Passive Immunization with Phospho-Tau Antibodies Reduces Tau Pathology and Functional Deficits in Two Distinct Mouse Tauopathy Models
Sethu Sankaranarayanan,Donna M. Barten,Laurel Vana,Nino Devidze,Ling Yang,Gregory W. Cadelina,Nina Hoque,Lynn B. DeCarr,Stefanie Keenan,Alan Lin,Yang Cao,Bradley Snyder,Bin Zhang,Magdalena Nitla,Gregg Hirschfeld,Nestor X. Barrezueta,Craig Polson,Paul D. Wes,Vangipuram S. Rangan,Angela Cacace,Charles F. Albright,Jere E. Meredith,John Q. Trojanowski,Virginia M.-Y. Lee,Kurt R. Brunden,Michael K. Ahlijanian +25 more
TL;DR: It is demonstrated that phospho-tau (p-t Tau) antibodies, PHF6 and PHF13, can prevent the induction of tau pathology in primary neuron cultures and provides evidence supporting the use of t Tau immunotherapy as a potential treatment option for AD and other tauopathies.
Journal ArticleDOI
The Amyloid-β Rise and γ-Secretase Inhibitor Potency Depend on the Level of Substrate Expression *
Catherine R. Burton,Jere E. Meredith,Donna M. Barten,Margi E. Goldstein,Carol M. Krause,Cathy J. Kieras,Lisa M. Sisk,Lawrence G. Iben,Craig Polson,Mark W. Thompson,Xu-Alan Lin,Jason A. Corsa,Tracey Fiedler,Maria Pierdomenico,Yang Cao,Arthur H. Roach,Joseph L. Cantone,Michael J. Ford,Dieter M. Drexler,Richard E. Olson,Yang Michael G,Bergstrom Carl P,Mcelhone Katharine E,Joanne J. Bronson,John E. Macor,Yuval Blat,Robert H. Grafstrom,Andrew M. Stern,Dietmar A. Seiffert,Robert Zaczek,Charles F. Albright,Jeremy H. Toyn +31 more
TL;DR: It is shown that the Aβ rise depends on the β-secretase-derived C-terminal fragment of APP (βCTF or C99 levels with low levels with high levels causing rises, while the N-terminally truncated form of Aβ, known as “p3,” formed by α- secretase cleavage, did not exhibit a rise.
Journal ArticleDOI
Discovery of Isonicotinamides as Highly Selective, Brain Penetrable, and Orally Active Glycogen Synthase Kinase-3 Inhibitors.
Guanglin Luo,Ling Chen,Catherine R. Burton,Hong Xiao,Prasanna Sivaprakasam,Carol M. Krause,Yang Cao,Nengyin Liu,Jonathan Lippy,Wendy Clarke,Kimberly Snow,Joseph Raybon,Vinod Arora,Matt Pokross,Kevin Kish,Hal A. Lewis,David R. Langley,John E. Macor,Gene M. Dubowchik +18 more
TL;DR: A class of isonicotinamides that are potent, highly kinase-selective GSK-3 inhibitors are presented, the members of which demonstrated oral activity in a triple-transgenic mouse model of AD.
Journal ArticleDOI
Ex Vivo Occupancy of γ-Secretase Inhibitors Correlates with Brain β-Amyloid Peptide Reduction in Tg2576 Mice
Margi E. Goldstein,Yang Cao,Tracey Fiedler,Jeremy H. Toyn,Lawrence G. Iben,Donna M. Barten,Maria Pierdomenico,Jason A. Corsa,C.V.C. Prasad,Richard E. Olson,Yu-Wen Li,Robert Zaczek,Charles F. Albright +12 more
TL;DR: The excellent correlation between brain Aβ40 andγ-secretase occupancy suggests that a positron emission tomography ligand for γ- secretase could be a valuable biomarker to determine whether γ -secretase inhibitors bind to their target in humans.
Journal ArticleDOI
Allele-Selective Knockdown of MYH7 Using Antisense Oligonucleotides.
Brian R. Anderson,Marianne Lerbech Jensen,Peter Hagedorn,Sean C. Little,Richard E. Olson,Ron Ammar,Bernadette Kienzle,John Ryan Thompson,Ivar M. McDonald,Stephen E. Mercer,Jonas Vikesaa,Bettina Nordbo,Larry Iben,Yang Cao,JoAnne E Natale,Greg Dalton-Kay,Angela Cacace,Bo Hansen,Maj Hedtjarn,Troels Koch,Linda J. Bristow +20 more
TL;DR: The ability of locked nucleic acid (LNA)-modified antisense oligonucleotides (ASOs) to selectively knock down mutant myosin transcripts by targeting single-nucleotide polymorphisms (SNPs) is investigated and suggests that SNP-targeting ASOs are a promising therapeutic modality for treating cardiac pathology.
