Journal ArticleDOI
Characterization of Hepatitis C Virus Core-Specific Immune Responses Primed in Rhesus Macaques by a Nonclassical ISCOM Vaccine
Noelle K. Polakos,Debbie Drane,John Cooper Cox,Philip Ng,Mark Selby,David Y. Chien,Derek T. O'Hagan,Michael Houghton,Xavier Paliard +8 more
TL;DR: Evidence is provided that Core-ISCOM is effective at inducing cellular and humoral immune responses in nonhuman primates and that it could serve as an adjuvant for the HCV envelope protein E1E2.
Journal ArticleDOI
Intrahepatic Genetic Inoculation of Hepatitis C Virus RNA Confers Cross-Protective Immunity
Amy J. Weiner,Xavier Paliard,Mark Selby,Angelica Medina-Selby,Doris Coit,Steve Nguyen,Joe Kansopon,Christopher L. Arian,Philip Ng,Jeffery Tucker,Chun-Ting Lee,Noelle K. Polakos,Jang Han,Shirley Wong,Hui-hua Lu,Steve Rosenberg,Kathy Brasky,David Y. Chien,George Kuo,Michael Houghton +19 more
TL;DR: It is demonstrated that the chimpanzee not only developed protective immunity to the homologous type 1a RNA after rechall challenge by IHGI but was also protected from chronic HCV infection after sequential rechallenge with 100 50% chimpanzee infectious doses of a heterologoustype 1a (H77) and 1b (HC-J4) whole-virus inoculum.
Journal ArticleDOI
Evaluation of hepatitis C virus glycoprotein E2 for vaccine design: an endoplasmic reticulum-retained recombinant protein is superior to secreted recombinant protein and DNA-based vaccine candidates.
Jens Heile,Yiu-Lian Fong,Domenico Rosa,Kim Berger,Giulietta Saletti,Susanna Campagnoli,Giuliano Bensi,Sabrina Capo,Steve Coates,Kevin Crawford,Christine Dong,Mark Wininger,Gary Baker,Larry Cousens,David Y. Chien,Philip Ng,Phillip Archangel,Guido Grandi,Michael Houghton,Sergio Abrignani +19 more
TL;DR: Comparing recombinant intracellular E2 protein to several E2-encoding DNA vaccines in mice found that protein immunization is superior to DNA in both the quantity and quality of the antibody response elicited.
Journal ArticleDOI
Cationic microparticles are a potent delivery system for a HCV DNA vaccine.
Derek T. O'Hagan,Manmohan Singh,Christine Dong,Mildred Ugozzoli,Kim Berger,Edward Glazer,Mark J. Selby,Mark Wininger,Philip Ng,Kevin Crawford,Xavier Paliard,Steven Coates,Michael Houghton +12 more
TL;DR: Cationic PLG microparticles with adsorbed HCV DNA generates potent immune responses, and PLG/DNA is selected for further evaluation in a non-human primate model.
Journal ArticleDOI
Hepatitis C virus polyprotein vaccine formulations capable of inducing broad antibody and cellular immune responses.
Michael Vajdy,Mark J. Selby,Angelica Medina-Selby,Doris Coit,Hall John A,Laura Tandeske,David Y. Chien,Celine Hu,Domenico Rosa,Manmohan Singh,Jina Kazzaz,Steve Nguyen,Steve Coates,Philip Ng,Sergio Abrignani,Yin-Ling Lin,Michael Houghton,Derek T. O'Hagan +17 more
TL;DR: The data suggest that the HCV polyprotein delivered with adjuvants induces broad B- and T-cell responses and could be a vaccine candidate against HCV.
