As the most important skeletal component in plants, the polysaccharide cellulose is an almost inexhaustible polymeric raw material with fascinating structure and properties. Formed by the repeated connection of D-glucose building blocks, the highly functionalized, linear stiff-chain homopolymer is characterized by its hydrophilicity, chirality, biodegradability, broad chemical modifying capacity, and its formation of versatile semicrystalline fiber morphologies. In view of the considerable increase in interdisciplinary cellulose research and product development over the past decade worldwide, this paper assembles the current knowledge in the structure and chemistry of cellulose, and in the development of innovative cellulose esters and ethers for coatings, films, membranes, building materials, drilling techniques, pharmaceuticals, and foodstuffs. New frontiers, including environmentally friendly cellulose fiber technologies, bacterial cellulose biomaterials, and in-vitro syntheses of cellulose are highlighted together with future aims, strategies, and perspectives of cellulose research and its applications.

1.1 Photodynamic Therapy and Imaging
The purpose of this review is to present the current state of the role of imaging in photodynamic therapy (PDT). In order for the reader to fully appreciate the context of the discussions embodied in this article we begin with an overview of the PDT process, starting with a brief historical perspective followed by detailed discussions of specific applications of imaging in PDT. Each section starts with an overview of the specific topic and, where appropriate, ends with summary and future directions. The review closes with the authors’ perspective of the areas of future emphasis and promise. The basic premise of this review is that a combination of imaging and PDT will provide improved research and therapeutic strategies.

PDT is a photochemistry-based approach that uses a light-activatable chemical, termed a photosensitizer (PS), and light of an appropriate wavelength, to impart cytotoxicity via the generation of reactive molecular species (Figure 1a). In clinical settings, the PS is typically administered intravenously or topically, followed by illumination using a light delivery system suitable for the anatomical site being treated (Figure 1b). The time delay, often referred to as drug-light interval, between PS administration and the start of illumination with currently used PSs varies from 5 minutes to 24 hours or more depending on the specific PS and the target disease. Strictly speaking, this should be referred to as the PS-light interval, as at the concentrations typically used the PS is not a drug, but the drug-light interval terminology seems to be used fairly frequently. Typically, the useful range of wavelengths for therapeutic activation of the PS is 600 to 800 nm, to avoid interference by endogenous chromophores within the body, and yet maintain the energetics necessary for the generation of cytotoxic species (as discussed below) such as singlet oxygen (1O2). However, it is important to note that photosensitizers can also serve as fluorescence imaging agents for which activation with light in the 400nm range is often used and has been extremely useful in diagnostic imaging applications as described extensively in Section 2 of this review. The obvious limitation of short wavelength excitation is the lack of tissue penetration so that the volumes that are probed under these conditions are relatively shallow.

Biological applications of optical forces

: The unpolarized absorption and circular dichroism spectra of the fundamental vibrational transitions of the chiral molecule, 4-methyl-2-oxetanone, are calculated ab initio. Harmonic force fields are obtained using Density Functional Theory (DFT), MP2, and SCF methodologies and a 5S4P2D/3S2P (TZ2P) basis set. DFT calculations use the Local Spin Density Approximation (LSDA), BLYP, and Becke3LYP (B3LYP) density functionals. Mid-IR spectra predicted using LSDA, BLYP, and B3LYP force fields are of significantly different quality, the B3LYP force field yielding spectra in clearly superior, and overall excellent, agreement with experiment. The MP2 force field yields spectra in slightly worse agreement with experiment than the B3LYP force field. The SCF force field yields spectra in poor agreement with experiment.The basis set dependence of B3LYP force fields is also explored: the 6-31G* and TZ2P basis sets give very similar results while the 3-21G basis set yields spectra in substantially worse agreements with experiment. jg

We present a comparison between two basically different optical detection systems: a confocal epifluorescence microscope, and a new evanescent wave detection system employing a parabolic optical element. In a microscope set-up, fluorescence light is collected within a cone around the optical axis, whereas in the evanescent light detector, fluorescence light is collected mainly at angles larger than the so-called critical angle of total internal reflection. Based on a thorough theoretical modeling of both experimental set-ups, comparison between the two detection systems is made Particularly, the optical detection efficiency is compared. 5

Comparison between a conventional epifluorescence microscope and a new highly efficient evanescent wave detector in single-molecule spectroscopic applications

Abstract. Flood marks are rarely utilized in hazard assessment, mainly because of a lack of data availability and accessibility and mistrust in their reliability. Challenging these common assumptions, we present an approach
for evaluation and practical utilization of flood marks by the example of
the Kinzig River, a Rhine tributary from the Black Forest with a history of
severe floods. We combined written documents describing flood marks with
field mapping at three study sites and collected information relating to 89 marks – about 50 % of them still preserved – which refer to ≥15 large floods between 1824 and 1991. The inclusion of a detailed historical-mark survey enabled an assessment of changes through time for each flood mark: they extend from small (±15 cm) imprecisions in mark heights to considerable uncertainties in position, height, and displayed date for some modified marks. Plausibility checks with further data nevertheless demonstrated good overall consistency. We then juxtaposed these marks with the current, modeled flood hazard maps. A wide agreement is apparent, in that the large majority of marks are situated at probable heights and within the modeled flooding area associated with extreme floods. For the few exceptions, we see plausible and historically sound reasons in changed local hydraulic conditions by flood protection walls, exceptional processes during a massive ice jam, and possibly also a local underestimation of hazard along Kinzig River tributaries. Overall, this study highlights (1) the broad availability of flood mark data, both on a larger spatial scale and with regard to already vanished marks, and (2) the significance of the marks, verified by further data, and also demonstrates (3) the possibility of a straightforward inclusion in hazard assessment. We thus encourage the systematic collection, maintenance, and integration of flood marks as responsible risk management, not least regarding their value in the wider context of risk awareness and memory.

Reliability of flood marks and practical relevance for flood hazard assessment in southwestern Germany

The understanding of processes occurring at the interface between two media are of prior importance in various fields of research, from material sciences to biology. A custom-made microscope objective based on the supercritical angle technique was developed in our group, allowing to probe these interfacial events by carrying out surface-sensitive and low invasive spectroscopy of aqueous samples. A biological example of particular interest is the comprehension of neurodegenerative diseases which seem caused by the interaction of specific peptides with the membrane of the neurons. Taking advantage of our optical setup, we used supercritical angle fluorescence spectroscopy to specifically monitor the interaction between a supported lipid bilayer (SLB) and the Amyloid β peptide, notably responsible of the Alzheimer disease. Different forms of the peptide (40 and 42 amino acids composition) were tested and the interfacial fluorescence measured to get information about the lipid integrity and mobility. The adsorption of the peptide was also characterized in terms of kinetic and affinity.
