Journal ArticleDOI
Expression, crystallization, and three-dimensional structure of the catalytic domain of human plasma kallikrein.
Jie Tang,Christine Yu,Steven R. Williams,Eric B. Springman,Douglas A. Jeffery,Paul A. Sprengeler,Alberto Estevez,Jun Sampang,William D. Shrader,Jeff Spencer,Wendy B. Young,Mary E. McGrath,Bradley A. Katz +12 more
TL;DR: Crystallization and x-ray crystal structure determination of both forms have yielded the first three-dimensional views of the catalytic domain of plasma kallikrein, and it is shown that the protease domain adopts a typical chymotrypsin-like serine protease conformation.
Journal ArticleDOI
Dissecting and designing inhibitor selectivity determinants at the S1 site using an artificial Ala190 protease (Ala190 uPA).
Bradley A. Katz,Christine Luong,Joseph D. Ho,John R. Somoza,Erik Gjerstad,Jie Tang,Steven R. Williams,Erik Verner,Richard L. Mackman,Wendy B. Young,Paul A. Sprengeler,Hedy Chan,Kyle Mortara,James W. Janc,Mary E. McGrath +14 more
TL;DR: Crystal structures of uPA complexes of novel, active site-directed arylguanidine and 2-aminobenzimidazole inhibitors of WT uPA, together with associated K(i) values, indicate a significant role of Ser190 in the binding of these classes of UPA inhibitors.
