy.
