Digital holography is an emerging field of new paradigm in general imaging applications. We present a review of a subset of the research and development activities in digital holography, with emphasis on microscopy techniques and applications. First, the basic results from the general theory of holography, based on the scalar diffraction theory, are summarized, and a general description of the digital holographic microscopy process is given, including quantitative phase microscopy. Several numerical diffraction methods are described and compared, and a number of representative configurations used in digital holography are described, including off-axis Fresnel, Fourier, image plane, in-line, Gabor, and phase-shifting digital holographies. Then we survey numerical techniques that give rise to unique capabilities of digital holography, including suppression of dc and twin image terms, pixel resolution control, optical phase unwrapping, aberration compensation, and others. A survey is also given of representative application areas, including biomedical microscopy, particle field holography, micrometrology, and holographic tomography, as well as some of the special techniques, such as holography of total internal reflection, optical scanning holography, digital interference holography, and heterodyne holography. The review is intended for students and new researchers interested in developing new techniques and exploring new applications of digital holography.

Principles and techniques of digital holographic microscopy

Femtosecond-laser micromachining (also known as inscription or writing) has been developed as one of the most efficient techniques for direct three-dimensional microfabrication of transparent optical materials. In integrated photonics, by using direct writing of femtosecond/ultrafast laser pulses, optical waveguides can be produced in a wide variety of optical materials. With diverse parameters, the formed waveguides may possess different configurations. This paper focuses on crystalline dielectric materials, and is a review of the state-of-the-art in the fabrication, characterization and applications of femtosecond-laser micromachined waveguiding structures in optical crystals and ceramics. A brief outlook is presented by focusing on a few potential spotlights.

Optical waveguides in crystalline dielectric materials produced by femtosecond-laser micromachining

The induced nonlinear electric dipole and higher moments in an atomic system, irradiated simultaneously by two or three light waves, are calculated by quantum-mechanical perturbation theory. Terms quadratic and cubic in the field amplitudes are included. An important permutation symmetry relation for the nonlinear polarizability is derived and its frequency dependence is discussed. The nonlinear microscopic properties are related to an effective macroscopic nonlinear polarization, which may be incorporated into Maxwell's equations for an infinite, homogeneous, anisotropic, nonlinear, dielectric medium. Energy and power relationships are derived for the nonlinear dielectric which correspond to the Manley-Rowe relations in the theory of parametric amplifiers. Explicit solutions are obtained for the coupled amplitude equations, which describe the interaction between a plane light wave and its second harmonic or the interaction between three plane electromagnetic waves, which satisfy the energy relationship ${\ensuremath{\omega}}_{3}={\ensuremath{\omega}}_{1}+{\ensuremath{\omega}}_{2}$, and the approximate momentum relationship ${\mathrm{k}}_{3}={\mathrm{k}}_{1}+{\mathrm{k}}_{2}+\ensuremath{\Delta}\mathrm{k}$. Third-harmonic generation and interaction between more waves is mentioned. Applications of the theory to the dc and microwave Kerr effect, light modulation, harmonic generation, and parametric conversion are discussed.

Interactions between Light Waves in a Nonlinear Dielectric

We present SLIM, a new optical method measuring optical pathlength changes of 0.3 nm spatially and 0.03nm temporally. SLIM combines two classic ideas in light imaging: Zernike’s phase contrast microscopyand Gabor’s holography.

Spatial light interference microscopy (SLIM)

This review gives an overview of techniques used for high-resolution jet printing that rely on electrohydrodynamically induced flows. Such methods enable the direct, additive patterning of materials with a resolution that can extend below 100 nm to provide unique opportunities not only in scientific studies but also in a range of applications that includes printed electronics, tissue engineering, and photonic and plasmonic devices. Following a brief historical perspective, this review presents descriptions of the underlying processes involved in the formation of liquid cones and jets to establish critical factors in the printing process. Different printing systems that share similar principles are then described, along with key advances that have been made in the last decade. Capabilities in terms of printable materials and levels of resolution are reviewed, with a strong emphasis on areas of potential application.

Ferroelectric Crystals for Photonic Applications

The dynamic evolution of the periodic poling into resist patterned LiNbO3 samples is investigated here by measuring the variation of the optical phase shift distribution across domain walls. In situ quantitative and high-contrast imaging is obtained through an interference microscope based on digital holography. Phase map movies of the evolving domains are presented and discussed. The method provides remarkable in situ information about the poling of ferroelectric crystals in the case of insulating constraints, under different voltage and resist conditions, providing a deeper understanding of the electric field periodic poling.

In situ investigation of periodic poling in congruent LiNbO3 by quantitative interference microscopy

We present a quick, simple and accurate digital holographic characterization of the Bessel beams produced by polymeric microaxicons. This technique allows the numerical reconstruction of both intensity and phase of the beam at whichever point starting from a single acquired hologram. From these data, it is possible to go back to the axicon structure, and to gather information about their characteristics. In particular, the focal length and the depth of focus of the axicon lens are experimentally measured, and the full width at half maximum of the beam is obtained too. The depth of focus, very large for a Bessel beam with respect to a Gaussian one, is successfully exploited for optical trapping of micrometric objects.

Characterization of Bessel beams generated by polymeric microaxicons

We propose an optical configuration in which floating particles in a microfluidic chamber can be characterized by an interference microscopy configuration to obtain quantitative phase-contrast maps. The configuration is simply made by two laser beams from the same laser source. One beam provides the optical forces for driving the particle along appropriate paths, but at same time works as the object illumination beam in the holographic microscope. The second beam plays the role of the reference beam, allowing recording of an interference fringe pattern (i.e., the digital hologram) in an out-of-focus image plane. The system and method are illustrated and experimental results are offered for polymeric particles as well as for in vitro cells with the aim to demonstrate the approach.

Driving and analysis of micro-objects by digital holographic microscope in microfluidics

Biomolecule detection is important in many fields, but obviously becomes increasingly difficult as concentrations decrease. Here, the authors demonstrate a method to dispense and concentrate biomolecules onto a target slide, greatly improving the detection limits.
