Wave front shaping, the ability to manipulate light fields both spatially and temporally, in complex media is an emerging field with many applications. This review summarizes how insights from mesoscopic scattering theory have direct relevance for optical wave control experiments and vice versa. The results are expected to have an impact on a number of fields ranging from biomedical imaging to nanophotonics, quantum information, and communication technology.

Light fields in complex media: Mesoscopic scattering meets wave control

Quantitative phase imaging (QPI) with its high-contrast images of optical phase delay (OPD) maps is often used for label-free single-cell analysis. Contrary to other imaging methods, sensitivity improvement has not been intensively explored because conventional QPI is sensitive enough to observe the surface roughness of a substrate that restricts the minimum measurable OPD. However, emerging QPI techniques that utilize, for example, differential image analysis of consecutive temporal frames, such as mid-infrared photothermal QPI, mitigate the minimum OPD limit by decoupling the static OPD contribution and allow measurement of much smaller OPDs. Here, we propose and demonstrate supersensitive QPI with an expanded dynamic range. It is enabled by adaptive dynamic range shift through a combination of wavefront shaping and dark-field QPI techniques. As a proof-of-concept demonstration, we show dynamic range expansion (sensitivity improvement) of QPI by a factor of 6.6 and its utility in improving the sensitivity of mid-infrared photothermal QPI. This technique can also be applied for wide-field scattering imaging of dynamically changing nanoscale objects inside and outside a biological cell without losing global cellular morphological image information.

Adaptive dynamic range shift (ADRIFT) quantitative phase imaging.

Three-dimensional imaging of biological cells is crucial for the investigation of cell biology, providing valuable information to reveal the mechanisms behind pathophysiology of cells and tissues. Recent advances in optical diffraction tomography (ODT) have demonstrated the potential for the study of various cells with its unique advantages of quantitative and label-free imaging capability. To provide insight on this rapidly growing field of research and to discuss its applications in biology and medicine, we present the summary of the ODT principle and highlight recent studies utilizing ODT with the emphasis on the applications to the pathophysiology of cells.

Optical diffraction tomography techniques for the study of cell pathophysiology

Holographic displays generate realistic 3D images that can be viewed without the need for any visual aids. They operate by generating carefully tailored light fields that replicate how humans see an actual environment. However, the realization of high-performance, dynamic 3D holographic displays has been hindered by the capabilities of present wavefront modulator technology. In particular, spatial light modulators have a small diffraction angle range and limited pixel number limiting the viewing angle and image size of a holographic 3D display. Here, we present an alternative method to generate dynamic 3D images by controlling volume speckle fields significantly enhancing image definition. We use this approach to demonstrate a dynamic display of micrometre-sized optical foci in a volume of 8 mm × 8 mm × 20 mm. Control of speckle fields yields high-definition 3D holographic images.

Recent advances in quantitative phase imaging (QPI) and artificial intelligence (AI) have opened up the possibility of an exciting frontier. The fast and label-free nature of QPI enables the rapid generation of large-scale and uniform-quality imaging data in two, three, and four dimensions. Subsequently, the AI-assisted interrogation of QPI data using data-driven machine learning techniques results in a variety of biomedical applications. Also, machine learning enhances QPI itself. Herein, we review the synergy between QPI and machine learning with a particular focus on deep learning. Furthermore, we provide practical guidelines and perspectives for further development.

Quantitative Phase Imaging and Artificial Intelligence: A Review

Recent advances in wavefront shaping techniques for biomedical applications

This article presents a comprehensive review of the laser-based guided ultrasonic waves propagation imaging (G-UPI) system and respective signal/data processing methods related to the nondestructive testing and evaluation of thin-walled structures. The primary goal of this study is to review and recognize various processing methods, explain the working principles of the most influential methods, and highlight outstanding capabilities. In addition, the suitability of the methods for multiple types of damage and defect in various materials and structures are presented. At the same time, success stories of difficult-to-inspect cases are highlighted. Its secondary goal is to compare and discuss the merits and demerits of the laser-scanning part of the system for ultrasound generation and acquisition to provide a guideline for scanning scheme or hardware selection. Finally, the potential challenges and prospects of the G-UPI are discussed. It is expected that this review would serve as an entrance key for newcomers and a reference point for researchers to explore the opportunities for further improvement in the laser ultrasound-based evaluation of critical engineering structures.

Guided ultrasonic waves propagation imaging: a review

We present hyperspectral holographic imaging of mouse brain tissues. Full-field optical amplitude and phase-delay images of a thin slice of mouse brain tissues are measured in the wavelengths ranging from 450 nm to 650 nm. To quantitatively and precisely measure the hyperspectral holographic imaging, we employed a recently develop swept-source diffraction phase microscopy [1, 2], which is composed of a custom-built wavelength-sweeping unit equipped with a supercontinuum source and diffraction phase microscopy, a common-path quantitative phase imaging system. We envision the present approach can provide a method for label-free quantification and diagnosis for biological tissue samples.
