Great attention is currently paid to the synthesis of polynuclear transition metal complexes and the study of their photochemical, photophysical, and electrochemical properties. This interest is stimulated, in particular, by attempts to design and construct multicomponent systems (often called supramolecular species) capable of performing useful lightand/or redox-induced functions.1-16 A great deal of investigations on mononuclear transition metal complexes had previously shown that several families of these compounds are very interesting from the electrochemical, photochemical, and photophysical viewpoints.17-22 The metalligand interaction, in fact, is often (i) weak enough to allow the manifestation of intrinsic properties of metal and ligands (e.g., ligand-centered and metalcentered absorption bands and redox waves) and, at the same time, (ii) strong enough to cause the appearance of new properties, characteristic of the whole compound (e.g., metal-to-ligand or ligand-tometal charge-transfer bands). On passing from mononuclear to polynuclear transition metal complexes, the situation becomes even more interesting because in the latter (supramolecular) compounds one can find, besides properties related to each metal-based component, properties related to the structure and composition of the whole array. A suitable choice of the mononuclear building blocks and bridging ligands and an appropriate design of the (supramolecular) structure can in fact allow the occurrence of very interesting and potentially useful processes such as energy transfer along predetermined pathways, photoinduced charge separation, multielectron exchange at a predetermined potential, etc. The knowledge on the luminescence and redox properties of polynuclear transition metal complexes is rapidly accumulating, but it is disperse in a great number of journals. We have made an attempt to collect the available results, and we present them together with some fundamental introductory concepts and a few comments. One of the main problems, of course, was to delimit the field of this review. Using personal criteria which are related to our own research interests, we decided to consider only polynuclear transition metal complexes that can be defined as supramolecular species (section 2.2) and that are reported to exhibit luminescence. For such compounds only, the electrochemical properties have also been reviewed. Furthermore, we decided to include only classical (Werner-type) polynuclear transition metal compounds where the number of metal-based units is exactly known and the connection between the metal centers is provided only by bridging ligands. The Stability of Cyclodextrin Complexes in Solution.

Cyclodextrins are cyclic oligomers of glucose that can form water-soluble inclusion complexes with small molecules and portions of large compounds. These biocompatible, cyclic oligosaccharides do not elicit immune responses and have low toxicities in animals and humans. Cyclodextrins are used in pharmaceutical applications for numerous purposes, including improving the bioavailability of drugs. Current cyclodextrin-based therapeutics are described and possible future applications discussed. Cyclodextrin-containing polymers are reviewed and their use in drug delivery presented. Of specific interest is the use of cyclodextrin-containing polymers to provide unique capabilities for the delivery of nucleic acids.

Cyclodextrin-based pharmaceutics: past, present and future

Cyclodextrins are extremely attractive components of artificial enzymes and other biomimetic materials. They are readily available, they bind hydrophobic substrates into their cavities in water solution, and they have two rims of hydroxyl groups (Figure 1) that can either react with substrates themselves or be used to attach other catalytic and functional groups. Of course, they have disadvantages. For one, unless they are extensively modified their complexes with substrates can be rather flexible and, perhaps, with unpredictable preferred geometry. They are also unstable to strong acid. Thus for some purposes such synthetic cavity species as calixerenes1 or synthetic macrocycles2-4 may have advantages. However, one of the chief advantages of cyclodextrins is highly attractivesthey are readily available, so it is possible to avoid the synthesis of a binding group and go directly to studies of what can be achieved with their use. Afterward, the lessons learned may be applied to other systems with advantage. This review will cover all the literature on reactions in which cyclodextrins bind substrates and then either catalyze their reactions or mimic a step in an enzymatic catalytic sequence. However, it will not describe work in which cyclodextrins simply change the course of a reaction without playing an obvious catalytic role involving substrate binding. For example, there are systems in which the main function of the cyclodextrin seems to be to complex a metal ion and keep it in solution.5-11 There are other studies in which binding into a cyclodextrin simply alters the selectivity of attack by an external reagent in some way12-24 or causes solubilization to facilitate phase transfer catalysis.12,25,26 Presumably such other areas are described elsewhere in this volume. While much work on artificial enzymes using cyclodextrins has been done in the author’s laboratory, and will be described, every effort is made to describe all the relevant work in the field. Several reviews of this subject already exist and should be consulted for further information.2,27-70 The readily Ronald Breslow, born in 1931 in Rahway, NJ, completed his B.A. in chemistry in1952, his M.A. in medical science in 1953, and his Ph.D. in chemistry in 1955 with R. B. Woodward, all at Harvard University. After a postdoctoral year with Alexander Todd in Cambridge, he came to Columbia University where he is now University Professor and Professor of Chemistry. His work on enzyme models, on novel conjugated aromatic and antiaromatic molecules, on electrochemical and hydrophobic methods in mechanistic chemistry, and on anticancer cytodifferentiating agents has been recognized by a number of awards, including the U.S. National Medal of Science. In 1996, he served as President of the American Chemical Society.

