﻿This article presents an overview of haematological parameters and factors affecting their values. Blood act as a pathological reflector of the status of the exposed animals to toxicants and other conditions. The examination of blood provides the opportunity to clinically investigate the presence of metabolites and other constituents in the body of animals and it plays a vital role in the physiological, nutritional and pathological status of an animal. Blood constituents change in relation to the physiological status of an animal. These changes are important in assessing the response of farm animals to various physiological situations. These changes are often caused by several factors; some of which are genetic and others, non-genetic. Age, sex, breed and management systems are among the factors that influence blood-based parameters of farm animals. It is important to establish baseline indices for these parameters on the basis of these factors and determine the effects of these factors on these indices.

Haematological Parameters and Factors Affecting Their Values

Biochemical investigations were carried out to assess the effect of consumption of crude oil contaminated catfish on the hepatocytes and performance of rat. Catfish (Clarias gariepinus) (n = 120) were grouped into 6 of 20 catfish each and held for 30 h in 5 different mixtures of crude oil polluted water (0.1, 0.25, 0.5, 0.75 and 1% v/v). Catfish in the control group were held in borehole water. At the expiration of 30 h, the catfish were harvested and used to formulate diet. Albino rats (n = 60) were grouped into 6 of 10 rats each and fed on the formulated diet for a period of 30 days. Compared with the control, the result showed no significant difference (p>0.05) in the average daily feed intake in all the groups of rats. However, a significant reduction (p<0.05) was observed in the overall body weight, the liver-to-body weight ratio as well as the serum concentrations of albumin and globulin of rat as the amount of crude oil in the diet increased. A significant increase (p<0.05) was also observed in the serum bilirubin concentration of rats fed on diet formulated with crude oil contaminated catfish when compared with the control. Histological analysis also revealed that the architectural arrangement of the liver was altered following the consumption of diet containing catfish exposed to crude oil polluted water. Overall, the data obtained indicate a possible adverse effect on the performance and impaired liver function in the rats fed on diet containing catfish exposed to crude oil polluted water.

Biochemical assessment of the effects of crude oil contaminated catfish (Clarias gariepinus) on the hepatocytes and performance of rat

A population-based-cross-sectional survey was carried out to investigate the potential risk of exposure to premium motor spirit (PMS) fumes in Calabar, Nigeria, among Automobile Mechanics (AM), Petrol Station Attendants (PSA) and the general population. Structured questionnaire was administered on the randomly chosen subjects to elicit information on their exposure to PMS. Duration of exposure was taken as the length of work in their various occupations. Venous blood was taken for methaemoglobin (MetHb) and packed cells volume (PCV). Mean MetHb value was higher in AM (7.3%) and PSA (5.8%) than in the subjects from the general population (2.7%). PCV was lower in PSA (30.8%), than AM (33.3%) and the subjects from the general population (40.8%). MetHb level was directly proportional, and PCV inversely related, to the duration of exposure. The study suggested increased exposure to petrol fumes among AM, PSA, and MetHb as a useful biomarker in determining the level of exposure to benzene in petrol vapour.

