Journal ArticleDOI
Intrahepatic Genetic Inoculation of Hepatitis C Virus RNA Confers Cross-Protective Immunity
Amy J. Weiner,Xavier Paliard,Mark Selby,Angelica Medina-Selby,Doris Coit,Steve Nguyen,Joe Kansopon,Christopher L. Arian,Philip Ng,Jeffery Tucker,Chun-Ting Lee,Noelle K. Polakos,Jang Han,Shirley Wong,Hui-hua Lu,Steve Rosenberg,Kathy Brasky,David Y. Chien,George Kuo,Michael Houghton +19 more
TL;DR: It is demonstrated that the chimpanzee not only developed protective immunity to the homologous type 1a RNA after rechall challenge by IHGI but was also protected from chronic HCV infection after sequential rechallenge with 100 50% chimpanzee infectious doses of a heterologoustype 1a (H77) and 1b (HC-J4) whole-virus inoculum.
Journal ArticleDOI
Hepatitis C virus polyprotein vaccine formulations capable of inducing broad antibody and cellular immune responses.
Michael Vajdy,Mark J. Selby,Angelica Medina-Selby,Doris Coit,Hall John A,Laura Tandeske,David Y. Chien,Celine Hu,Domenico Rosa,Manmohan Singh,Jina Kazzaz,Steve Nguyen,Steve Coates,Philip Ng,Sergio Abrignani,Yin-Ling Lin,Michael Houghton,Derek T. O'Hagan +17 more
TL;DR: The data suggest that the HCV polyprotein delivered with adjuvants induces broad B- and T-cell responses and could be a vaccine candidate against HCV.
Journal ArticleDOI
Use of a Novel Hepatitis C Virus (HCV) Major-Epitope Chimeric Polypeptide for Diagnosis of HCV Infection
David Y. Chien,Phillip Arcangel,Angelica Medina-Selby,Doris Coit,Mark A. Baumeister,Steve Nguyen,Carlos George-Nascimento,Alexander Gyenes,George Kuo,Pablo Valenzuela +9 more
TL;DR: The results indicate that MEFA-6 exposes all of the major immunogenic epitopes from the seven functional regions of the HCV genome, and its excellent sensitivity and specificity for the detection of clinical seroconversion are demonstrated by this assay.
Journal ArticleDOI
Design of Novel Conformational and Genotype-Specific Antigens for Improving Sensitivity of Immunoassays for Hepatitis C Virus-Specific Antibodies
Sansan Lin,Phillip Arcangel,Angelica Medina-Selby,Doris Coit,Philip Ng,Steve Nguyen,Colin S. McCoin,Alex Gyenes,Celine Hu,Laura Tandeske,Bruce Phelps,David Y. Chien +11 more
TL;DR: An HCV antibody assay that employs a conformational protein, NS3NS4a PI (with functional protease and helicase activities), and a linear fusion protein, multiple-epitope fusion antigen 7.1 or 7.2 may have the potential to replace current HCV immunoassays for better sensitivity.
Journal ArticleDOI
Hepatitis B virus e antigen specific epitopes and limitations of commercial anti-HBe immunoassays.
Mark A. Baumeister,Angelica Medina-Selby,Doris Coit,Steve Nguyen,Carlos George-Nascimento,Alexander Gyenes,Pablo Valenzuela,George Kuo,David Y. Chien +8 more
TL;DR: It was found that early anti‐HBe seroconversion antibodies appear to be conformational, whereas later serconversion, more typically associated with the clearance of HBeAg, is characterized by the presence of antibodies to the linear H beAg epitopes.