Dimers containing /?-cyclodextrin (/?-CD) moieties at both ends of dicarboxylic acids were prepared by the reaction of bis(m-nitrophenyl) dicarboxylate with a large excess of ft-CD, and were purified by dialysis and gel chromatography. The interaction of the dimers with potassium 2-p-toluidinylnaphthalene-6-sulfonate (TNS) was studied by the fluorescence technique and compared with that of /?-CD and poly(acryloyl-/?-CD). The dimers were found to bring about a much greater enhancement and blue shift of the TNS fluorescence than /?-CD but less than the polymer. The succinate dimer enhanced the TNS fluorescence twofold more than the glutarate dimer. Dissociation constants and stoichiometries were determined by fluorometric titration and continuous variation methods. The dimer complexes showed exclusively 2 : 1 stoichiometry (CD : TNS), even in the concentration range in which /?-CD showed 1 : 1 stoichiometry. The 2 : 1 complexes of the dimers were much more stable than the 2 : 1 complex of fi-CD and were comparable to that of the polymer. Thus, two CD units linked by the chain were found to cooperate in binding one TNS molecule.

Cooperative Binding by Cyclodextrin Dimers

A novel series of polymethylene-linked heterobinuclear complexes of polypyridine ruthenium(II)/osmium(II) complex Ru(II)(bpy)2Mebpy–(CH2)n–MebpyOs(II)(bpy)2(bpy=2,2′-bipyridine and n=2, 3, 5, and 7), 1, was prepared. The photophysical behavior was examined in various solvents. The emission spectra of 1 (excitation wavelength: 455nm) showed a nearly complete quenching of RuII→π*(bpy) metal-to-ligand charge transfer (MLCT) emission and the enhancement of OsII→π*(bpy) MLCT emission. The luminescence lifetime measurements by a time-correlated single photon-counting method provided evidence that intramolecular energy transfer is a significant pathway for the observed emission quenching. The rate constants of the intramolecular energy transfer in ethanol are 5.3×108, 3.3×108, 1.3×108, and 1.0×108s−1 for 1(n=2, 3, 5, and 7), respectively. Intramolecular Energy Transfer in Covalently Linked Polypyridine Ruthenium(II)/Osmium(II) Binuclear Complexes. Ru(II)(bpy)2Mebpy– (CH2)n–MebpyOs(II)(bpy)2 (n=2, 3, 5, and 7)

A condensation polymer (β-CD-E) of β-CD with epichlorohydrin was compared with β-CD and poly(β-CD acrylate) in regard to inclusion behavior in an aqueous solution. The stability of the inclusion complexes of β-CD-E with substrates of a single guest part is smaller than that of β-CD because of steric hindrance by substituents on β-CD. On the other hand, the stability of β-CD-E complexes for substrates with two guest parts is much larger than that of β-CD and is somewhat smaller than that of poly(β-CD acrylate). This is due to the cooperation in binding of the adjacent two CD units on a polymer chain.
