Parasitoids and predators of herbivores have evolved and function within a multitrophic context. Consequently, their physiology and behavior are in­ fluenced by elements from other trophic levels such as their herbivore victim (second trophic level) and its plant food (first trophic level) (126) . Natural enemies base their foraging decisions on information from these different trophic levels, and chemical information plays an important role. This review is restricted to the ecology of chemical information from the first and second trophic levels. The importance of so-called infochemicals, a subcategory of semiochemicals, in foraging by parasitoids and predators has been well documented (e.g. reviewed in 31, 78, Il l , 183, 185) , and we do not intend to repeat the details. But because of a lack of testable hypotheses, all this research is conducted rather haphazardly: the total puzzle of infochemical use has not been solved for any natural enemy species. Here we approach the use of infochemicals by natural enemies from an evolutionary and ecological standpoint. Our basic concept is that information from the first and second trophic levels differs in availability and in reliability, a difference that shapes the way infochemicals are used by a species. Cytochrome P450 systems--biological variations of electron transport chains.

P450BM3 (CYP102A1), a fatty acid hydroxylase from Bacillus megaterium, has been extensively studied over a period of almost forty years. The enzyme has been redesigned to catalyse the oxidation of non-natural substrates as diverse as pharmaceuticals, terpenes and gaseous alkanes using a variety of engineering strategies. Crystal structures have provided a basis for several of the catalytic effects brought about by mutagenesis, while changes to reduction potentials, inter-domain electron transfer rates and catalytic parameters have yielded functional insights. Areas of active research interest include drug metabolite production, the development of process-scale techniques, unravelling general mechanistic aspects of P450 chemistry, methane oxidation, and improving selectivity control to allow the synthesis of fine chemicals. This review draws together the disparate research themes and places them in a historical context with the aim of creating a resource that can be used as a gateway to the field.

P450BM3 (CYP102A1): connecting the dots

Stored-product insects can cause postharvest losses, estimated from up to 9% in developed countries to 20% or more in developing countries. There is much interest in alternatives to conventional insecticides for controlling stored-product insects because of insecticide loss due to regulatory action and insect resistance, and because of increasing consumer demand for product that is free of insects and insecticide residues. Sanitation is perhaps the first line of defense for grain stored at farms or elevators and for food-processing and warehouse facilities. Some of the most promising biorational management tools for farm-stored grain are temperature management and use of natural enemies. New tools for computer-assisted decision-making and insect sampling at grain elevators appear most promising. Processing facilities and warehouses usually rely on trap captures for decision-making, a process that needs further research to optimize.

Biorational approaches to managing stored-product insects.

I review the recent literature on cuticular hydrocarbons (CHs) in Drosophila. First, the major structural features of CHs are examined in a variety of species with regard to phylogeny. The genetic bases of the CH variation between and within species have been investigated with some of the genes involved characterized and manipulated. The effect of non-genetic factors as temperature, food and development is also examined with regard to CH production. Using a model involving the stimulating or the inhibiting role of CHs, it is possible to speculate on the mechanisms of CH perception and on the role(s) that these substances could play on sexual isolation and on the evolution of pheromonal communication.

Carnivorous arthropods use volatile infochemicals emitted from prey-infested plants in their foraging behavior. Although several volatile components are common among plant species, the compositions differ among prey-plant complexes. Studies showed that the predatory mite Neoseiulus womersleyi is attracted only to previously experienced plant volatiles. In this study, we identified the attractant components in prey-induced plant volatiles of two prey-plant complexes. N. womersleyi reared on Tetranychus kanzawai-infested tea leaves showed significant preference for a mixture of three synthetic compounds [mimics of the T. kanzawai-induced tea leaves volatiles: (E)-beta-ocimene, (E)-4,8-dimethyl-1,3,7-nonatriene (DMNT), and (E,E)-alpha-farnesene] at a level comparable to that for T. kanzawai-induced tea plant volatiles. However, mixtures lacking any of these compounds did not attract the predatory mites. Likewise, N. womersleyi reared on T. urticae-infested kidney bean plants showed a significant preference for a mixture of four synthetic compounds [mimics of the T. urticae-induced kidney bean volatiles: DMNT, methyl salicylate (MeSA), beta-caryophyllene, and (E,E)-4,8,12-trimethyl-1,3,7,11-tridecatetraene] at a level comparable to that for T. urticae-induced kidney bean volatiles. The absence of any of the four compounds resulted in no attraction. These results indicate that N. womersleyi can use at least four volatile components to identify prey-infested plants.

Cytochrome P450foxy (P450foxy, CYP505) is a fused protein of cytochrome P450 (P450) and its reductase isolated from the fungus Fusarium oxysporum, which catalyzes the subterminal (omega-1 approximately omega-3) hydroxylation of fatty acids. Here, we produced, purified and characterized a fused recombinant protein (rP450foxy) using the Escherichia coli expression system. Purified rP450foxy was catalytically and spectrally indistinguishable from the native protein, but most of the rP450foxy was recovered in the soluble fraction of E. coli cells unlike the membrane-bound native protein. The results are consistent with our notion that the native protein is targeted to the membrane by a post-translational modification mechanism. We also discovered that P450foxy could use shorter saturated fatty acid chains (C9 and C10) as a substrate. The regiospecificity (omega-1 approximately omega-3) of hydroxylation due to the enzymatic reaction for the short substrates (decanoate, C10; undecanoate, C11) was the same as that for longer substrates. Steady state kinetic studies showed that the kcat values for all substrates tested (C9-C16) were of the same magnitude (1200-1800 min-1), whereas the catalytic efficiency (kcat/Km) was higher for longer fatty acids. Substrate inhibition was observed with fatty acid substrates longer than C13, and the degree of inhibition increased with increasing chain length. This substrate inhibition was not apparent with P450BM3, a bacterial counterpart of P450foxy, which was the first obvious difference in their catalytic properties to be identified. Kinetic data were consistent with the inhibition due to binding of the second substrate. We discuss the inhibition mechanism based on differences between P450foxy and P450BM3 in key amino acid residues for substrate binding.