The impact of health and environmental hazards, associated with the constituents of gasoline, on occupationally exposed workers has been recorded over the past few decades. However, the scientific literature on their pathogenic potential remains incomplete, which could affect the current understanding of the associated health risks. This review provides current information based on recently improved research techniques to evaluate gasoline toxicity profiles for humans. Our current knowledge provides insight into the intricate mechanism of gasoline-induced adverse effects, including the formation of reactive metabolites via bio-activation and subsequent generation of reactive oxygen species (ROS) and oxidative stress, which are involved in multiple mechanisms that are central to the aetiology of gasoline-induced toxicity. These mechanisms include covalent binding to deoxyribonucleic acid (DNA), leading to oxidative damage, tumor-suppression gene activity, and activation of pro-oncogenes. Furthermore, it results in induction of autoimmunity and local inflammatory responses, disruption of multiple neurotransmitters and immune cell function, derangement of various enzyme activities (e.g., sodiumpotassium adenosine triphosphate (Na+/K+/ATPase) activity, cytochrome P450 (CYP450), nitric oxide synthase (NOS), antioxidant enzyme activities, etc.), conjugation of bile, and non-specific cell membrane interaction, leading to damage of the membrane lipid bilayer and proteins. Available data suggests that exposure to gasoline or gasoline constituents have the potential to cause different types of illnesses. The data highlights the need to maintain safety measures via suitable research, medical surveillance, regulatory control, life style modification, early detection, and intervention to minimize exposure and manage suspected cases. Cadmium and mercury are among the most toxic and dangerous environmental pollutants that may cause fatal implications. Vitamin C is an important chain-breaking antioxidant and enzyme co-factor against heavy metals. The objective of the present study was to evaluate the toxicological effects of cadmium chloride, mercuric chloride, and their co-administration on biochemical parameters of blood serum and metal bioaccumulation in kidneys and also to elucidate the protective effect of vitamin C in rabbits against these metals. In the current research, cadmium chloride (1.5 mg/kg), mercuric chloride(1.2 mg/kg), and vitamin C (150 mg/kg of body weight) were orally administered to eight treatment groups of the rabbits (1, control; 2, vitamin; 3, CdCl2; 4, HgCl2; 5, vitamin + CdCl2; 6, vitamin + HgCl2; 7, CdCl2 + HgCl2, and 8, vitamin + CdCl2 + HgCl2). After the biometric measurements of all experimental rabbits, biochemical parameters viz. creatinine, cystatin C, uric acid, and alkaline phosphatase (ALP) and metal bioaccumulation were determined using commercially available kits and atomic absorption spectrophotometer, respectively. The levels of creatinine (28.3 ± 1.1 μmol/l), cystatin C (1932.5 ± 38.5 ηg/ml), uric acid (4.8 ± 0.1 mg/day), and ALP (51.6 ± 1.1 IU/l) were significantly (P < 0.05) increased due to administration of mercuric chloride but in the presence of vitamin C, the effects of mercuric chloride on creatinine (21.9 ± 1.4 μmol/l), cystatin C (1676.2 ± 42.2 ηg/ml), uric acid (3.9 ± 0.1 mg/day), and ALP (43.3 ± 0.8 IU/l) were less as compared to metal-exposed specimens. Similar results were found in rabbits treated with cadmium chloride and vitamin C and also with co-administration of both metals and vitamin C. Because of the bio-accumulative nature of cadmium chloride and mercuric chloride, these metals were accumulated in kidneys of rabbits, which might lead to deleterious effects. The results of the present study provide an insight into the toxicity of the cadmium chloride, mercuric chloride, and/or their combination on biochemical parameters as well as kidneys of the rabbits and the ameliorating potential of vitamin C against these metals is also evaluated.

Renal toxicity of heavy metals (cadmium and mercury) and their amelioration with ascorbic acid in rabbits

Blood cell profile among experimental rabbits associated with crude oil ingestion were evaluated and was significantly (P<0.05) altered. Erythrocytes decreased linearly (P<0.05) indicating an anemic condition. The decrease also affected dependable factors such as packed cell volume (PVC) and haemoglobin (Hb). Leukocyctes, the main defense cells of the animal decreased linearly (P<0.05) further indicating susceptibility of the animal to stress and infection. Granulolytic leukocytes, which include neutrophils and eosinophils increased linearly (P<0.05). This increase is a physiological response to stress. Lymphocytes, antibody forming leukocytes decreased linearly, indicating a response to stress and susceptibility of the animal to infection. Crude oil fraction present in the diet has serious consequences on hematological parameters in animals.

Biochemical blood parameters in semi-adult rabbits experimentally fed crude oil contaminated diets

The influence of maize-milling waste on the reproductive performance of rabbits was assessed. The treatments comprised of autoclaved and unautoclaved maize-milling waste, groundnut cake/maize meal diets formulated with 0 (I), 25 (II), 50 (III), 75 (IV) and 100% (V) replacement of maize with maize-milling waste. Each of the diets contains 18% crude protein and was unpelleted. There were two parity periods and the duration was 15 weeks. In the first parity period, the conception rates were 33.33, 100, 75, 66.66 and 75% for treatments I, II, III, IV and V, respectively, while the second parity conceptions rates were 50, 75 and 50% for treatments II, III and IV, respectively. No conceptions were observed for treatments I and V. The first breeding trials had abortion rates of 50 and 25% for treatments II and V, respectively. Autoclaved maize-milling waste was used for the second breeding trials. Autoclaving maize milling waste was observed to improve the nutritional value of the diets and neither abortion nor still births were seen for the second parity period. Therefore, simple diet comprising maize-milling waste and groundnut cake can support normal growth and reproduction of rabbits for meat production.
