Chemical reactivity, metabolic oxirane formation and biological reactivity of chlorinated ethylenes in the isolated perfused rat liver preparation

Although various bicyclo[3.3.0]octane derivatives have been known for several decades, the chemistry of this ring system and the higher polyquinanes has developed particularly rapidly in the last decade. These developments have been stimulated by a wide range of interests and have encompassed a broad spectrum of research chemists. One may now safely claim that polyquinanes are readily available molecules. The more complex members of this class, particularly those endowed with all-cis stereo-chemistry, take on a “sphere-like” topology and conformational rigidity. As sphericality increases, the ability of a molecule to exclude solvation from its inner core is enhanced. Can smaller molecules or atoms be occluded therein? And what about completely enclosed structures such as the pentagonal dodecahedrane? What unique properties will characterize compounds of this type? It is the quest for such information that will propel the field of polyquinane chemistry rapidly forward in the years to come. The increasing complexity of natural products having multiply fused cyclopentanoid rings will generate a comparably high level of chemical investigation in this field.

The development of polyquinane chemistry

The metabolic transformation of chlorinated aliphatic hydrocarbons to reactive intermediates is predominantly determined by the number of chlorine substitutions and the type of C-C bonds. Due to their electron-attracting effect, chlorine residues exert a destabilization in alkanes, resulting in metabolic conversions to either alkenes (dechlorination or dehydrochlorination) or free radicals. Chlorinated alkenes are enzymatically oxidized to epoxides which may be hydrolized (enzymatically or nonenzymatically), react with cellular nucleophiles, or rearrange to either chlorinated aldehydes or acyl chlorides. In the series of chlorinated ethylenes (tetra-, tri-, 1,2-cis- and trans-di-, 1,1-di-, and monochloroethylene) the metabolites thus far identified are identical to or biological conjugates of the expected thermal rearrangement products of the epoxides, with one important exception: trichloroethylene. Die Dehydrochlorierung der Pentachlorbutadiene 1 und 3 liefert Perchlorbutenin (2), das je nach der angewandten Temperatur vorzugsweise zum α- (80–100°) bzw, β-C8Cl8 (120 bis 180°) dimerisiert. Fur β-C8Cl8 wird durch Hydrolyse zu 8 und Oxydation zu 9 die Struktur des Perchlorbenzocyclobutens 7 bewiesen. α-C8Cl8 besitzt auf Grund des 13C-NMR-Spektrums die Struktur 11 eines Bicyclo[4.2.0]octatriens-(1.5.7). Chemisch ist jedoch die aus dem NQR-Spektrum abgeleitete Alternativformel 12 nicht sicher auszuschliesen. In Ameisensaure oder unter dem Einflus von Ag⊖-Ionen in Methanol bzw. Chloroform lagert sich α-C8Cl8 in das Cyclooctatetraenderivat 32 (γ-C8Cl8) um. Durch Ozonisierung des α-Isomeren werden die Dihydrophthalsaurederivate 13, 19 und 21 erhalten, die durch Dechlorierung oder Allylumlagerung und 1.7-Fragmentierung glatt in die zugehorigen Phthalsaure- bzw. Benzoesaurederivate ubergehen. Nucleophile Agenzien greifen α-C8Cl8 sehr leicht aromatenbildend an, wobei die entsprechenden Derivate von 7 entstehen. An 7 verlaufen die nucleophilen Substitutionen bedeutend schwerer.



Structure and Reactivity of the Dimers of Perchlorobutenyne



Dehydrochlorination of the pentachlorobutadienes 1 and 3 yields perchlorobutenyne (2). Dimerization of 2 at different temperatures leads to the formation of α- (80–100°) or β-C8Cl8 (120–180°). β-C8Cl8 is identified as perchlorobenzocyclobutene 7 by hydrolysis to 8 and oxidation to 9. On the basis of its 13C-n.m.r. spectrum α-C8Cl8 has the bicyclo[4.2.0]octa-1.5.7-triene structure 11; the alternative formula 12 concluded from the n.q.r. spectrum, cannot be excluded certainly by chemical experiments. In formic acid or under the influence of Ag⊕-ions in methanol or chloroform α-C8Cl8 is transformed into the cyclooctatetraene derivative 32 (γ-C8Cl8). On ozonolysis the α-isomer yields derivatives of dihydrophthalic acid 13, 19 and 21. These compounds are converted into the corresponding derivatives of phthalic and benzoic acids by dechlorination or allylic rearrangement and 1.7-fragmentation. α-C8Cl8 is very easily attacked by nucleophilic agents, thus forming aromatic products derived from 7. Nucleophilic substitution of 7 is not as vigorous as with α-C8Cl8.

