Estimating The Efficiency Of Phagocytic Neutrophil Cells And Studying Its Risk Factors Among Diabetic Foot Ulcers

DNA is the central molecule in cells. The correct f unction of cells depends on the structure of DNA, a nd any mutation led to drives a wave of cellular multiplic ation associated with gradual increases in cancer o ccurring. The effect of paracetamol on DNA was studied using prol iferative lymphocyte in vitro. DNA fragmentation wa s measured by quantitative method using (DPA). This s tudy showed, that the Paracetamol drug at all conce ntrations (500µg/ml; 250µg/ml; and 125µg/ml) showed potent in duction of DNA fragmentation in DNA after different exposure periods (24hrs; 48hrs; and 72hrs) respecti vely. Current results showed, that the paracetamol at 500 µg/ml had highly apoptotic activity against normal lympho cytes,the percentage of fragmented DNA was 47.13 %, 69.32%, and 72.81% at 24hrs, 48hrs, and 72hrs respectively. The results showed significant effect (p< 0.05) of paracetamol on treated cells at 250 µg/ml after 24hrs of exposure time, the percentage o f DNA fragmentation was 51.94%, 60.00% and 57.89%, after 24hrs, 48hrs, and 72hrs of exposure time, respectively. However, the ability of paracetamol at low concentrations (125 µg/ml) were achieved a significant fragmentation (p ≤ 0.05) on lymphocytes DNA; percentages of DNA fragmentation was 44.76%, a nd 61.66%, and 70.17% respectively after 24hrs, 48h rs, and 72hrs of incubation period in. Indeed, the inhi bition rate was increased with increasing time of i ncubation. On the other hand, present results showed low percenta ge of DNA fragmentation (21.14%, 21.78%, and 23.42% ) in untreated cells after different exposure periods (2 4, 48, and 72hrs) respectively.

 Background: Foot ulcers complications in diabetes mellitus (DM) patients are one of the significant medical problems and an economic burden. The aim of this study is to assessment role of phagocytic neutrophil cells and its relationship with the incidence of diabetic foot ulcers (DFU) infection of diabetic patients. A total of 60 venipuncture blood samples collected from diabetes mellitus, diabetic foot ulcer patients and healthy persons as control group. 20 swabs from the DFU patients were collected and processed for culture and susceptibility test after the ulcers classified according to Wagner’s grades system. Phagocytic cells activity test was performed to determine the efficiency of phagocytic neutrophil cells in diabetic patients. Results: Gram positive bacteria were the most prevalent in the DFU patients 57.1% with statistical significant relationship between the type of bacteria and grades of Wagner’s classification followed by Gram negative bacteria in high grades of ulcers. Wagner’s ulcers grade 1 and 2 were the most prevalence in DFU patients 30%. There was a weak negative correlation between the efficiency of phagocytic neutrophil cells activity and grade ulcers classified (r = -0.323). Amikacin and ciprofloxacin were the most effective antibiotics against 90.5% and 81% of the bacterial isolates respectively, whereas ampicillin, cefepime and cefadroxil were less effective antibiotics against the bacterial isolates.Conclusion: When the grade of ulcer increased, the bacterial resistance to antibiotics increased, and this was emphasis the correlation with prevalent of Gram negative bacteria in the high grade of ulcers with high resistance of antibiotics. In contrast, the grade of ulcer increased, the efficiency of phagocytic neutrophil cells decreased.

Estimation of Phagocytic Activity in Diabetic Patients (Type 1 , 2) in Mosul City

Our study includes follow up the effect of excessive dose of paracetamol in some individuals on nonspecific immunological parameters, the study estimates total white blood cell count and calculates the absolute number of neutrophils, lymphocytes and monocytes in blood, and estimation of the effectiveness of phagocytic cells of neutrophils. The study included the collection of (30) blood samples from individuals used paracetamol with excessive dose with age range between (20-40) years of both sexes, blood samples individuals donors do not use paracetamol as control samples were used as well. The results showed no significant differences in number of total white blood cell count and number of lymphocytes for individuals with excessive dose of paracetamol compared with control, on another hand the results showed a significant increase in number of neutrophils and monocytes compared with control, concerning the phagocytic activity, the results showed a significant reduction in phagocytic activity in individuals with excessive dose of paracetamol compared with control.
