— Anthocyanins are water-soluble pigments found in all plant tissues throughout the plant kingdom. Our understanding of anthocyanin biosynthesis and its molecular control has greatly improved in the last decade. The adaptive advantages of anthocyanins, especially in non-reproductive tissues, is much less clear. Anthocyanins often appear transiently at specific developmental stages and may be induced by a number of environmental factors including visible and UVB radiation, cold temperatures and water stress. The subsequent production and localization of anthocyanins in root, stem and especially leaf tissues may allow the plant to develop resistance to a number of environmental stresses. This article reviews the environmental induction of anthocyanins and their proposed importance in ameliorating environmental stresses induced by visible and UVB radiation, drought and cold temperatures.

Environmental Significance of Anthocyanins in Plant Stress Responses

The function of anthocyanins in green, vegetative tissues has always been a contentious issue. Here we evaluate their proposed photoprotective function since recent findings have shown that anthocyanins reduce photoinhibition and photobleaching of chlorophyll under light stress conditions. Anthocyanins generally accumulate in peripheral tissues exposed to high irradiance, although there are some exceptions (e.g. accumulation in abaxial leaf tissues and in obligatory shade plants) and accumulation is usually transient. Anthocyanin accumulation requires light and generally coincides with periods of high excitation pressure and increased potential for photo-oxidative damage due to an imbalance between light capture, CO2 assimilation and carbohydrate utilization (e.g. greening of developing tissues, senescence and adverse environmental conditions). Light attenuation by anthocyanin may help to re-establish this balance and so reduce the risk of photo-oxidative damage. Although it has been suggested that anthocyanins may act as antioxidants, the association between anthocyanins and oxidative stress appears to relate to the ability of anthocyanins to reduce excitation pressure and, hence, the potential for oxidative damage. The various aspects of anthocyanin induction and pigmentation presented here are compatible with, and support, the proposed general role of anthocyanins as photoprotective light screens in vegetative tissues.

Anthocyanins in vegetative tissues: a proposed unified function in photoprotection

The roles of flavonoids in plant defence against pathogens, herbivores, and environmental stress are reviewed and their significant contribution to plant resistance is discussed. The induction of flavonoids is of particular interest for gathering evidence of their roles. Tools are mentioned which may enhance flavonoid biosynthesis and accumulation. These include metabolic engineering and UV light. The induction of defence-related flavonoids is modified by other determining factors and competition between growth and secondary metabolism may exist. In an evolutionary context, stress-related oxidative pressure may have been a major trigger for the distribution and abundance of flavonoids. UV protection is one of their most significant, or even the most significant, functional role for flavonoids. The multi-functionality of these compounds, however, often complicates the interpretation of experimental results but, overall, it supports the importance of flavonoids.

Accumulation of 3-deoxyanthocyanidin phytoalexins and resistance to Colletotrichum sublineolum in sorghum

Sorghum (Sorghum bicolor L. Moench) accumulates the anthocyanin cyanidin 3-dimalonyl glucoside in etiolated mesocotyls in response to light. Inoculation with the nonpathogenic fungus Cochliobolus heterostrophus drastically reduced the light-induced accumulation of anthocyanin by repressing the transcription of the anthocyanin biosynthesis genes encoding flavanone 3-hydroxylase, dihydroflavonol 4-reductase, and anthocyanidin synthase. In contrast to these repression effects, fungal inoculation resulted in the synthesis of the four known 3-deoxyanthocyanidin phytoalexins and a corresponding activation of genes encoding the key branch-point enzymes in the phenylpropanoid pathway, phenylalanine ammonia-lyase and chalcone synthase. In addition, a gene encoding the pathogenesis-related protein PR-10 was strongly induced in response to inoculation. The accumulation of phytoalexins leveled off by 48 h after inoculation and was accompanied by a more rapid increase in the rate of anthocyanin accumulation. The results suggest that the plant represses less essential metabolic activities such as anthocyanin synthesis as a means of compensating for the immediate biochemical and physiological needs for the defense response.

Reduction of light-induced anthocyanin accumulation in inoculated sorghum mesocotyls : Implications for a compensatory role in the defense response

A sorghum cDNA clone was isolated by differential screening of a cDNA library prepared from mesocotyls (cultivar DK18) inoculated with fungal pathogenes. The deduced translation product shows sequence similarity to a family of intracellular pathogenesis-related proteins (PR-10) with a potential ribonuclease function. We studied the accumulation of PR-10 and chalcone synthase (CHS) transcripts in mesocotyls following inoculation with Cochliobolus heterostrophus or Colletotrichum sublineolum. CHS is involved in phytoalexin synthesis in sorghum. Coordinate expression of PR-10 and CHS genes was localized in the area of inoculation along with the accumulation of phytoalexins. C. heterostrophus is a nonpathogen of sorghum and cytological studies indicated that cultivar DK18 is resistant to C. sublineolum, a sorghum pathogen. We demonstrated that the two fungi triggered different time courses of plant defense reactions. Inoculation with C. heterostrophus resulted in rapid accumulation of PR-10 and CHS transcripts after appressoria had become mature. Accumulation of these transcripts was delayed in plants inoculated with C. sublineolum until penetration of host tissue had been completed and infection vesicles had formed. Results suggest that different recognition events are involved in the expression of resistance to the two fungi used or that C. sublineolum suppresses the nonspecific induction of defense responses.