Konstitution und Reaktivität der dimeren Perchlorbutenine

Perchlor-COT1 lagert sich bei 180°C in die Dihydropentalene 2 und 4 um. Die Chlorierung von 1 oder 2 fuhrt zu dem Tetrahydropentalen 5a, wahrend die von 4 das Tetrahydropentalen 7a ergibt. Bei weiterer Chlorierung gehen 5a und 7a in das gleiche Hexahydropentalen 8 uber. Die Struktur dieser Verbindungen ergibt sich am ihren 13C-NMR-Spektren und den chemischen Reaktionen von 2, 4 und 5a mit Sauren und nucleophilen Agentien zu den Derivaten 9–14. Besonders charakteristisch ist die Dechlorierung von 2 oder 4 zu einem roten Dimeren des Perchlorpentalens (16).

Cyclooctatetraen‐Dihydropentalen‐Umlagerung von Perchlor‐COT und einige Polyhalogenderivate des Bicyclo [3.3.0] octans

Die Dimerisation der per- und polyhalogenierten Benzocyclobutadiene 2 und 10, die durch Dehalogenierung bzw. Dehydrohalogenierung aus den Vorstufen 1 und 3 bzw. 9 erzeugt werden, verlauft insofern ungewohnlich, als sie nur „lineare” Dimere ergibt. In Gegenwart von 1,3-Diphenylisobenzofuran last sich 10a in dem Diels-Alder Addukt 14 abfangen. Die Dimeren 4 und 12 lagern sich in siedendem Toluol in die entsprechenden Dibenzo[a,e]-cyclooctene 6 und 13 um. Das Perchlorderivat 6a spaltet bei 400°C Chlor ab zum Dibenzopentalen 7.



Dimers of Polyhalogenated Benzocyclobutadienes



The dimerization of per- and polyhalogenated benzocyclobutadienes 2 and 10, prepared by dehalogenation of 1 and 3 or dehydrohalogenation of 9, proceeds unusually to give only „linear” dimers. When 10a is generated in the presence of 1,3-diphenylisobenzofuran the Diels-Alder addukt 14 is obtained. In boiling toluene the dimers 4 and 12 are isomerized to dibenzo[a,e]cyclooctenes 6 and 13. The perchlorinated derivative 6a splitts off chlorine at 400°C to form a dibenzopentalene 7.

Dimere polyhalogenierter Benzocyclobutadiene

Die bicyclische Perchlorverbindung 1 liefert mit primaren aliphatischen Aminen die Diimine 3a–d. Mit Athylenimin lassen sich vier Aziridinreste unter Bildung von 4 einfuhren. Offenkettige sekundare Amine spalten dagegen den Cyclobutenring und liefern o-dichlormethylsubstituierte Benzamide 6. Hierbei wechselt ein Halogenatom seinen Platz am Vierring, wie die Umwandlung der Bromverbindung 16 in das Amid 17 veranschaulicht. Auch durch Ammoniak erfahrt 1 eine Ringoffnung. Dabei wird ein C-Atom als Formamid abgespalten und das Benzonitril 18c gebildet. Durch Variation der Reaktionsbedingungen ist das Diimin 3f fasbar, das ebenso wie 3a–d mit Basen in 18c ubergefuhrt werden kann. Mogliche Mechanismen fur die Ringspaltung von 1 werden diskutiert.



Polyhalogenated Bicyclo[4.2.0]octa-1,5,7-trienes, V. Aromatizing Nucleophilic Substitutions with Aliphatic Amines and Ammonia



The bicyclic perchloro compound 1 reacts with primary aliphatic amines to yield the diimines 3a–d. With ethylene imine four aziridino groups are introduced to form 4, while openchained secondary amines effect cleavage of the four-membered ring to yield o-dichloromethyl-substituted benzamides 6. In this reaction a halogen atom is transferred from one position to the other within the cyclobutene system, as demonstrated by the conversion of the bromo compound 16 to the amide 17. Ammonia, too, causes ring opening of 1, but the reaction is connected with loss of one carbon atom to give the benzonitrile 18c and formamide. Under special conditions the diimine 3fis obtained from 1 and ammonia, which may be transformed as well as 3a–d into 18c by bases. Possible mechanisms for the ring cleavage reactions of 1 are discussed.
