ABPP-HT - high-throughput activity-based profiling of deubiquitylating enzyme inhibitors in a cellular context
TL;DR: A new methodology named ABPP-HT (high-throughput-compatible activity-based protein profiling), implementing a semi-automated proteomic sample preparation workflow that increases the throughput capabilities of the classical ABPP workflow approximately ten times while preserving its enzyme profiling characteristics.
Posted ContentDOI
The chaperonin CCT8 controls proteostasis essential for T cell maturation, selection, and function
Bergithe E Oftedal,Stefano Maio,Adam E. Handel,Madeleine P. J. White,Duncan Howie,Simon J. Davis,N Prevot,Ioanna A. Rota,Mary E. Deadman,Benedikt M. Kessler,Roman Fisher,Nikolaus S. Trede,Erdinc Sezgin,Rick M. Maizels,Georg A. Holländer +14 more
TL;DR: It is demonstrated that normal T cell biology is critically dependent on CCT-controlled proteostasis and that its absence is incompatible with protective immunity.
Posted ContentDOI
Age-dependent changes in protein incorporation into collagen-rich tissues of mice by in vivo pulsed SILAC labelling
Yoanna Ariosa-Morejon,Alberto Santos,Roman Fisher,Simon J. Davis,Philip D. Charles,Rajesh V. Thakker,A.K. Wann,Tonia L. Vincent +7 more
TL;DR: This article used in vivo pulsed SILAC labelling to quantify new protein incorporation into cartilage, bone, skin, and plasma of mice across the life course and reported highly dynamic matrisome turnover in bone and cartilage during skeletal maturation which was markedly reduced after skeletal maturity.
Posted ContentDOI
Interplay Between KRAS and LZTR1 Protein Turnover, Controlled by CUL3/LZTR1 E3 Ubiquitin Ligase, is Disrupted by KRAS Mutations
Andreas Damianou,Z. Liang,Benedikt M. Kessler,F. Lassen,George Vere,S. Hester,Philip D. Charles,Adan Pinto-Fernandez,Roman Fisher,A. Santos-Delgado +9 more
TL;DR: In this article, the presence of known interactors of KRAS including a-RAF and LZTR1, which varied in abundance with wild type and KRAS mutants, was demonstrated by quantitative proteomics.

Cardiac Atrial Compartmentalisation Proteomics: A Modified Density Gradient Method to Analyse Endo-lysosomal Proteins
Thamali Ayagama,Samuel J. Bose,Rebecca A. Capel,David A. Priestman,Georgina Berridge,Roman Fisher,Antony Galione,Frances M. Platt,Holger Kramer,Rebecca A.B. Burton +9 more
TL;DR: In this paper, the authors describe a label-free proteomics method suitable for small cardiac tissue biopsies based on density-separated fractionation, which allows study of endo-lysosomal (EL) proteins.
