Journal ArticleDOI
Effect of Acetylator Phenotype on the Rate at Which Procainamide Induces Antinuclear Antibodies and the Lupus Syndrome
TL;DR: It is concluded that acetylator phenotype influences the rate at which procainamide induces antinuclear antibodies and probably the lupus syndrome.
Journal ArticleDOI
Polymorphism of propafenone metabolism and disposition in man: clinical and pharmacokinetic consequences.
Lyle A. Siddoway,Katherine A. Thompson,C. B. McAllister,Thomas J. Wang,Grant R. Wilkinson,Dan M. Roden,Raymond L. Woosley +6 more
TL;DR: It is concluded that propafenone is metabolized via the same cytochrome P-450 responsible for debrisoquine's 4-hydroxylation, and that its pharmacokinetics and concentration-response relationships and the incidence of central nervous system side effects are different in patients of different debrisoquines metabolic phenotype.
Journal ArticleDOI
Influence of opioid agonists on cardiac human ether-a-go-go-related gene K(+) currents.
Alexander N. Katchman,Kelly A. McGroary,Michael J. Kilborn,Craig A. Kornick,Paolo L Manfredi,Raymond L. Woosley,Steven N. Ebert +6 more
TL;DR: It is demonstrated that LAAM and methadone can block IHERG in transfected cells at clinically relevant concentrations, thereby providing a plausible mechanism for the adverse cardiac effects observed in some patients receiving LAAM or Methadone.
Journal ArticleDOI
Predicting the Unpredictable: Drug-Induced QT Prolongation and Torsades de Pointes.
TL;DR: Prevention of TdP in the future may require inclusion of both genetic and clinical predictors into CDSS, which can scan the patient's electronic health records for clinical risk factors predictive of diLQTS and warn when a drug that can cause TDP is prescribed.
Journal ArticleDOI
QT prolongation and torsades de pointes among methadone users: reports to the FDA spontaneous reporting system.
TL;DR: Recent case series have associated the synthetic opioid, methadone, with QT prolongation and torsades de pointes (TdP) ventricular arrhythmia.
